    Survey of indoor air quality in residences of WIC participants in Lincoln, Nebraska

    Since 1992, Women, Infants, and Children at the Lincoln Lancaster County Health Department has provided supplemental food benefits, nutrition information, breastfeeding support, immunization referrals, and dental services to support low-income families and their children. However, statewide, the retention rate has been dropping since 2009, and if this continues, it can result in adverse health outcomes. As part of the Service Learning activities, this study aims to evaluate current services on clients’ experiences and improve their satisfaction by conducting the WIC participant-centered services survey. This cost-effective intervention program has been working successfully, however it does not provide the information of environmental health. To understand WIC enrolled families’ home environment, as part of the Capstone Experience we conducted the indoor air quality survey to investigate existing indoor air contaminants and related health problems. This survey was a questionnaire designed to collect information of sources of air pollution in homes (cleaning practices, natural and forced ventilation, lead, mold, tobacco smoke, etc.) and resident’s respiratory symptoms. The data was collected by face-to-face interviews at the two WIC clinics, LLCHD and satellite office, from 50 individuals who have lived in their current residence more than six months. The results were used to identify present residential air contaminants that can cause respiratory symptoms. Health effects caused by indoor air pollution is preventable by good maintenance and housekeeping practices. The goal of the project is to increase the mother’s knowledge about indoor air quality in low-income communities and support them to reduce contaminants for better health outcomes. Ultimately, the information collected by surveys can be used to plan a support program that increases mother’s awareness of air pollutants and prevention

    Alpha-tocopherol exerts protective function against the mucotoxicity of particulate matter in amphibian and human goblet cells

    Exposure to particulate matter (PM) in ambient air is known to increase the risk of cardiovascular disorders and mortality. The cytotoxicity of PM is mainly due to the abnormal increase of reactive oxygen species (ROS), which damage cellular components such as DNA, RNA, and proteins. The correlation between PM exposure and human disorders, including mortality, is based on long-term exposure. In this study we have investigated acute responses of mucus-secreting goblet cells upon exposure to PM derived from a heavy diesel engine. To this end, we employed the mucociliary epithelium of amphibian embryos and human Calu-3 cells to examine PM mucotoxicity. Our data suggest that acute exposure to PM significantly impairs mucus secretion and results in the accumulation of mucus vesicles in the cytoplasm of goblet cells. RNA-seq analysis revealed that acute responses to PM exposure significantly altered gene expression patterns; however, known regulators of mucus production and the secretory pathway were not significantly altered. Interestingly, pretreatment with alpha-tocopherol nearly recovered the hyposecretion of mucus from both amphibian and human goblet cells. We believe this study demonstrates the mucotoxicity of PM and the protective function of alpha-tocopherol on mucotoxicity caused by acute PM exposure from heavy diesel engines

    Improved Surgical Technique for Heterotopic Aortic Transplantation in Mice

    Transplant arteriosclerosis is the main limitation for long-term survival of solid organ transplant recipients. Animal models would provide invaluable tools to investigate the cellular and molecular mechanisms underlying the pathogenesis of transplant arteriosclerosis, as well as for studies with novel drugs and other reagents for the prevention of the disease. We have therefore developed a modified technique for aortic transplantation in mice. The central suture ligation of the recipient abdominal aorta allowed a simpler end-to-side anastomosis of a segment of the donor thoracic aorta into the infrarenal portion of the recipient abdominal aorta. Using this technique, the overall survival rate was 94%. We also observed typical aspects of chronic rejection of the aortic allografts not observed with isografts. Our new technique is relatively easy to perform and has a low incidence of thrombosis, thus being useful for studying various aspects of transplant arteriosclerosis

    Microcurrent therapy as the nonpharmacological new protocol against Alzheimer’s disease

    IntroductionAlzheimer’s disease (AD) poses an increasing global health challenge and is marked by gradual cognitive deterioration, memory impairment, and neuroinflammation. Innovative therapeutic approaches as non-pharmacological protocol are urgently needed with side effect risk of drugs. Microcurrent therapy, a non-invasive modality involving low-level electrical currents, has emerged as a potential solution to address AD’s complex pathogenesis. This study investigates the optimal application of microcurrent therapy as a clinical protocol for AD, utilizing a comprehensive approach that integrates behavioral assessments and neuroinflammation evaluation in a mouse model of dementia.Methods and resultsThe results reveal that microcurrent therapy holds promise in ameliorating memory impairment and reducing neuroinflammation in AD. Behavioral assessments, including the Novel Object Recognition Test (NOR) and Radial Arm Maze Test (RAM), demonstrated improved cognitive function following microcurrent therapy. Furthermore, microcurrent therapy inhibited expression of neuroinflammatory proteins, including ionized calcium binding adaptor molecule 1 (Iba1), and glial fibrillary acidic protein (GFAP) in current-treated group. Mechanistic insights suggest that microcurrent therapy may modulate neuroinflammation through the regulation of MAPK signaling pathways.ConclusionThis study emphasizes the prospect of microcurrent therapy as a safe and efficacious non-pharmacological strategy for Alzheimer’s disease (AD), providing optimism to the countless individuals impacted by this debilitating ailment. These results contribute to the developments of an innovative clinical protocol for AD and recovery from neurological injury, underscoring the significance of investigating unconventional therapeutic approaches for addressing this complex condition

    TRAO Survey of Nearby Filamentary Molecular clouds, the Universal Nursery of Stars (TRAO FUNS) I. Dynamics and Chemistry of L1478 in the California Molecular Cloud

    "TRAO FUNS" is a project to survey Gould Belt's clouds in molecular lines. This paper presents its first results on the central region of the California molecular cloud, L1478. We performed On-The-Fly mapping observations using the Taedeok Radio Astronomy Observatory (TRAO) 14m single dish telescope equipped with a 16 multi-beam array covering \sim1.0 square degree area of this region using C18^{18}O (1-0) mainly tracing low density cloud and about 460 square arcminute area using N2_{2}H+^{+} (1-0) mainly tracing dense cores. CS (2-1) and SO (3221)(3_{2}-2_{1}) were also used simultaneously to map \sim440 square arcminute area of this region. We identified 10 filaments by applying the dendrogram technique to the C18^{18}O data-cube and 8 dense N2_{2}H+^{+} cores by using {\sc FellWalker}. Basic physical properties of filaments such as mass, length, width, velocity field, and velocity dispersion are derived. It is found that L1478 consists of several filaments with slightly different velocities. Especially the filaments which are supercritical are found to contain dense cores detected in N2_{2}H+^{+}. Comparison of non-thermal velocity dispersions derived from C18^{18}O and N2_{2}H+^{+} for the filaments and dense cores indicates that some of dense cores share similar kinematics with those of the surrounding filaments while several dense cores have different kinematics with those of their filaments. This suggests that the formation mechanism of dense cores and filaments can be different in individual filaments depending on their morphologies and environments.Comment: 25 pages, 15 figures, accepted for publication in Ap

    HIV-1 subtypes and drug resistance mutations among female sex workers varied in different cities and regions of the Democratic Republic of Congo

    Background Complex mosaic structures of HIV-1 were found in the Democratic Republic of Congo (DRC). Currently, there is limited information on the circulating HIV-1 strains, the distribution of these strains and antiretroviral (ART) resistant viruses in different regions of the country, and the HIV-1 strains harbored by the high-risk groups like female sex workers (FSW) reported to be the source of recombinant and ART resistant viruses. Methods Dried Blood Spots (DBS), collected from 325 infected FSWs in ten cities from 2012 DRC HIV/STI Integrated Biological and Behavioral Surveillance Survey, were tested for HIV-1 genotypes and antiretroviral resistance mutations. Regional segregation of HIV-1 clades was detected using phylogenetics. The significance for differences in HIV-1 subtype and drug resistance mutations were evaluated using Chi-square tests. Results There were 145 (env) and 93 (pol) sequences analyzed. Based on env sequences, the predominant subtype was A1 (44%), and recombinants as defined pol sequences comprised 35% of the total sample. Paired sequences of pol and env from DRC FSW revealed mosaic recombinant in 54% of the sequences. Distinct geographic distributions of different HIV-1 subtypes and recombinants were observed.     Mitogenesis of Vascular Smooth Muscle Cell Stimulated by Platelet-Derived Growth Factor-bb Is Inhibited by Blocking of Intracellular Signaling by Epigallocatechin-3- O

    Epigallocatechin gallate (EGCG) is known to exhibit antioxidant, antiproliferative, and antithrombogenic effects and reduce the risk of cardiovascular diseases. Key events in the development of cardiovascular disease are hypertrophy and hyperplasia according to vascular smooth muscle cell proliferation. In this study, we investigated whether EGCG can interfere with PDGF-bb stimulated proliferation, cell cycle distribution, and the gelatinolytic activity of MMP and signal transduction pathways on RAOSMC when it was treated in two different ways—cotreatment with PDGF-bb and pretreatment of EGCG before addition of PDGF-bb. Both cotreated and pretreated EGCG significantly inhibited PDGF-bb induced proliferation, cell cycle progression of the G0/G1 phase, and the gelatinolytic activity of MMP-2/9 on RAOSMC. Also, EGCG blocked PDGF receptor-β (PDGFR-β) phosphorylation on PDGF-bb stimulated RAOSMC under pretreatment with cells as well as cotreatment with PDGF-bb. The downstream signal transduction pathways of PDGFR-β, including p42/44 MAPK, p38 MAPK, and Akt phosphorylation, were also inhibited by EGCG in a pattern similar to PDGFR-β phosphorylation. These findings suggest that EGCG can inhibit PDGF-bb stimulated mitogenesis by indirectly and directly interrupting PDGF-bb signals and blocking the signaling pathway via PDGFR-β phosphorylation. Furthermore, EGCG may be used for treatment and prevention of cardiovascular disease through blocking of PDGF-bb signaling
