    High-resolution vessel wall imaging for quantitatively and qualitatively evaluating in-stent stenosis of intracranial aneurysms

    BackgroundIt is critical to accurately and noninvasively evaluate the stented parent artery of intracranial aneurysms (IAs) with endovascular treatment.ObjectiveTo investigate high-resolution vessel wall imaging (HR-VWI) for quantitative and qualitative evaluation of in-stent stenosis (ISS) in IAs treated with stent placement (SP).MethodsFifty-five patients (58 aneurysms) underwent HR-VWI, contrast-enhanced (CE)-HR-VWI, CE-MR angiography (MRA), time-of-flight (TOF)-MRA, and digital subtraction angiography (DSA) six months after SP, and the reliability of quantitative stent lumen measurements was evaluated by intraclass correlation coefficient (ICC) analysis. Agreement and correlation of quantitative evaluation were estimated by comparing the four MR imaging modalities with DSA. The diagnostic performance for >0%, ≥25%, and ≥50% of ISS degrees and overall diagnostic accuracy for the ISS degrees of the four MR imaging modalities were calculated to qualitative evaluation.ResultsThe reliability of CE-HR-VWI and HR-VWI for ISS quantitative measurements was excellent (ICC 0.955–0.989). The agreement and correlation of CE-HR-VWI, HR-VWI versus DSA for ISS quantitative measurements were better than those of CE-MRA and TOF-MRA (p < 0.05). The diagnostic performance for distinguishing the degree of ISS >0%, ≥25%, and ≥50% by CE-HR-VWI and HR-VWI was superior to CE-MRA and TOF-MRA, and their overall diagnostic accuracy was 96.55 and 94.83%, respectively. HR-VWI and CE-HR-VWI were not statistically significant in the quantitative and qualitative evaluation of ISS performance (p > 0.05).ConclusionHR-VWI and CE-HR-VWI have similar performance and value in the quantitative and qualitative evaluation of ISS, and HR-VWI without contrast media could be used as an ideal long-term follow-up approach after SP treatment for IAs

    Two-stage robust planning method for distribution network energy storage based on load forecasting

    A two-stage robust planning method for energy storage in distribution networks based on load prediction is proposed to address the uncertainty of active load in energy storage planning. First, considering the uncertainty of active load, a short-term load forecasting model combining the mutual information method and BiLSTM is established based on k-means++ clustering. Second, based on the results of load forecasting, a comprehensive norm-constrained uncertainty set is constructed, and a two-stage robust model for distribution network energy storage planning is established. The first stage aims to minimize the annual investment cost of the energy storage system, while the second stage aims to minimize the daily operating cost of the distribution network. At the same time, a second-order cone relaxation transformation model with non-convex constraints is introduced to ultimately achieve the optimal economy of the distribution network in energy storage planning. Finally, the effectiveness of the proposed method and model is validated on the IEEE 33-node distribution network model using the MATLAB platform

    Prognosis of HIV Patients Receiving Antiretroviral Therapy According to CD4 Counts: A Long-term Follow-up study in Yunnan, China

    We aim to evaluate the overall survival and associated risk factors for HIV-infected Chinese patients on antiretroviral therapy (ART). 2517 patients receiving ART between 2006 and 2016 were prospectively enrolled in Yunnan province. Kaplan-Meier analyses and Cox proportional hazard regression analyses were performed. 216/2517 patients died during a median 17.5 (interquartile range [IQR] 6.8-33.2) months of follow-up. 82/216 occurred within 6 months of starting ART. Adjusted hazard ratios were10.69 (95%CI 2.38-48.02, p = 0.002) for old age, 1.94 (95%CI 1.40-2.69, p < 0.0001) for advanced WHO stage, and 0.42 (95%CI 0.27-0.63, p < 0.0001) for heterosexual transmission compared to injecting drug users. Surprisingly, adjusted hazard ratios comparing low CD4 counts group (<50 cells/μl) with high CD4 counts group (≥500 cells/μl) within six months after starting ART was 20.17 (95%CI 4.62-87.95, p < 0.0001) and it declined to 3.57 (95%CI 1.10-11.58, p = 0.034) afterwards. Age, WHO stage, transmission route are significantly independent risk factors for ART treated HIV patients. Importantly, baseline CD4 counts is strongly inversely associated with survival in the first six months; whereas it becomes a weak prognostic factor after six months of starting ART

    A correlation analysis of HHV infection and its predictive factors in an HIV-seropositive population in Yunnan, China

    Human herpesviruses (HHVs) have a particularly high prevalence in certain high-risk populations and cause increased morbidity and mortality in patients with acquired immunodeficiency syndrome (AIDS). Screening and treating subclinical HHV infections reduce human immunodeficiency virus (HIV) infection incidence, disease progression, and transmission. However, there are few studies on HHVs, HIV coinfection rates, and their related risk factors. We aimed to clarify the prevalence of all eight HHVs in peripheral blood samples collected from HIV-positive patients, and explore the association of HHV infection in HIV-positive patients in an HIV-seropositive population in Yunnan. We recruited 121 HIV-positive patients with highly active antiretroviral therapy (HAART) and 45 healthy individuals. All the eight HHVs were detected using polymerase chain reaction and their epidemiological information and clinical data were collected and statistically analyzed. A high prevalence of HHVs (89.3%) was observed in individuals with HIV infections and with herpes simplex virus (HSV)-2 (65.3%), and HSV-1 (59.5%) being the most common. Coinfection with more than two different HHVs was more common in patients with HIV infections receiving HAART (72.7%) than in healthy controls. Older age, being married, higher HIV-1 plasma viral loads, and use of antiviral protease inhibitors were independently correlated with an increased frequency of HHVs, but we found no association with CD4 count, WHO HIV clinical stage, and HIV infection duration. Our findings are of great significance for the prevention of HHV opportunistic infection in patients with AIDS and their clinical treatment

    Modeling Rett Syndrome Using TALEN-Edited MECP2 Mutant Cynomolgus Monkeys

    Gene-editing technologies have made it feasible to create nonhuman primate models for human genetic disorders. Here, we report detailed genotypes and phenotypes of TALEN-edited MECP2 mutant cynomolgus monkeys serving as a model for a neurodevelopmental disorder, Rett syndrome (RTT), which is caused by loss-of-function mutations in the human MECP2 gene. Male mutant monkeys were embryonic lethal, reiterating that RTT is a disease of females. Through a battery of behavioral analyses, including primate-unique eye-tracking tests, in combination with brain imaging via MRI, we found a series of physiological, behavioral, and structural abnormalities resembling clinical manifestations of RTT. Moreover, blood transcriptome profiling revealed that mutant monkeys resembled RTT patients in immune gene dysregulation. Taken together, the stark similarity in phenotype and/or endophenotype between monkeys and patients suggested that gene-edited RTT founder monkeys would be of value for disease mechanistic studies as well as development of potential therapeutic interventions for RTT

    Study of Xuanhuang Pill in protecting against alcohol liver disease using ultra-performance liquid chromatography/time-of-flight mass spectrometry and network pharmacology

    IntroductionXuanhuang Pill (XHP) is a traditional Chinese medicine oral formula composed of 10 herbs. This study aims to verify the hepatoprotective activity of XHP and explain its possible mechanism.MethodsThe hepatoprotective activity of XHP was evaluated by constructing a mouse model of alcoholic liver disease, and the mechanism of XHP was preliminarily explained by utilizing ultra-performance liquid chromatography/time-of-flight mass spectrometry (UPLC-QTOF/MS), proteomics and network pharmacology.ResultsThe current study demonstrated that treatment with XHP ameliorated acute alcohol-induced liver injury in mice by significantly reducing alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels and triglycerides (TGs) and malondialdehyde (MDA) content. Remarkably, treatment also increased superoxide dismutase (SOD) activity and glutathione (GSH) content. UPLC-QTOF/MS, 199 compounds were identified as within the make-up of the XHP. Network pharmacology analysis showed that 103 targets regulated by 163 chemical components may play an important role in the protective liver effect mediated by XHP. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis suggest that the HIF-1, FoxO, PI3K-Akt, insulin, and thyroid hormone signaling pathways are key modulators of XHP’s effects. Finally, eight key targets including Mapk1, Mapk3, Akt1, Map2k1, Pik3ca, Pik3cg, Raf1, and Prkca were verified by molecular docking and proteomics analysis, which provide insight into the hepatoprotective effect observed with XHP treatment.ConclusionIn summary, these results improved upon knowledge of the chemical composition and the potential mechanisms of hepatoprotective action of oral XHP treatment, providing foundational support for this formulation as a viable therapeutic option for alcoholic liver disease

    P_VggNet: A convolutional neural network (CNN) with pixel-based attention map.

    Attention maps have been fused in the VggNet structure (EAC-Net) [1] and have shown significant improvement compared to that of the VggNet structure. However, in [1], E-Net was designed based on the facial action unit (AU) center and for facial AU detection only. Thus, for the use of attention maps in every image type, this paper proposed a new convolutional neural network (CNN) structure, P_VggNet, comprising the following parts: P_Net and VggNet with 16 layers (VggNet-16). The generation approach of P_Net was designed, and the P_VggNet structure was proposed. To prove the efficiency of P_VggNet, we designed two experiments, which indicated that P_VggNet could more efficiently extract image features than VggNet-16

    Hydro-Geochemistry of the River Water in the Jiulongjiang River Basin, Southeast China: Implications of Anthropogenic Inputs and Chemical Weathering

    This study focuses on the chemical weathering process under the influence of human activities in the Jiulongjiang River basin, which is the most developed and heavily polluted area in southeast China. The average total dissolved solid (TDS) of the river water is 116.6 mg/L and total cation concentration ( TZ + ) is 1.5 meq/L. Calcium and HCO 3 &#8722; followed by Na + and SO 4 2 &#8722; constitute the main species in river waters. A mass balance based on cations calculation indicated that the silicate weathering (43.3%), carbonate weathering (30.7%), atmospheric (15.6%) and anthropogenic inputs (10.4%) are four reservoirs contributing to the dissolved load. Silicates (SCW) and carbonates (CCW) chemical weathering rates are calculated to be approximately 53.2 ton/km2/a and 15.0 ton/km2/a, respectively. When sulfuric and nitric acid from rainfall affected by human activities are involved in the weathering process, the actual atmospheric CO 2 consumption rates are estimated at 3.7 &#215; 105 mol/km2/a for silicate weathering and 2.2 &#215; 105 mol/km2/a for carbonate weathering. An overestimated carbon sink (17.4 Gg C / a ) is about 27.0% of the CO 2 consumption flux via silicate weathering in the Jiulongjiang River basin, this result shows the strong effects of anthropogenic factors on atmospheric CO 2 level and current and future climate change of earth

    Distribution, Sources, and Water Quality Assessment of Dissolved Heavy Metals in the Jiulongjiang River Water, Southeast China

    In this study, the concentration of eight dissolved heavy metals (Ti, Cr, Mn, Fe, Ni, Mo, Sb, and Ba) in 42 water samples from the Jiulongjiang River, southeast China, were determined by inductively coupled plasma mass spectrometry (ICP-MS). Multivariate statistical methods, including correlation analysis (CA) and factor and principal component analysis (FA/PCA), were analyzed to identify the sources of the elements. Water quality index (WQI) and health risk assessment, including hazard quotient (HQ) and hazard index (HI), were used to evaluate water quality and the impacts on human health. Our results were compared with the drinking water guidelines reported by China, the World Health Organization (WHO), and the United States Environmental Protection Agency (US EPA), revealing that Ti, Mn, and Sb were not within approved limits at some sites and might be the main pollutants in the drainage basin. Based on the spatial distributions, Ti, Mn, Fe, Ni, and Mo showed good similarity, indicating that they might come from similar sources along the river. The CA results also showed that Ti, Mn, Fe, Ni, and Mo had a high correlation coefficient. The FA/PCA results identified three principal components (PC) that accounted for 79.46% of the total variance. PC 1 suggested that a mixed lithogenic and urban land source contributed to Ti, Mn, Fe, Ni, and Mo; PC 2 showed that Cr, Ni, and Mo were influenced by the discharge of industrial effluents; Sb had a strong loading on PC 3, which was controlled by mining activities. The results of the WQI indicated that the water in the Jiulongjiang River was basically categorized as excellent water, but the water quality levels in site W5 and N4 were poorer due to urban land use. Hazard quotient and HI values showed that Sb was a potential threat to human health, indicating that preventive actions should be considered in regard to mining activities in the upper reaches of Beixi stream
