    A novel single amino acid deletion impairs fibronectin function and causes familial glomerulopathy with fibronectin deposits: case report of a family

    Abstract Background Glomerulopathy with fibronectin deposits is an autosomal dominant disease associated with proteinuria, hematuria, hypertension and renal function decline. Forty percent of the cases are caused by mutations in FN1, the gene that encodes fibronectin. Case presentation This report describes two cases of Glomerulopathy with fibronectin deposits, involving a 47-year-old father and a 14-year-old son. The renal biopsies showed glomeruli with endocapillary hypercellularity and large amounts of mesangial and subendothelial eosinophilic deposits. Immunohistochemistry for fibronectin was markedly positive. Whole exome sequencing identified a novel FN1 mutation that leads to an amino-acid deletion in both patients (Ile1988del), a variant that required primary amino-acid sequence analysis for assessment of pathogenicity. Our primary sequence analyses revealed that Ile1988 is very highly conserved among relative sequences and is positioned in a C-terminal FN3 domain containing heparin- and fibulin-1-binding sites. This mutation was predicted as deleterious and molecular mechanics simulations support that it can change the tertiary structure and affect the complex folding and its molecular functionality. Conclusion The current report not only documents the occurrence of two GFND cases in an affected family and deeply characterizes its anatomopathological features but also identifies a novel pathogenic mutation in FN1, analyzes its structural and functional implications, and supports its pathogenicity.https://deepblue.lib.umich.edu/bitstream/2027.42/152212/1/12882_2019_Article_1507.pd

    ALDH1A2 (RALDH2) genetic variation in human congenital heart disease

    Abstract\ud \ud \ud \ud Background\ud \ud Signaling by the vitamin A-derived morphogen retinoic acid (RA) is required at multiple steps of cardiac development. Since conversion of retinaldehyde to RA by retinaldehyde dehydrogenase type II (ALDH1A2, a.k.a RALDH2) is critical for cardiac development, we screened patients with congenital heart disease (CHDs) for genetic variation at the ALDH1A2 locus.\ud \ud \ud \ud Methods\ud \ud One-hundred and thirty-three CHD patients were screened for genetic variation at the ALDH1A2 locus through bi-directional sequencing. In addition, six SNPs (rs2704188, rs1441815, rs3784259, rs1530293, rs1899430) at the same locus were studied using a TDT-based association approach in 101 CHD trios. Observed mutations were modeled through molecular mechanics (MM) simulations using the AMBER 9 package, Sander and Pmemd programs. Sequence conservation of observed mutations was evaluated through phylogenetic tree construction from ungapped alignments containing ALDH8 s, ALDH1Ls, ALDH1 s and ALDH2 s. Trees were generated by the Neighbor Joining method. Variations potentially affecting splicing mechanisms were cloned and functional assays were designed to test splicing alterations using the pSPL3 splicing assay.\ud \ud \ud \ud Results\ud \ud We describe in Tetralogy of Fallot (TOF) the mutations Ala151Ser and Ile157Thr that change non-polar to polar residues at exon 4. Exon 4 encodes part of the highly-conserved tetramerization domain, a structural motif required for ALDH oligomerization. Molecular mechanics simulation studies of the two mutations indicate that they hinder tetramerization. We determined that the SNP rs16939660, previously associated with spina bifida and observed in patients with TOF, does not affect splicing. Moreover, association studies performed with classical models and with the transmission disequilibrium test (TDT) design using single marker genotype, or haplotype information do not show differences between cases and controls.\ud \ud \ud \ud Conclusion\ud \ud In summary, our screen indicates that ALDH1A2 genetic variation is present in TOF patients, suggesting a possible causal role for this gene in rare cases of human CHD, but does not support the hypothesis that variation at the ALDH1A2 locus is a significant modifier of the risk for CHD in humans.Work supported by grants from Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) 01/000090; 00/030722; 01/142381; 02/113402; 03/099982; 04/116068; 04/157044 and Conselho Nacional de Desenvolvimento Científico e Tecnológico 481872/20078.     Risk assessment of work-related musculoskeletal disorders at a workplace from metalworking industry

    With constant changes at work, industries demands workers to perform small movements with high frequencies. The repetitive movements are frequently present in metalworking industry and constitute an important Work-Related Upper Limb Musculoskeletal Disorders (WRMSD) risk factor. The main goals of this study were: (i) asses the risk of developing WRMSD, by applying the method Rapid Upper Limbs Assessment (RULA); (ii) suggest possible preventive measures; (iii) with RULA action level results, rank priorities for implementing these measures; (iv) explore if the implemented rotation scheme decreases risk of developing WRMSD. (v) examine association of pain, discomfort and nuisance reported by workers. with presence of WRMSD risk factors in the workplace, through Nordic Musculoskeletal Questionnaire and Ergonomic Workplace Analysis application RULA’ results showed that these workers are exposed to a high risk of WRMSD development, being in agreement with reported symptoms pointed out by the questionnaire applied. Additionationally, the obtained results demonstrated that the task rotation scheme seems to be insufficient relatively to the WRMSD prevention. Therefore, a more complete study is needed in order to achieve better ergonomic interventions on workplaces of metalworking industry.(undefined)info:eu-repo/semantics/publishedVersio

    Illuminance and whole body vibration evaluation in a company from footwear industry in Portugal

    The work in storage areas, using mechanical means of handling loads, as well as office work, may lead employee's to multiple risks exposure. Therefore, the aim of this study was to assess whole body vibration (WBV) exposure of employees at a footwear industry, as well as to determine the illuminance levels in different tasks. The results were compared with the recommended levels in Portuguese legislation, Decree-Law number 46/2006, for WBV, and in ISO 8995-1: 2002, for illuminance. With the obtained results, effects on employees' health, safety, comfort and productivity due to these physical agents were analysed, being suggested improvement recommendations. As a result of the environmental assessment it was found that, for some tasks done at Storage, the mean values of illuminance on the working plan, and respective neighbourhood, are lower than the recommended value established in ISO 8995-1: 2002. Regarding occupational exposure to WBV, it was concluded that employees are not exposed to values above the limit value established in Portuguese legislation
