    Dynamics of African swine fever virus shedding and excretion in domestic pigs infected by intramuscular inoculation and contact transmission

    African swine fever virus (ASFV) is a highly virulent swine pathogen that has spread across Eastern Europe since 2007 and for which there is no effective vaccine or treatment available. The dynamics of shedding and excretion is not well known for this currently circulating ASFV strain. Therefore, susceptible pigs were exposed to pigs intramuscularly infected with the Georgia 2007/1 ASFV strain to measure those dynamics through within- and between-pen transmission scenarios. Blood, oral, nasal and rectal fluid samples were tested for the presence of ASFV by virus titration (VT) and quantitative real-time polymerase chain reaction (qPCR). Serum was tested for the presence of ASFV-specific antibodies. Both intramuscular inoculation and contact transmission resulted in development of acute disease in all pigs although the experiments indicated that the pathogenesis of the disease might be different, depending on the route of infection. Infectious ASFV was first isolated in blood among the inoculated pigs by day 3, and then chronologically among the direct and indirect contact pigs, by day 10 and 13, respectively. Close to the onset of clinical signs, higher ASFV titres were found in blood compared with nasal and rectal fluid samples among all pigs. No infectious ASFV was isolated in oral fluid samples although ASFV genome copies were detected. Only one animal developed antibodies starting after 12 days post-inoculation. The results provide quantitative data on shedding and excretion of the Georgia 2007/1 ASFV strain among domestic pigs and suggest a limited potential of this isolate to cause persistent infection

    Scaling in the crossover from random to correlated growth

    In systems where deposition rates are high compared to diffusion, desorption and other mechanisms that generate correlations, a crossover from random to correlated growth of surface roughness is expected at a characteristic time t_0. This crossover is analyzed in lattice models via scaling arguments, with support from simulation results presented here and in other authors works. We argue that the amplitudes of the saturation roughness and of the saturation time scale as {t_0}^{1/2} and t_0, respectively. For models with lateral aggregation, which typically are in the Kardar-Parisi-Zhang (KPZ) class, we show that t_0 ~ 1/p, where p is the probability of the correlated aggregation mechanism to take place. However, t_0 ~ 1/p^2 is obtained in solid-on-solid models with single particle deposition attempts. This group includes models in various universality classes, with numerical examples being provided in the Edwards-Wilkinson (EW), KPZ and Villain-Lai-Das Sarma (nonlinear molecular-beam epitaxy) classes. Most applications are for two-component models in which random deposition, with probability 1-p, competes with a correlated aggregation process with probability p. However, our approach can be extended to other systems with the same crossover, such as the generalized restricted solid-on-solid model with maximum height difference S, for large S. Moreover, the scaling approach applies to all dimensions. In the particular case of one-dimensional KPZ processes with this crossover, we show that t_0 ~ nu^{-1} and nu ~ lambda^{2/3}, where nu and lambda are the coefficients of the linear and nonlinear terms of the associated KPZ equations.     SUPLEMENTAÇÃO FOSTATADA PARA BOVINOS DE CORTE EM CAMPOS NATURAIS DA SERRA DO SUDESTE, RIO GRANDE DO SUL - Nos campos naturais da Região agroecológica Serra do Sudeste, foi conduzido um estudo abrangente sobre a influência de fatores de solo e de clima sobre a dinâmica estacional da disponibilidade de minerais na cadeia solo-pastagem-animal. O estudo foi realizado em duas propriedades rurais no 2º (Área 1, Fazenda Esperança) e no 4º (Área 2, Fazenda São Thomaz) sub-distritos do município de Piratini, RS, cujas características de solo, da vegetação e topográficas são representativas da Região. A interpretação dos teores minerais no solo indicou que apenas o fósforo e o sódio são os elementos em deficiência. O estudo revelou que os solos da região apresentam baixos teores de fósforo durante o ano todo, sendo menores no inverno e maiores no verão. Os teores de fósforo na vegetação do campo natural são insuficientes para suprir as exigências nutricionais dos animais em pastejo. Assim sendo, os teores de fósforo no soro de sangue dos animais em pastejo estão abaixo do limite de normalidade no verão, outono e inverno; na primavera encontra-se no limite da normalidade para novilhas (8,0 mg/100 ml). Como foi evidente a deficiência de fósforo para os animais na maior parte do ano, recomenda-se a suplementação com fósforo durante todo o ano. Em função dos baixos teores de sódio no solo e na forragem, o fornecimento de misturas minerais contendo cloreto de sódio + fósforo é imprescindível. Ou então misturas com sais proteinados + fósforo, para ajudar a utilização deste nutriente em períodos de deficiência de proteína, como no inverno.bitstream/item/31322/1/comunicado-170.pd

    Endometriosis: A Rare Cause of Large Bowel Obstruction.

    Large bowel obstruction can result in significant morbidity and mortality, especially in cases of acute complete obstruction. There are many possible causes, the most common in adults being colorectal cancer. Endometriosis is a benign disease, and the most affected extragenital location is the bowel, especially the rectosigmoid junction. However, transmural involvement and acute occlusion are very rare events. We report an exceptional case of acute large bowel obstruction as the initial presentation of endometriosis. The differential diagnosis of colorectal carcinoma may be challenging, and this case emphasizes the need to consider intestinal endometriosis in females at a fertile age presenting with gastrointestinal symptoms and an intestinal mass causing complete large bowel obstruction.info:eu-repo/semantics/publishedVersio

    SP-Sephadex equilibrium chromatography of bradykinin and related peptides: Application to trypsin-treated human plasma

    An analytical method is deseribed for the separation of bradykinin, Lys-bradykinin, and Met-Lys-bradykinin by equilibrium chromatography on SP-Sephadex C-25 eluted in 0.02 Tris-HCl buffer, pH 8.10, 0.12 NaCl. A second elution buffer, 0.02 Tris-HCl buffer, pH 7.70, 0.06 NaCl, serves as a second parameter for the identification of bradykinin and also separates the hormone from plasma bradykinin-potentiating peptides. Ten to one-hundred nanomoles of each peptide can be recovered in high yields, identified by elution position, and measured by bioassay with the isolated guinea pig ileum. The identification of bradykinin as the peptide released by trypsin acting on acid-denatured plasma is documented as an illustration of the method

    Contribution of Trf4/5 and the Nuclear Exosome to Genome Stability Through Regulation of Histone mRNA Levels in Saccharomyces cerevisiae

    Balanced levels of histones are crucial for chromosome stability, and one major component of this control regulates histone mRNA amounts. The Saccharomyces cerevisiae poly(A) polymerases Trf4 and Trf5 are involved in a quality control mechanism that mediates polyadenylation and consequent degradation of various RNA species by the nuclear exosome. None of the known RNA targets, however, explains the fact that trf mutants have specific cell cycle defects consistent with a role in maintaining genome stability. Here, we investigate the role of Trf4/5 in regulation of histone mRNA levels. We show that loss of Trf4 and Trf5, or of Rrp6, a component of the nuclear exosome, results in elevated levels of transcripts encoding DNA replication-dependent histones. Suggesting that increased histone levels account for the phenotypes of trf mutants, we find that TRF4 shows synthetic genetic interactions with genes that negatively regulate histone levels, including RAD53. Moreover, synthetic lethality of trf4Δ rad53Δ is rescued by reducing histone levels whereas overproduction of histones is deleterious to trf's and rrp6Δ mutants. These results identify TRF4, TRF5, and RRP6 as new players in the regulation of histone mRNA levels in yeast.
