    JNK Signalling Regulates Self-Renewal of Proliferative Urine-Derived Renal Progenitor Cells via Inhibition of Ferroptosis

    With a global increase in chronic kidney disease patients, alternatives to dialysis and organ transplantation are needed. Stem cell-based therapies could be one possibility to treat chronic kidney disease. Here, we used multipotent urine-derived renal progenitor cells (UdRPCs) to study nephrogenesis. UdRPCs treated with the JNK inhibitor—AEG3482 displayed decreased proliferation and downregulated transcription of cell cycle-associated genes as well as the kidney progenitor markers—SIX2, SALL1 and VCAM1. In addition, levels of activated SMAD2/3, which is associated with the maintenance of self-renewal in UdRPCs, were decreased. JNK inhibition resulted in less efficient oxidative phosphorylation and more lipid peroxidation via ferroptosis, an iron-dependent non-apoptotic cell death pathway linked to various forms of kidney disease. Our study is the first to describe the importance of JNK signalling as a link between maintenance of self-renewal and protection against ferroptosis in SIX2-positive renal progenitor cells

    X-Ray Scattering at FeCo(001) Surfaces and the Crossover between Ordinary and Normal Transitions

    In a recent experiment by Krimmel et al. [PRL 78, 3880 (1997)], the critical behavior of FeCo near a (001) surface was studied by x-ray scattering. Here the experimental data are reanalyzed, taking into account recent theoretical results on order-parameter profiles in the crossover regime between ordinary and normal transitions. Excellent agreement between theoretical expectations and the experimental results is found.Comment: 9 pages, Latex, 1 PostScript figure, to be published in Phys.Rev.

    Technical Note: Characterisation of a DUALER instrument for the airborne measurement of peroxy radicals during AMMA 2006

    A DUALER (dual-channel airborne peroxy radical chemical amplifier) instrument has been developed and optimised for the airborne measurement of the total sum of peroxy radicals during the AMMA (African Monsoon Multidisciplinary Analyses) measurement campaign which took place in Burkina Faso in August 2006. The innovative feature of the instrument is that both reactors are sampling simultaneously from a common pre-reactor nozzle while the whole system is kept at a constant pressure to ensure more signal stability and accuracy. <br><br> Laboratory experiments were conducted to characterise the stability of the NO<sub>2</sub> detector signal and the chain length with the pressure. The results show that airborne measurements using chemical amplification require constant pressure at the luminol detector. Wall losses of main peroxy radicals HO<sub>2</sub> and CH<sub>3</sub>O<sub>2</sub> were investigated. The chain length was experimentally determined for different ambient mixtures and compared with simulations performed by a chemical box model. <br><br> The DUALER instrument was successfully mounted within the German DLR-Falcon. The analysis of AMMA data utilises a validation procedure based on the O<sub>3</sub> mixing ratios simultaneously measured onboard. The validation and analysis procedure is illustrated by means of the data measured during the AMMA campaign. The detection limit and the accuracy of the ambient measurements are also discussed

    Amine-Gold Linked Single-Molecule Junctions: Experiment and Theory

    The measured conductance distribution for single molecule benzenediamine-gold junctions, based on 59,000 individual conductance traces recorded while breaking a gold point contact in solution, has a clear peak at 0.0064 G0_{0} with a width of ±\pm 40%. Conductance calculations based on density functional theory (DFT) for 15 distinct junction geometries show a similar spread. Differences in local structure have a limited influence on conductance because the amine-Au bonding motif is well-defined and flexible. The average calculated conductance (0.046 G0_{0}) is seven times larger than experiment, suggesting the importance of many-electron corrections beyond DFT
