    Voltammetry under a Controlled Temperature Gradient

    Electrochemical measurements are generally done under isothermal conditions. Here we report on the application of a controlled temperature gradient between the working electrode surface and the solution. Using electrochemical sensors prepared on ceramic materials with extremely high specific heat conductivity, the temperature gradient between the electrode and solution was applied here as a second driving force. This application of the Soret phenomenon increases the mass transfer in the Nernst layer and enables more accurate control of the electrode response enhancement by a combination of diffusion and thermal diffusion. We have thus studied the effect of Soret phenomenon by cyclic voltammetry measurements in ferro/ferricyanide. The time dependence of sensor response disappears when applying the Soret phenomenon, and the complicated shape of the cyclic voltammogram is replaced by a simple exponential curve.     Heart transplantation for cardiac light chain amyloidosis with subsequent autologous stem cell transplantation

    AbstractOur report describes a case of 57-year-old man with manifest heart failure on the basis of cardiac amyloidosis, which was detected by endomyocardial biopsy. Due to the heart failure, the patient was unable to undergo myeloablative therapy. We changed our previous decision for conservative therapy of heart failure and the patient underwent heart transplantation. Autologous stem cell transplantation was performed 6 months later. After the successful stem cell transplantation, the serum free light chain lambda levels promptly decreased. One year later, their levels started again to increase. Chemotherapy was therefore initiated. The patient has now completed the seventh cycle of chemotherapy in good condition. The graft function is normal and the latest endomyocardial biopsy revealed no amyloid

    Brownfields do not "only live twice": The possibilities for heritage preservation and the enlargement of leisure time activities in Brno, the Czech Republic

    Central Europe is replete with legacy contaminated sites, commonly called “brownfields”. The question is what can be done to remediate them and make them again safe and useful to society. This question is addressed in post-socialistic city of Brno, the Czech Republic. Our research assesses public perceptions of such sites that are currently utilized for the leisure time activities. Special attention is paid to public views of heritage preservation as an option for brownfield regeneration. The principal aim of the paper is to measure public support of heritage preservation through the conversion of brownfields to leisure activities and tourism. Data were gathered by means of the questionnaire survey with visitors of four brownfields currently being used for leisure time activities (n = 130). It was revealed that the awareness of the visitors of the issue of brownfield regeneration is very low, and that importantly affects the perception of heritage preservation of individual sites. Further, the opinions of the respondents are significantly influenced by the local contexts of individual brownfields (predominantly by the situation and the technical state). However, it might be stated that in locations where the historical state of building has already been repaired, historical heritage is perceived more positively

    Alterations in the balance between skeletogenesis and adipogenesis is a pathogenic feature in multiple skeletal disorders. Clinically, enhanced bone marrow adiposity in bones impairs mobility and increases fracture risk, reducing the quality of life of patients. The molecular mechanism that underlies the balance between skeletogenesis and adipogenesis is not completely understood but alterations in skeletal progenitor cells’ differentiation pathway plays a key role. We recently demonstrated that parathyroid hormone (PTH)/PTH-related peptide (PTHrP) control the levels of DEPTOR, an inhibitor of the mechanistic target of rapamycin (mTOR), and that DEPTOR levels are altered in different skeletal diseases. Here, we show that mutations in the PTH receptor-1 (PTH1R) alter the differentiation of skeletal progenitors in two different skeletal genetic disorders and lead to accumulation of fat or cartilage in bones. Mechanistically, DEPTOR controls the subcellular localization of TAZ (transcriptional co-activator with a PDZ-binding domain), a transcriptional regulator that governs skeletal stem cells differentiation into either bone and fat. We show that DEPTOR regulation of TAZ localization is achieved through the control of Dishevelled2 (DVL2) phosphorylation. Depending on nutrient availability, DEPTOR directly interacts with PTH1R to regulate PTH/PTHrP signaling or it forms a complex with TAZ, to prevent its translocation to the nucleus and therefore inhibit its transcriptional activity. Our data point DEPTOR as a key molecule in skeletal progenitor differentiation; its dysregulation under pathologic conditions results in aberrant bone/fat balance

    Effect of fluid resuscitation on mortality and organ function in experimental sepsis models

    Introduction Several recent studies have shown that a positive fluid balance in critical illness is associated with worse outcome. We tested the effects of moderate vs. high-volume resuscitation strategies on mortality, systemic and regional blood flows, mitochondrial respiration, and organ function in two experimental sepsis models. Methods 48 pigs were randomized to continuous endotoxin infusion, fecal peritonitis, and a control group (n = 16 each), and each group further to two different basal rates of volume supply for 24 hours [moderate-volume (10 ml/kg/h, Ringer's lactate, n = 8); high-volume (15 + 5 ml/kg/h, Ringer's lactate and hydroxyethyl starch (HES), n = 8)], both supplemented by additional volume boli, as guided by urinary output, filling pressures, and responses in stroke volume. Systemic and regional hemodynamics were measured and tissue specimens taken for mitochondrial function assessment and histological analysis. Results Mortality in high-volume groups was 87% (peritonitis), 75% (endotoxemia), and 13% (controls). In moderate-volume groups mortality was 50% (peritonitis), 13% (endotoxemia) and 0% (controls). Both septic groups became hyperdynamic. While neither sepsis nor volume resuscitation strategy was associated with altered hepatic or muscle mitochondrial complex I- and II-dependent respiration, non-survivors had lower hepatic complex II-dependent respiratory control ratios (2.6 +/- 0.7, vs. 3.3 +/- 0.9 in survivors; P = 0.01). Histology revealed moderate damage in all organs, colloid plaques in lung tissue of high-volume groups, and severe kidney damage in endotoxin high-volume animals. Conclusions High-volume resuscitation including HES in experimental peritonitis and endotoxemia increased mortality despite better initial hemodynamic stability. This suggests that the strategy of early fluid management influences outcome in sepsis. The high mortality was not associated with reduced mitochondrial complex I- or II-dependent muscle and hepatic respiration

    A multi-purpose, multi-rotor drone system for long-range and high-altitude volcanic gas plume measurements

    A multi-rotor drone has been adapted for studies of volcanic gas plumes. This adaptation includes improved capacity for high-altitude and long-range, real-time SO2 concentration monitoring, long-range manual control, remotely activated bag sampling and plume speed measurement capability. The drone is capable of acting as a stable platform for various instrument configurations, including multi-component gas analysis system (MultiGAS) instruments for in situ measurements of SO2, H2S, and CO2 concentrations in the gas plume and portable differential optical absorption spectrometer (MobileDOAS) instruments for spectroscopic measurement of total SO2 emission rate, remotely controlled gas sampling in bags and sampling with gas denuders for posterior analysis on the ground of isotopic composition and halogens. The platform we present was field-tested during three campaigns in Papua New Guinea: in 2016 at Tavurvur, Bagana and Ulawun volcanoes, in 2018 at Tavurvur and Langila volcanoes and in 2019 at Tavurvur and Manam volcanoes, as well as in Mt. Etna in Italy in 2017. This paper describes the drone platform and the multiple payloads, the various measurement strategies and an algorithm to correct for different response times of MultiGAS sensors. Specifically, we emphasize the need for an adaptive flight path, together with live data transmission of a plume tracer (such as SO2 concentration) to the ground station, to ensure optimal plume interception when operating beyond the visual line of sight. We present results from a comprehensive plume characterization obtained during a field deployment at Manam volcano in May 2019. The Papua New Guinea region, and particularly Manam volcano, has not been extensively studied for volcanic gases due to its remote location, inaccessible summit region and high level of volcanic activity. We demonstrate that the combination of a multi-rotor drone with modular payloads is a versatile solution to obtain the flux and composition of volcanic plumes, even for the case of a highly active volcano with a high-altitude plume such as Manam.     Quality control and quantification in IG/TR next-generation sequencing marker identification: protocols and bioinformatic functionalities by EuroClonality-NGS

    Assessment of clonality, marker identification and measurement of minimal residual disease (MRD) of immunoglobulin (IG) and T cell receptor (TR) gene rearrangements in lymphoid neoplasms using next-generation sequencing (NGS) is currently under intensive development for use in clinical diagnostics. So far, however, there is a lack of suitable quality control (QC) options with regard to standardisation and quality metrics to ensure robust clinical application of such approaches. The EuroClonality-NGS Working Group has therefore established two types of QCs to accompany the NGS-based IG/TR assays. First, a central polytarget QC (cPT-QC) is used to monitor the primer performance of each of the EuroClonality multiplex NGS assays; second, a standardised human cell line-based DNA control is spiked into each patient DNA sample to work as a central in-tube QC and calibrator for MRD quantification (cIT-QC).     Surgical liver resection in multimodal therapy of hepatic malignat affections: A 4-year study

    BACKGROUND: The goal of our study was to evaluate our hitherto 4-year surgical treatment of liver malignancies. METHODS: Our team performed 43 liver resections from 1997 to 2000. Of these, 10 were primary tumours and 33 metastases. The most frequent indication for resection in our sample was for colorectal cancer - in total 27 patients. Metastatic pancreatic cancer or metastases of malignant melanoma account for rarer indications to surgery We considered CT arterioportography (CTAP) as the most significant preoperative examination from all available radiological methods. RESULTS Proper resection of the liver is possible even without special technical equipment. No patient between the ages of 22 - 82 years died in the 30-day postoperative period. Our postoperative morbidity was 18.6%. The most frequent complication was prolonged biliary secretion from drains. In our study, recurrent liver malignancies occurred most frequently within 6 months from the primary resection. CONCLUSION: Radical resection procedure may be facilitated for the greatest number of patients with primary or metastatic liver malignancies by the cohesive cooperation of a number of specialists
