Upregulation of Barrel GABAergic Neurons Is Associated with Cross-Modal Plasticity in Olfactory Deficit
Background: Loss of a sensory function is often followed by the hypersensitivity of other modalities in mammals, which secures them well-awareness to environmental changes. Cellular and molecular mechanisms underlying cross-modal sensory plasticity remain to be documented. Methodology/Principal Findings: Multidisciplinary approaches, such as electrophysiology, behavioral task and immunohistochemistry, were used to examine the involvement of specific types of neurons in cross-modal plasticity. We have established a mouse model that olfactory deficit leads to a whisking upregulation, and studied how GABAergic neurons are involved in this cross-modal plasticity. In the meantime of inducing whisker tactile hypersensitivity, the olfactory injury recruits more GABAergic neurons and their fine processes in the barrel cortex, as well as upregulates their capacity of encoding action potentials. The hyperpolarization driven by inhibitory inputs strengthens the encoding ability of their target cells. Conclusion/Significance: The upregulation of GABAergic neurons and the functional enhancement of neuronal networks may play an important role in cross-modal sensory plasticity. The Genome of Ganderma lucidum Provide Insights into Triterpense Biosynthesis and Wood Degradation
BACKGROUND: Ganoderma lucidum (Reishi or Ling Zhi) is one of the most famous Traditional Chinese Medicines and has been widely used in the treatment of various human diseases in Asia countries. It is also a fungus with strong wood degradation ability with potential in bioenergy production. However, genes, pathways and mechanisms of these functions are still unknown. METHODOLOGY/PRINCIPAL FINDINGS: The genome of G. lucidum was sequenced and assembled into a 39.9 megabases (Mb) draft genome, which encoded 12,080 protein-coding genes and ∼83% of them were similar to public sequences. We performed comprehensive annotation for G. lucidum genes and made comparisons with genes in other fungi genomes. Genes in the biosynthesis of the main G. lucidum active ingredients, ganoderic acids (GAs), were characterized. Among the GAs synthases, we identified a fusion gene, the N and C terminal of which are homologous to two different enzymes. Moreover, the fusion gene was only found in basidiomycetes. As a white rot fungus with wood degradation ability, abundant carbohydrate-active enzymes and ligninolytic enzymes were identified in the G. lucidum genome and were compared with other fungi. CONCLUSIONS/SIGNIFICANCE: The genome sequence and well annotation of G. lucidum will provide new insights in function analyses including its medicinal mechanism. The characterization of genes in the triterpene biosynthesis and wood degradation will facilitate bio-engineering research in the production of its active ingredients and bioenergy
LICSTER -- A Low-cost ICS Security Testbed for Education and Research
Unnoticed by most people, Industrial Control Systems (ICSs) control entire
productions and critical infrastructures such as water distribution, smart grid
and automotive manufacturing. Due to the ongoing digitalization, these systems
are becoming more and more connected in order to enable remote control and
monitoring. However, this shift bears significant risks, namely a larger attack
surface, which can be exploited by attackers. In order to make these systems
more secure, it takes research, which is, however, difficult to conduct on
productive systems, since these often have to operate twenty-four-seven.
Testbeds are mostly very expensive or based on simulation with no real-world
physical process. In this paper, we introduce LICSTER, an open-source low-cost
ICS testbed, which enables researchers and students to get hands-on experience
with industrial security for about 500 Euro. We provide all necessary material
to quickly start ICS hacking, with the focus on low-cost and open-source for
education and research

Achieving stable nitritation for mainstream deammonification by combining free nitrous acid-based sludge treatment and oxygen limitation
Stable nitritation is a critical bottleneck for achieving autotrophic nitrogen removal using the energy-saving mainstream deammonification process. Herein we report a new strategy to wash out both the Nitrospira sp. and Nitrobacter sp. from the treatment of domestic-strength wastewater. The strategy combines sludge treatment using free nitrous acid (FNA) with dissolved oxygen (DO) control in the nitritation reactor. Initially, the nitrifying reactor achieved full conversion of NH4+ to NO3-. Then, nitrite accumulation at similar to 60% was achieved in the reactor when 1/4 of the sludge was treated daily with FNA at 1.82 mg N/L in a side-stream unit for 24 h. Fluorescence in-situ hybridization (FISH) revealed FNA treatment substantially reduced the abundance of nitrite oxidizing bacteria (NOB) (from 23.0 +/- 4.3 to 5.3 +/- 1.9%), especially that of Nitrospira sp. (from 15.7 +/- 3.9 to 0.4 +/- 0.1%). Nitrite accumulation increased to similar to 80% when the DO concentration in the mainstream reactor was reduced from 2.5-3.0 to 0.3-0.8 mg/L. FISH revealed the DO limitation further reduced the abundance of NOB (to 2.1 +/- 1.0%), especially that of Nitrobacter sp. (from 4.9 +/- 1.2 to 1.8 +/- 0.8%).
