    Technological Progress in Generation of Induced Pluripotent Stem Cells for Clinical Applications

    Reprogramming of somatic cells into induced pluripotent stem cells (iPSCs) is achieved by viral-mediated transduction of defined transcription factors. Generation of iPSCs is of great medical interest as they have the potential to be a source of patient-specific cells. For the eventual goal of clinical application, it is necessary to overcome the limitations of low reprogramming efficiency and chromosomal abnormalities due to viral DNA integration. In this paper, we summarize the current state of reprogramming technology for generation of iPSCs and also discuss potential approaches to the development of safe iPSCs for personalized cell-based replacement therapy

    CD9, a Major Platelet Cell Surface Glycoprotein, is a ROCA Antigen and Is Expressed in the Nervous System

    We previously generated a monoclonal antibody (mAb), ROCA1, which binds preferentially to rostral versus caudal sympathetic ganglia and intercostal nerves. Two other mAbs, ROCA2 and B2C11, bind to the same structures but not in rostrocaudal gradients. All three mAbs recognize a 26 kDa cell surface protein. Amino acid sequence data obtained from the affinity purified 26 kDa protein showed some homology with human CD9, a tetraspan protein implicated in intercellular signaling in hematopoietic cells. Using the PCR, we obtained cDNA clones representing the entire rat CD9 coding sequence from sciatic nerve and sympathetic ganglia. ROCA1, ROCA2, and B2C11 each immunoprecipitate a 26 kDa protein from CHO cells stably transfected with one of the clones, demonstrating that the ROCA cell surface antigen is indeed rat CD9. We find that CD9 mRNA is widely expressed, with particularly high levels present in a number of neural tissues. In situ hybridization demonstrates that peripheral neurons and Schwann cells, as well as adrenal chromaffin cells express CD9 mRNA. Consistent with immunoblot analyses showing that, unlike the ROCA1 epitope, the 26 kDa protein is not expressed in a rostrocaudal gradient, we find similar levels of rat CD9 mRNA in rostral and caudal intercostal nerves. In developing postnatal rat sciatic nerve, CD9 mRNA levels are coordinately regulated with the expression of myelin genes. These results provide another example of a cell surface protein expressed by both hematopoietic and neural cells, and suggest a role for CD9 in intercellular signaling in the nervous system

    Comparison of quality of life and related factors according to premature menopause using the Korea National Health and Nutrition Examination Survey (2014–2017)

    Purpose The purpose of this study was to compare the health‐related quality of life of premature menopausal women with that of normal postmenopausal women and to identify factors related to quality of life in these two groups. Methods A secondary analysis was conducted of a sample of 5,910 postmenopausal women drawn from women aged 19–79 years, using raw data from the Korea National Health and Nutrition Survey between 2014 and 2017. Quality of life was compared between premature menopausal women and normal postmenopausal women using the t‐test and chi‐square test, and factors related to quality of life were analyzed by multivariate logistic regression using SAS version 9.4. Results There was no significant difference in quality of life between premature menopausal women and normal postmenopausal women. Subjective health status and symptoms of depression were identified as factors related to quality of life in women with premature menopause, while engaging in aerobic physical activity, subjective health status, stress, and symptoms of depression were found to be related to quality of life among normal postmenopausal women. Conclusion It is necessary to support self‐care measures for both premature and normal menopausal women to adapt to changes caused by menopause, and multidimensional strategies should be developed considering postmenopausal women’s subjective health status and symptoms of depression to improve their quality of life

    Cell Type-Specific Responses to Wingless, Hedgehog and Decapentaplegic Are Essential for Patterning Early Eye-Antenna Disc in Drosophila

    The Drosophila eye-antenna imaginal disc (ead) is a flattened sac of two-layered epithelia, from which most head structures are derived. Secreted morphogens like Wingless (Wg), Hedgehog (Hh), and Decapentaplegic (Dpp) are important for early patterning of ead, but the underlying mechanisms are still largely unknown. To understand how these morphogens function in the ead of early larval stages, we used wg-LacZ and dpp-Gal4 markers for the examination of wild-type and mutant eads. We found that the ead immediately after hatching was crescent-shaped with the Bolwig’s nerve at the ventral edge, suggesting that it consists of dorsal domain. In a subsequent step, transcriptional induction of dpp in the cells along the Bolwig’s nerve was followed by rapid growth of the ventral domain. Both Wg and Hh were required for the formation of the ventral domain. Wg was crucial for the growth of the entire ead, but Hh was essential for cell division only in the dorsal domain. In the ventral domain, Hh regulated dpp transcription. Based on these data, we propose that signaling among distinct groups of cells expressing Wg, Dpp, or Hh in the ead of the first-instar larvae are critical for coordinated growth and patterning of ead

    Assessment of vitamin B6 status in Korean patients with newly diagnosed type 2 diabetes

    The purpose of this study was to assess vitamin B6 intake and status in Korean patients with newly diagnosed type 2 diabetes. Sixty-four patients with newly diagnosed type 2 diabetes and 8-11% glycated hemoglobin (A1C), along with 28 age-matched non-diabetic subjects, participated. Dietary vitamin B6 intake was estimated by the 24 hour recall method and plasma pyridoxal 5'-phosphate (PLP) was measured. There was a significant difference in daily total calorie intake between the diabetic and non-diabetic groups (1,917 ± 376 vs 2,093 ± 311 kcal). There were no differences in intake of total vitamin B6 (2.51 ± 0.91 vs 2.53 ± 0.81 mg/d) or vitamin B6/1,000 kcal (1.31 ± 0.42 vs 1.20 ± 0.32 mg) between the diabetic and non-diabetic groups, andI intakes of total vitamin B6 were above the Korean RDA in both groups (180.0 ± 57.9 vs 179.0 ± 65.4). There was a higher percentage of diabetic subjects whose plasma PLP concentration was < 30 nmol/L compared to non-diabetic group. Plasma PLP levels tended to be lower in the diabetic subjects than in the non-diabetic subjects, although the difference was not statistically significant due to a large standard deviation (80.0 ± 61.2 nmol/L vs 68.2 ± 38.5 nmol/L). Nevertheless, plasma PLP levels should be monitored in pre-diabetic patients with diabetic risk factors as well as in newly diagnosed diabetic patients for long-term management of diabetes, even though this factor is not a major risk factor that contributes to the development of degenerative complications in certain patients

    Phenotyping of rice in salt stress environment using high-throughput infrared imaging

    Phenotyping of rice (Oryza sativa L. cv. Donggin) in salt stress environment using infrared imaging was conducted. Results were correlated with the most frequently used physiological parameters such as stomatal conductance, relative water content and photosynthetic parameters. It was observed that stomatal conductance (R2 = –0.618) and relative water content (R2 = –0.852) were significantly negatively correlated with average plant temperature (thermal images), while dark-adapted quantum yield (Fv/Fm, R2 = –0.325) and performance index (R2 = –0.315) were not consistent with plant temperature. Advantages of infrared thermography and utilization of this technology for the selection of stress tolerance physiotypes are discussed in detail
