Background : Diabetes mellitus is the world wide metabolic disease and lead to higher mortality in every years around the globe. This conditions lead to increased the insensity and duration of inflammation, thus impair the healing proccesses. Fibroblast as the major component of proliferative phase in healing decreased and the collagenases proteases enzim MMP-9 increased in diabetes. Hence, the ulceration as the most common oral disease become harder to heal and take a long periods. Zinc Sulphate as the trace elements has been used in years as the wounds treatment. This mineral has the important role in healing phase, by inducing the signalling, proliferations and migrations of the healing phase components. Objectives : The aim of this research, is to examine and analyze the effect of the topical zinc sulphate 1 % therapy in fibroblast and MMP-9 expressions in mucosa tissue of wistar rat with diabetic conditions.Method : The research uses post test only controls group design. Fifthy eight wistar rat 2-3 month olds, 150-200 gram in weight. The samples divided into four group, normal, diabetes, normal + ZnSO4 and diabetes+ZnSO4 . Diabetes induced by streptozotocin 30 mg diluted in 1,33 ml buffer citrates. After treatment, the specimens were staining with HE for fibroblast and immunohistochemistry for MMP-9 expressions, and analyzed using one way anova. Result : The uses of zinc sulphate 1% therapy in traumatic ulceration, increase the fibroblast number and decrease the MMP-9 expressions in diabetic conditions showed significant different (p<0,05). Conclusion : After therapy of topically zinc sulphate the number of fibroblast were significantly increased and the expressions of MMP-9 were significantly decreased. key words : diabetes melitus, traumatic ulcer, zinc sulphate, fibroblast, MMP-
