    Progress of the acyl-Coenzyme A thioester hydrolase family in cancer

    In recent years, the acyl-Coenzyme A thioester hydrolase family (ACOTs) has received wide attention as a key link in lipid metabolism. This family is a class of enzymes that catalyze the hydrolysis of fatty acyl-Coenzyme A, disrupting the thioester bond present within acyl-CoA ester molecules to produce free fatty acids (FFA) and the corresponding coenzyme A (CoA). Such enzymes play a very important role in lipid metabolism through maintaining appropriate levels of intracellular FFA and fatty acyl-CoA as well as CoA. It is broadly divided into two distinct subgroups, the type-I α/β-hydrolase fold enzyme superfamily and the type-II ‘hot dog’ fold superfamily. There are currently four human type-I genes and eight human type-II genes. Although the two subgroups catalyze the same reaction, they are not structurally similar, do not share the same sequence homology, and differ greatly in protein executive functions. This review summarizes the classification of the acyl-CoA thioester hydrolase family, an overview of the structural sequences, and advances in digestive, respiratory, and urinary systemic tumors.     Effects of Tea Residue Extracts with Different Molecular Weight on the Pasting Characteristics of Potato Starch

    Tea residues are the remaining residue of tea after processing and utilization, which are rich in multiple active components. To investigate the effects of different types and molecular weights of tea residue extracts on the pasting characteristics of potato starch (PS), the ethanol extract (TRE), water extract (TRW) and alkali extract (TRA) of tea residue were obtained by continuous extraction method. On this basis, the different molecular weights of ethanol extract (TRE-1, 30 kDa) and water extract (TRW-1, 100 kDa) were prepared by a membrane separation. The effects of different tea residue extracts on the viscosity properties were investigated, and the microstructure of potato starch added with tea residue extract was observed by scanning electron microscopy (SEM). The results showed that different types and molecular weights of tea residue extracts could significantly (PTRW-2>TRE-2>TRW-1>TRE-1. The peak viscosity of potato starch was gradually decreased with the increase of different extracts. After adding 10% TRA, TRW-2, TRE-2, TRW-1 and TRE-1, the peak viscosity of potato starch was 4624, 5013, 5431, 5911 and 6195 cP, respectively. TRE-2, TRW-2 and TRA could better promote the link between potato starch fragments and result in a more complete and smooth lamellar structure, compared with TRE-1, TRW-1. In summary, the addition of different types and molecular weights of tea residue extracts could effectively inhibit the gelatinization of potato starch, and the inhibitory effect of 10% alkali extract was the best

    Advances in circulating tumor cells for early detection, prognosis and metastasis reduction in lung cancer

    Globally, lung cancer stands as the leading type of cancer in terms of incidence and is the major source of mortality attributed to cancer. We have outlined the molecular biomarkers for lung cancer that are available clinically. Circulating tumor cells (CTCs) spread from the original location, circulate in the bloodstream, extravasate, and metastasize, forming secondary tumors by invading and establishing a favorable environment. CTC analysis is considered a common liquid biopsy method for lung cancer. We have enumerated both in vivo and ex vivo techniques for CTC separation and enrichment, examined the advantages and limitations of these methods, and also discussed the detection of CTCs in other bodily fluids. We have evaluated the value of CTCs, as well as CTCs in conjunction with other biomarkers, for their utility in the early detection and prognostic assessment of patients with lung cancer. CTCs engage with diverse cells of the metastatic process, interfering with the interaction between CTCs and various cells in metastasis, potentially halting metastasis and enhancing patient prognosis

    Influencing factors of medication literacy among community-dwelling older adult patients with hypertension: a study based on social learning theory

    Objective: This study aimed to examine the factors affecting medication literacy in community-dwelling older adults with hypertension, guided by social learning theory. It sought to analyze the pathways these factors influenced and provide a theoretical foundation for designing targeted intervention programs.Study design: This is a cross-sectional study.Methods: From October 2022 to February 2023, a total of 432 community-dwelling older adults with hypertension from Linghe District, Guta District, and Taihe District, Jinzhou City, Liaoning Province, China, were selected using convenience sampling. Data were collected using a socio-demographic questionnaire, a medication literacy questionnaire, the Brief Illness Perception Questionnaire, the General Self-efficacy Scale, and the Perceived Social Support Scale. The collected data were analyzed with Kruskal-Wallis and Mann-Whitney tests, correlation analysis, multiple stepwise regression analysis, and structural equation modeling (SEM).Results: The medication literacy score among the participants was 3.83 ± 1.91. Multi-factor analysis revealed key factors affecting their medication literacy, including blood pressure-control status, utilization of community health education resources, receiving guidance for medication usage, marital status, number of annual visits, social support, self-efficacy, and disease perception. The SEM based on social learning theory showed that general self-efficacy mediated the relationship between social support, disease perception, and medication literacy.Conclusion: The present study developed a model and provided potential intervention strategies to improve medication literacy, knowledge, and safety among community-dwelling older adults with hypertension, considering the relationships between the identified variables

    Diel vertical migration of Peridiniopsis niei, Liu et al., a new species of dinoflagellates in an eutrophic bay of Three-Gorge Reservoir, China

    In spring, a typical dinoflagellate (Peridiniopsis niei Liu et al.) constitutes most of the phytoplankton biomass in most of eutrophic bay of Three-Gorge Reservoir (TGR, China). There are few field observations on vertical migration of the members of the genus Peridiniopsis. We studied diel vertical migration of the new species (Peridiniopsis niei) at sampling stations A and B in an eutrophic bay (Xiangxi Bay) of TGR during both day and night in March 2007. The present study suggests that vertical migration by Peridiniopsis niei is similar in nature to the migration patterns observed for other dinoflagellates. Solar irradiance incident was an important factor regulating the ascent and descent of Peridiniopsis niei. The vertical descent of Peridiniopsis niei during nighttime was restricted when the thermocline (Temperature gradient > 1A degrees C m(-1)) was present in the station B

    A metabolomics-based analysis of the metabolic pathways associated with the regulation of branched-chain amino acids in rats fed a high-fructose diet

    Previous studies have shown that the elevated levels of circulating branched-chain amino acids (BCAAs) are associated with the development of insulin resistance and its complications, including obesity, type 2 diabetes, cardiovascular disease and some cancers. However, animal models that can mimic the metabolic state of chronically elevated BCAAs in humans are rare. Therefore, the aim of this study was to establish the above animal model and analyse the metabolic changes associated with high BCAA levels. Sixteen 8-week-old Sprague–Dawley (SD) rats were randomly divided into two groups and given either a high fructose diet or a normal diet. BCAA levels as well as blood glucose and lipid levels were measured at different time points of feeding. The mRNA expression levels of two key enzymes of BCAA catabolism, ACAD (acyl-CoA de hydrogenase) and BCKDH (branched-chain α-keto acid dehydrogenase), were measured by qPCR, and the protein expression levels of these two enzymes were analysed by immunoh istochemistry. Finally, the metabolite expression differences between the two groups were analysed by Q300 metabolomics technology. Our study confirms that defects in the catabolic pathways of BCAAs lead to increased levels of circulating BCAAs, resulting in disorders of glucose and lipid metabolism characterized by insulin resistance by affecting metabolic pathways associated with amino acids and bile acids

    Metabolic perturbation of Streptomyces albulus by introducing NADP-dependent glyceraldehyde 3-phosphate dehydrogenase

    The available resources of Streptomyces represent a valuable repository of bioactive natural products that warrant exploration. Streptomyces albulus is primarily utilized in the industrial synthesis of ε-poly-L-lysine (ε-PL). In this study, the NADP-dependent glyceraldehyde 3-phosphate dehydrogenase (GapN) from Streptococcus mutans was heterologously expressed in S. albulus CICC11022, leading to elevated intracellular NADPH levels and reduced NADH and ATP concentrations. The resulting perturbation of S. albulus metabolism was comprehensively analyzed using transcriptomic and metabolomic methodologies. A decrease in production of ε-PL was observed. The expression of gapN significantly impacted on 23 gene clusters responsible for the biosynthesis of secondary metabolites. A comprehensive analysis revealed a total of 21 metabolites exhibiting elevated levels both intracellularly and extracellularly in the gapN expressing strain compared to those in the control strain. These findings underscore the potential of S. albulus to generate diverse bioactive natural products, thus offering valuable insights for the utilization of known Streptomyces resources through genetic manipulation

    Does the Upgrading of Provincial Development Zones Improve the Cities’ Economic Efficiency? Evidence from a Quasi-experiment of Heterogeneous Development Zones

     The expansion of China's development zones has already made significant contributions to economic development, as well as provided practical guidance for other developing countries to implement development zone practice. This paper analyzes how industry characteristics, high-technology zone hierarchy differences, regional differences, and openness differences affect the causal relationship between high-tech zones (HTZ) establishment and innovation of information service enterprises. The study constructs a complete empirical analysis framework with the help of the difference-indifference model (DID) and interaction term model. The empirical results show that, compared with the control group, HTZ establishes more innovation intentions and increases the innovation scale, thus significantly enhancing innovation levels. Compared with the non-high-tech zone sample, the HTZ has a significant positive effect on the innovation ability of technology service enterprises in the cities. Meanwhile, through a comparison of the regression results of different regions, it is found that the establishment of high-tech zones has the best effect on enterprise innovation ability in the eastern region, followed by the northeastern, central, and western regions. The following policy recommendations are proposed for optimizing the construction and management of high technology zones and promoting enterprise innovation development

    With the continuous progress of computer and laser measurement technology, non-contact measurement based on laser scanning technology has been more and more applied in the industrial production process. Since the 1990s, 3D laser scanning technology used as a new technology has been developed rapidly. The 3D laser scanning technology, also known as High Definition Surveying (HDS), was based on the principle of laser ranging. The technology on the surface of a measured object can record some features of large dense points, including 3d coordinates, reflectance and texture information, so as to rapidly reconstruct 3D models of a measured target and various map data such as the line, the surface and the body. Meanwhile, it is a revolutionary leap that the technology improves the traditional single point measurement to the surface measurement, and realizes the digital reconstruction of the objects. Especially, for some conditions such as contact measurement and complex component detection that cannot be measured, 3D laser scanning technology has a natural advantage in the field of industrial design and measurement
