    Rapid and non-destructive method to assess in the vineyardgrape berry anthocyanins under different seasonal andwater conditions

    Abstract Background and Aims: Monitoring of anthocyanins (Anth) in winegrape (Vitis vinifera L.) is fundamental for the production of top-quality red wines. This work was aimed at testing a new fluorescence-based sensor for Anth detection in the vineyard. Significance of the Study: The present study showed a new important innovative technology for viticulture. The sensor evaluation of the large spatial and temporal heterogeneity in Anth accumulation can be useful as support parameter in the harvest date decision or for vineyard zoning of phenolic maturity. Methods and Results: Anth in grape (cv. Aleatico) bunches attached to the vine were monitored non-destructively in the field using a fluorescence-based sensor during the 2008 and 2009 seasons and under different water regimes. The ANTHRG index = log(far-red-fluorescenceR/far-red-fluorescenceG), with fluorescence signals excited with red (R) and green (G) light, was inversely correlated through an exponential function (r2 = 0.875) to the Anth concentration derived from the HPLC analysis of berry skin extracts. ANTHRG was effective in detecting the earlier ripening process in 2009 with respect to 2008 and differences in the Anth accumulation between seasons and in relation to different water regimes.Water deficit imposed in 2009 enhanced Anth concentration in berries because of a reduction in berry size but also an increase of Anth biosynthesis. This effect was observed by both destructive and ANTHRG non-destructive measurements. Conclusions: Our results show that the employed fluorescence sensor represents a reliable, rapid and non-invasive tool for monitoring and determining Anth accumulation in situ

    Trans-heterozygosity for mutations enhances the risk of recurrent/chronic pancreatitis in patients with Cystic Fibrosis

    Background: Recurrent (RP) and chronic pancreatitis (CP) may complicate Cystic Fibrosis (CF). It is still unknown if mutations in genes involved in the intrapancreatic activation of trypsin (IPAT) or in the pancreatic secretion pathway (PSP) may enhance the risk for RP/CP in patients with CF.Methods: We enrolled: 48 patients affected by CF complicated by RP/CP and, as controls 35 patients with CF without pancreatitis and 80 unrelated healthy subjects. We tested a panel of 8 genes involved in the IPAT, i.e. PRSS1, PRSS2, SPINK1, CTRC, CASR, CFTR, CTSB and KRT8 and 23 additional genes implicated in the PSP.Results: We found 14/48 patients (29.2%) with mutations in genes involved in IPAT in the group of CF patients with RP/CP, while mutations in such genes were found in 2/35 (5.7%) patients with CF without pancreatitis and in 3/80 (3.8%) healthy subjects (p < 0.001). Thus, we found mutations in 12 genes of the PSP in 11/48 (22.9%) patients with CF and RP/CP. Overall, 19/48 (39.6%) patients with CF and RP/CP showed one or more mutations in the genes involved in the IPAT and in the PSP while such figure was 4/35 (11.4%) for patients with CF without pancreatitis and 11/80 (13.7%) for healthy controls (p < 0.001).Conclusions: The trans-heterozygous association between CFTR mutations in genes involved in the pathways of pancreatic enzyme activation and the pancreatic secretion may be risk factors for the development of recurrent or chronic pancreatitis in patients with CF

    Potential of a multiparametric optical sensor for determining in situ the maturity components of red and white vitis vinifera wine grapes

    A non-destructive fluorescence-based technique for evaluating Vitis vinifera L. grape maturity using a portable sensor (Multiplex ®) is presented. It provides indices of anthocyanins and chlorophyll in Cabernet Sauvignon, Merlot and Sangiovese red grapes and of flavonols and chlorophyll in Vermentino white grapes. The good exponential relationship between the anthocyanin index and the actual anthocyanin content determined by wet chemistry was used to estimate grape anthocyanins from in field sensor data during ripening. Marked differences were found in the kinetics and the amount of anthocyanins between cultivars and between seasons. A sensor-driven mapping of the anthocyanin content in the grapes, expressed as g/kg fresh weight, was performed on a 7-ha vineyard planted with Sangiovese. In the Vermentino, the flavonol index was favorably correlated to the actual content of berry skin flavonols determined by means of HPLC analysis of skin extracts. It was used to make a non-destructive estimate of the evolution in the flavonol concentration in grape berry samplings. The chlorophyll index was inversely correlated in linear manner to the total soluble solids (°Brix): it could, therefore, be used as a new index of technological maturity.     Effects of different water regimes on berry metabolism and composition of Aleatico grapevine

    This work reports the results of a trial carried out on ‘Aleatico’ grapevine, a cultivar cultivated along Tuscany coasts and in Elba Island, from which a typical dessert wine is produced after partial postharvest dehydration of berries. Two different water regimes were applied in the vineyard: not irrigated (water stress, WS) and partially irrigated (moderate water deficit, MWD). WS conditions determined a decrease of stem water potential (SWP) that reached values lower than - 1.4 Mpa, whereas in MWD these SWP levels were registered only for short periods. Significant differences in stomata conductance and net photosynthetic rate values were observed between the two treatments. At harvest, berry weight was reduced of about 20% in WS plants. An effect of similar magnitude was detected for skin weight, while seed weight was not affected. The sugar accumulation process was more pronounced in WS berries and this was paralleled by higher titratable acidity values both at veraison and harvest. Skin anthocyanin content was determined in laboratory and by a fluorimetric method, using the optical sensor Multiplex®2, an innovative portable instrument based on chlorophyll fluorescence, that nondestructively analyzes the berries in the field, under natural light. WS induced a significant increase of anthocyanin biosynthesis as their content resulted to be higher than that of MWD samples. The cultivar ‘Aleatico’ appeared to be quite tolerant to water stress conditions and this information could be useful also for setting up targeted postharvest dehydration strategies to produce dessert wines. An additional interesting result is represented by the validation of the use of a portable nondestructive instrument to measure skin anthocyanin content directly in the vineyard
