    Frequent EGFR Positivity and Overexpression in High-Grade Areas of Human MPNSTs

    Malignant peripheral nerve sheath tumours (MPNSTs) are highly malignant and resistant. Transformation might implicate up regulation of epidermal growth factor receptor (EGFR). Fifty-two MPNST samples were studied for EGFR, Ki-67, p53, and survivin expression by immunohistochemistry and for EGFR amplification by in situ hybridization. Results were correlated with clinical data. EGFR RNA was also quantified by RT-PCR in 20 other MPNSTs and 14 dermal neurofibromas. Half of the patients had a neurofibromatosis type 1 (NF1). EGFR expression, detected in 86% of MPNSTs, was more frequent in NF1 specimens and closely associated with high-grade and p53-positive areas. MPNSTs expressed more EGFR transcripts than neurofibromas. No amplification of EGFR locus was observed. NF1 status was the only prognostic factor in multivariate analysis, with median survivals of 18 and 43 months for patients with or without NF1. Finally, EGFR might become a new target for MPNSTs treatment, especially in NF1-associated MPNSTs

    JOYS+: mid-infrared detection of gas-phase SO2_2 emission in a low-mass protostar. The case of NGC 1333 IRAS2A: hot core or accretion shock?

    JWST/MIRI has sharpened our infrared eyes toward the star formation process. This paper presents the first mid-infrared detection of gaseous SO2_2 emission in an embedded low-mass protostellar system. MIRI-MRS observations of the low-mass protostellar binary NGC 1333 IRAS2A are presented from the JWST Observations of Young protoStars (JOYS+) program, revealing emission from the SO2 Μ3_2~\nu_3 asymmetric stretching mode at 7.35 micron. The results are compared to those derived from high-angular resolution SO2_2 data obtained with ALMA. The SO2_2 emission from the Îœ3\nu_3 band is predominantly located on ∌50−100\sim50-100 au scales around the main component of the binary, IRAS2A1. A rotational temperature of 92±892\pm8 K is derived from the Îœ3\nu_3 lines. This is in good agreement with the rotational temperature derived from pure rotational lines in the vibrational ground state (i.e., Îœ=0\nu=0) with ALMA (104±5104\pm5 K). However, the emission of the Îœ3\nu_3 lines is not in LTE given that the total number of molecules predicted by a LTE model is found to be a factor 2×1042\times10^4 higher than what is derived for the Îœ=0\nu=0 state. This difference can be explained by a vibrational temperature that is ∌100\sim100 K higher than the derived rotational temperature of the Îœ=0\nu=0 state. The brightness temperature derived from the continuum around the Îœ3\nu_3 band of SO2_2 is ∌180\sim180 K, which confirms that the Îœ3=1\nu_3=1 level is not collisionally populated but rather infrared pumped by scattered radiation. This is also consistent with the non-detection of the Îœ2\nu_2 bending mode at 18-20 micron. Given the rotational temperature, the extent of the emission (∌100\sim100 au in radius), and the narrow line widths in the ALMA data (3.5 km/s), the SO2_2 in IRAS2A likely originates from ice sublimation in the central hot core around the protostar rather than from an accretion shock at the disk-envelope boundary.Comment: 19 pages, 17 figures, accepted for publication in A&A, abstract abbreviate

    Clinical characteristics and prognosis of osteosarcoma in young children: a retrospective series of 15 cases

    <p>Abstract</p> <p>Background</p> <p>Osteosarcoma is the most common primary bone malignancy in childhood and adolescence. However, it is very rare in children under 5 years of age. Although studies in young children are limited in number, they all underline the high rate of amputation in this population, with conflicting results being recently reported regarding their prognosis.</p> <p>Methods</p> <p>To enhance knowledge on the clinical characteristics and prognosis of osteosarcoma in young children, we reviewed the medical records and histology of all children diagnosed with osteosarcoma before the age of five years and treated in SFCE (Société Française des Cancers et leucémies de l'Enfant) centers between 1980 and 2007.</p> <p>Results</p> <p>Fifteen patients from 7 centers were studied. Long bones were involved in 14 cases. Metastases were present at diagnosis in 40% of cases. The histologic type was osteoblastic in 74% of cases. Two patients had a relevant history. One child developed a second malignancy 13 years after osteosarcoma diagnosis.</p> <p>Thirteen children received preoperative chemotherapy including high-dose methotrexate, but only 36% had a good histologic response. Chemotherapy was well tolerated, apart from a case of severe late convulsive encephalopathy in a one-year-old infant. Limb salvage surgery was performed in six cases, with frequent mechanical and infectious complications and variable functional outcomes.</p> <p>Complete remission was obtained in 12 children, six of whom relapsed. With a median follow-up of 5 years, six patients were alive in remission, seven died of their disease (45%), in a broad range of 2 months to 8 years after diagnosis, two were lost to follow-up.</p> <p>Conclusions</p> <p>Osteosarcoma seems to be more aggressive in children under five years of age, and surgical management remains a challange.</p

    Molecules with ALMA at Planet-forming Scales (MAPS). XVI. Characterizing the impact of the molecular wind on the evolution of the HD 163296 system

ST/R000549/1, ST/T000287/1, MR/T040726/1).During the main phase of evolution of a protoplanetary disk, accretion regulates the inner-disk properties, such as the temperature and mass distribution, and in turn, the physical conditions associated with planet formation. The driving mechanism behind accretion remains uncertain; however, one promising mechanism is the removal of a fraction of angular momentum via a magnetohydrodynamic (MHD) disk wind launched from the inner tens of astronomical units of the disk. This paper utilizes CO isotopologue emission to study the unique molecular outflow originating from the HD 163296 protoplanetary disk obtained with the Atacama Large Millimeter/submillimeter Array. HD 163296 is one of the most well-studied Class II disks and is proposed to host multiple gas-giant planets. We robustly detect the large-scale rotating outflow in the 12CO J = 2 - 1 and the 13CO J = 2 - 1 and J = 1 - 0 transitions. We constrain the kinematics, the excitation temperature of the molecular gas, and the mass-loss rate. The high ratio of the rates of ejection to accretion (5-50), together with the rotation signatures of the flow, provides solid evidence for an MHD disk wind. We find that the angular momentum removal by the wind is sufficient to drive accretion though the inner region of the disk; therefore, accretion driven by turbulent viscosity is not required to explain HD 163296's accretion. The low temperature of the molecular wind and its overall kinematics suggest that the MHD disk wind could be perturbed and shocked by the previously observed high-velocity atomic jet.     Malaria and vitamin A deficiency in African children: a vicious circle?

    Vitamin A deficiency and malaria are both highly prevalent health problems in Africa. Vitamin A deficiency affects over 30 million children, most of whom are in the age-group (under five years) most affected by malaria. Vitamin A deficiency increases all-cause mortality in this part of the population, and malaria is an important cause of death in children at this age. A low serum retinol concentration (a marker of vitamin A deficiency) is commonly found in children suffering from malaria, but it is not certain whether this represents pre-existing vitamin A deficiency, a contribution of malaria to vitamin A deficiency, or merely an acute effect of malaria on retinol metabolism or binding. In this paper, available evidence in support of a causal relationship in each direction between vitamin A deficiency and malaria is reviewed. If such a relationship exists, and especially if this is bidirectional, interventions against either disease may convey an amplified benefit for health

    Characterization of Macrophages and Osteoclasts in the Osteosarcoma Tumor Microenvironment at Diagnosis: New Perspective for Osteosarcoma Treatment?

    Biological and histopathological techniques identified osteoclasts and macrophages as targets of zoledronic acid (ZA), a therapeutic agent that was detrimental for patients in the French OS2006 trial. Conventional and multiplex immunohistochemistry of microenvironmental and OS cells were performed on biopsies of 124 OS2006 patients and 17 surgical (“OSNew”) biopsies respectively. CSF-1R (common osteoclast/macrophage progenitor) and TRAP (osteoclast activity) levels in serum of 108 patients were correlated to response to chemotherapy and to prognosis. TRAP levels at surgery and at the end of the protocol were significantly lower in ZA+ than ZA− patients (padj = 0.0011; 0.0132). For ZA+-patients, an increase in the CSF-1R level between diagnosis and surgery and a high TRAP level in the serum at biopsy were associated with a better response to chemotherapy (p = 0.0091; p = 0.0251). At diagnosis, high CD163+ was associated with good prognosis, while low TRAP activity was associated with better overall survival in ZA− patients only. Multiplex immunohistochemistry demonstrated remarkable bipotent CD68+/CD163+ macrophages, homogeneously distributed throughout OS regions, aside osteoclasts (CD68+/CD163−) mostly residing in osteolytic territories and osteoid-matrix-associated CD68−/CD163+ macrophages. We demonstrate that ZA not only acts on harmful osteoclasts but also on protective macrophages, and hypothesize that the bipotent CD68+/CD163+ macrophages might present novel therapeutic targets

In this paper, we address the major challenges of 3D sequential integration: in particular, the control of molecular bonding allows us to obtain pristine quality top active layer. With the help of Solid Phase Epitaxy, we can match the performance of top FET, processed at low temperature (600°C), with the bottom FET devices. Finally, the development of a stable salicide enables to retain bottom performance after top FET processing. Overcoming these major technological issues offers a wide range of applications

    A far-ultraviolet-driven photoevaporation flow observed in a protoplanetary disk.

    Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photodissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, which affects planet formation within the disks. We report James Webb Space Telescope and Atacama Large Millimeter Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modeling their kinematics and excitation allowed us to constrain the physical conditions within the gas. We quantified the mass-loss rate induced by the FUV irradiation and found that it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk
