    Pronounced activation of protein kinase C, ornithine decarboxylase and c-jun proto-oncogene by paraquat-generated active oxygen species in WI-38 human lung cells

    AbstractParaquat (methyl viologen, PQ) is a widely used herbicide that produces oxygen-derived free radicals and severely injures human lungs. In this study we examined the effects of PQ on the protein kinase C (PKC), ornithine decarboxylase (ODC) and c-jun oncogene expression in WI-38 human lung cells. Exposure of cells to 25–200 μM PQ resulted in an increase of [3H]phorbol dibutyrate (PDBu) binding and PKC redistribution in a dose-dependent manner. Interestingly, a superoxide dismutase mimic, 4-hydroxyl-2,2,6,6-tetramethylpiperidine-1-oxyl (Tempol, 2.5 mM) and catalase (400 μg/ml) could significantly reduce the PQ-stimulated increase of phorbol ester binding and particular PKC phosphorylatiog activity, but dimethylsulfoxide (DMSO, 1.5%), an effective ·OH trapping agent, failed to prevent this stimulation. In addition, an endogenous substrate of PKC, 80 kDa protein, was found to be highly phosphorylated in intact WI-38 cells treated with 50 AM PQ. The increase of phosphorylated proteins could be completely or partly abolished by Tempol or catalase, but only the phosphorylation of 80 kDa protein was diminished by protein kinase C inhibitor, 1-(5-isoquinolinyl-sulfonyl)-2-methylpiperazine (H-7). A maximal peak of ODC activity was observed at 6 h of treatment with 50 μM PQ. PQ induced activity was reduced at the following rates, Tempol 85%, DMSO 80% and catalase 45%, but H-7 failed to do so. Furthermore, we found that the level of c-jun mRNA was transiently increased by PQ and the peak appeared at 1 h of treatment. When correlated with the PKC result, Tempol, catalase and H-7 all effectively blocked PQ-elicited c-jun transcript expression, but DMSO only exhibited a weakly inhibitory effect. We therefore propose that superoxide anion (O2− and H2O2 generated by PQ could activate PKC and lead to induction of c-jun gene expression; on the other hand, O2− and ·OH might trigger other kinase pathways to elevate ODC activity. Finally, the sequential expression of c-jun oncogene, and ODC may cooperate to relieve the oxidative damages elicited by PQ

Geophysical Research Letters, 48(18), (2021): e2021GL094104, https://doi.org/10.1029/2021GL094104.Interannual variations in global mean sea level (GMSL) closely correlate with the evolution of El Niño-Southern Oscillation. However, GMSL differences occur in extreme El Niños; for example, in the 2015–2016 and 1997–1998 El Niños, the peak GMSL during the mature stage of the former (9.00 mm) is almost 2.5 times higher than the latter (3.72 mm). Analyses from satellite and reanalysis data sets show that the disparity in GMSL is primarily due to barystatic (ocean mass) changes. We find that the 2015–2016 event developed not purely as an Eastern Pacific El Niño event but with Central Pacific (CP) El Niño forcing. CP El Niños contribute to a stronger negative anomaly of global terrestrial water storage and subsequent higher barystatic heights. Our results suggest that the mechanism of hydrology-related interannual variations of GMSL should be further emphasized, as more CP El Niño events are projected to occur.This study was supported by a grant of MOST 106-2111-M-002-010-MY4 to National Taiwan University

    Site classification and Vs30 estimation of free-field TSMIP stations using the logging data of EGDT

    The Engineering Geological Database for TSMIP (EGDT), the Taiwan Strong Motion Instrumentation Program, has been under construction by the National Center for Research on Earthquake Engineering and the Central Weather Bureau in Taiwan since 2000. Site characterization, comprising surface investigations and logging measurements, was carried out throughout the project. We provide a set of specifications and a description to help users understand the subject matter of the database. EGDT contains 469 surveyed stations, 439 of which were drilled and the logging measurements completed. Of these, 385 had logging data reaching at least 30 m, and we used these to examine and determine the most accurate extrapolation of Vs30 (the average S-wave velocity of the top 30 m of strata) for the other 54 stations with velocity profiles less than 30 m. The chosen method assumed that the bottom velocity is identical from the actual depth of the hole to a distance of 30 m, that is, the Bottom Constant Velocity (BCV) method. In order to utilize other existing boreholes which have only N values but no velocities in the future, the empirical S-wave velocity equations for seven different regions and the whole of Taiwan were evaluated by a multivariable analysis. Henceforth, for those existing boreholes which have an N profile less than 30 m, the S-wave velocity profile can first be calculated by empirical S-wave velocity equations, and then Vs30 can be estimated by reliable extrapolation. Some other studies of site classifications of TSMIP stations were compared with our results to demonstrate the necessity of reclassification. Ultimately, the Vs30 values of the 439 drilled free-field TSMIP stations were derived and the new site classification was achieved according to the Vs30-based provisions of the National Earthquake Hazards Reduction Program

    Simulating Shallow Soil Response Using Wave Propagation Numerical Modelling in the Western Plain of Taiwan

    This study used the results from 45 microtremor array measurements to construct a shallow shear wave velocity structure in the western plain of Taiwan. We constructed a complete 3D velocity model based on shallow and tomography models for our numerical simulation. There are three major subsurfaces, engineering bedrock (VS = 600 m s-1), Pliocene formation and Miocene formation, constituted in the shallow model. The constant velocity is given in each subsurface. We employed a 3D-FD (finite-differences) method to simulate seismic wave propagation in the western plain. The aim of this study was to perform a quantitative comparison of site amplifications and durations obtained from empirical data and numerical modelling in order to obtain the shallow substructure soil response. Modelling clearly revealed that the shallow substructure plays an important role in strong ground motion prediction using 3D simulation. The results show significant improvements in effective shaking duration and the peak ground velocity (PGV) distribution in terms of the accuracy achieved by our developed model. We recommend a high-resolution shallow substructure as an essential component in future seismic hazard analyses

    HBV replication is significantly reduced by IL-6

    Interleukin-6 (IL-6) is a pleiotropic cytokine with pivotal functions in the regulation of the biological responses of several target cells including hepatocytes. The level of serum IL-6 has been reported to be elevated in patients with chronic hepatitis B, cirrhosis and hepatocellular carcinoma and represents the best marker of HBV-related clinical progression as compared with several other cytokines. In this study, we found that IL-6 was able to effectively suppress hepatitis B virus (HBV) replication and prevent the accumulation of HBV covalently closed circular DNA (cccDNA) in a human hepatoma cell line. We also demonstrated that the suppression of HBV replication by IL-6 requires concurrently a moderate reduction of viral transcripts/core proteins and a marked decrease in viral genome-containing nucleocapsids. Studies on the stability of existing viral capsids suggest that the IL-6 effect on the reduction of genome-containing nucleocapsids is mediated through the prevention of the formation of genome-containing nucleocapsids, which is similar to the effect of interferons. However, IFN-α/β and IFN-γ did not participate in the IL-6-induced suppression of HBV replication. Taken together, our results will provide important information to better understand the role of IL-6 in the course of HBV infection

    A randomized controlled trial of a homeâ based training programme to decrease depression in family caregivers of persons with dementia

    AimsThe aim of this study was to explore distinct trajectories of caregiversâ depressive symptoms and the effects of a training programme on these trajectories over 18 months after the programme.BackgroundOverall effects of caregiverâ training programmes on family caregiversâ depressive symptoms have been reported, but few studies explored distinct courses of changes in caregiversâ depressive symptoms and followed up intervention effects on these distinct courses.DesignRandomized clinical trial.MethodsFamily caregivers (n = 116) were randomly assigned into experimental (n = 57) and control (n = 59) groups. The experimental group received the training programme with telephone consultation and the control group received written educational materials and social telephone followâ ups. Caregiversâ depressive symptoms were assessed from June 2009 â March 2012 by selfâ completed questionnaires before, at 2 weeks and 3, 6, 12 and 18 months after the intervention. Groups of individual trajectories were distinguished using groupâ based trajectory modelling.ResultsCaregiversâ depressive symptoms fell into three stable trajectories: nonâ depressed, mildly blue and depressed. After controlling for covariates, caregivers who received the caregiverâ training programme were less likely than those who did not experience persistent depressive symptoms (b = â 1·92, odds ratio = 0·15, P < 0·05).ConclusionDepressive symptoms of family caregivers of persons with dementia were relatively stable and followed three distinct courses: nonâ depressed, mildly blue and depressed. Therefore, caregiversâ depressive symptoms should be assessed as early as possible. Caregivers in the experimental group had a lower probability of persistent depressive symptoms than caregivers in the control group. Therefore, this training programme can be used by healthcare providers for persons with dementia and their caregivers.     MG63 Osteoblast-Like Cells Exhibit Different Behavior when Grown on Electrospun Collagen Matrix versus Electrospun Gelatin Matrix

    Electrospinning is a simple and efficient method of fabricating a non-woven polymeric nanofiber matrix. However, using fluorinated alcohols as a solvent for the electrospinning of proteins often results in protein denaturation. TEM and circular dichroism analysis indicated a massive loss of triple-helical collagen from an electrospun collagen (EC) matrix, and the random coils were similar to those found in gelatin. Nevertheless, from mechanical testing we found the Young's modulus and ultimate tensile stresses of EC matrices were significantly higher than electrospun gelatin (EG) matrices because matrix stiffness can affect many cell behaviors such as cell adhesion, proliferation and differentiation. We hypothesize that the difference of matrix stiffness between EC and EG will affect intracellular signaling through the mechano-transducers Rho kinase (ROCK) and focal adhesion kinase (FAK) and subsequently regulates the osteogenic phenotype of MG63 osteoblast-like cells. From the results, we found there was no significant difference between the EC and EG matrices with respect to either cell attachment or proliferation rate. However, the gene expression levels of OPN, type I collagen, ALP, and OCN were significantly higher in MG63 osteoblast-like cells grown on the EC than in those grown on the EG. In addition, the phosphorylation levels of Y397-FAK, ERK1/2, BSP, and OPN proteins, as well as ALP activity, were also higher on the EC than on the EG. We further inhibited ROCK activation with Y27632 during differentiation to investigate its effects on matrix-mediated osteogenic differentiation. Results showed the extent of mineralization was decreased with inhibition after induction. Moreover, there is no significant difference between EC and EG. From the results of the protein levels of phosphorylated Y397-FAK, ERK1/2, BSP and OPN, ALP activity and mineral deposition, we speculate that the mechanism that influences the osteogenic differentiation of MG63 osteoblast-like cells on EC and EG is matrix stiffness and via ROCK-FAK-ERK1/2

    Incidence Rates of Enterovirus 71 Infections in Young Children during a Nationwide Epidemic in Taiwan, 2008–09

    Enterovirus 71 (EV71) was first isolated in California, USA, in 1969. Since then, EV71 has been identified globally. Recently, EV71 caused several life-threatening outbreaks in young children in tropical Asia. Development of EV71 vaccines becomes national priority in several Asia countries including Taiwan. To design clinical trials of EV71 vaccines, age-specific incidence rates of EV71 infections are required to identify target populations, estimate disease burdens, select endpoints of clinical efficacy, and estimate sample size. In Taiwan, nationwide EV71 epidemics occurred every 3–4 years but age-specific incidences of EV71 infection are not available. In 2006, we initiated a prospective cohort study in northern Taiwan to recruit neonates and follow up them. In 2008–09, a nationwide EV71 epidemic occurred and we found that age-specific incidence rates of EV71 infection increased from 1.71 per 100 person-years at 0–6 months of age to 4.09, 5.74, and 4.97 per 100 person-years at 7–12, 13–24, and 25–36 months of age, respectively. The cumulative incidence rate was 15% by 36 months of age, and 29% of EV71 infections were asymptomatic in young children. These findings would be helpful to development of EV71 vaccines in Taiwan and other Asian tropical countries
