    A developmental study of drawing expression in young children

    The present study examined the development of drawing expression in young children, especially the transition between intellectual realism and visual realism, by comparing what they drew with imagination and what they drew while looking at a model. Forty-five children (three age groups: 3-4, 4-5, and 5-6 year olds; n = 15, 16, 14, respectively) participated in this study. First, they were instructed to draw a lion without looking at anything (the imagination task). Next, they drew a lion while looking at a photo of a lion (the imitation task). We checked on each drawing for the presence of lion’s component parts (eyes, mouth, nose, ears, mane, head, body, four legs, tail, and color). In addition, each drawing was evaluated whether it seemed to lion. The results revealed the following: 1) Young children were able to draw more lion’s parts on the imitation task than the imagination task. 2) Four-five year-olds and 5-6 year-olds were able to draw more parts than 3-4 year-olds. 3) The transition from intellectual realism to visual realism occurred approximately between five and six years of age. 4) It was shown by evaluation that older children’s drawing had captured the more distinctive features of lion than younger children’s one

    Lactoferrin-like Immunoreactivity in Distinct Neuronal Populations in the Mouse Central Nervous System

    Lactoferrin (Lf) is an iron-binding glycoprotein mainly found in exocrine secretions and the secondary granules of neutrophils. In the central nervous system (CNS), expression of the Lf protein has been reported in the lesions of some neurodegenerative disorders such as Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis, as well as in the aged brain. Lf is primarily considered an iron chelator, protecting cells from potentially toxic iron or iron-requiring microorganisms. Other biological functions of Lf include immunomodulation and transcriptional regulation. However, the roles of Lf in the CNS have yet to be fully clarified. In this study, we raised an antiserum against mouse Lf and investigated the immunohistochemical localization of Lf-like immunoreactivity (Lf-LI) throughout the CNS of adult mice. Lf-LI was found in some neuronal populations throughout the CNS. Intense labeling was found in neurons in the olfactory systems, hypothalamic nuclei, entorhinal cortex, and a variety of brainstem nuclei. This study provides detailed information on the Lf-LI distribution in the CNS, and the findings should promote further understanding of both the physiological and pathological significance of Lf in the CNS

    Association study of the vesicular monoamine transporter 1 (VMAT1) gene with schizophrenia in a Japanese population

    BACKGROUND: Vesicular monoamine transporters (VMATs) mediate accumulation of monoamines such as serotonin, dopamine, adrenaline, and noradrenaline from the cytoplasm into storage organelles. The VMAT1 (alternatively solute carrier family 18: SLC18A1) regulates such biogenic amines in neuroendocrine systems. The VMAT1 gene maps to chromosome 8p21.3, a locus with strong evidence of linkage with schizophrenia. A recent study reported that a non-synonymous single nucleotide polymorphism (SNP) of the gene (Pro4Thr) was associated with schizophrenia. METHODS: We attempted to replicate this finding in a Japanese sample of 354 schizophrenics and 365 controls. In addition, we examined 3 other non-synonymous SNPs (Thr98Ser, Thr136Ile, and Val392Leu). Genotyping was performed by the TaqMan allelic discrimination assay. RESULTS: There was no significant difference in genotype or allele distribution of the three SNPs of Pro4Thr, Thr136Ile, or Val392Leu between patients and controls. There was, however, a significant difference in genotype and allele distributions for the Thr98Ser polymorphism between the two groups (P = 0.01 for genotype and allele). When sexes were examined separately, significant differences were observed in females (P = 0.006 for genotype, P = 0.003 for allele), but not in males. The Thr98 allele was more common in female patients than in female controls (odds ratio 1.69, 95% CI 1.19–2.40, P = 0.003). Haplotype-based analyses also provided evidence for a significant association in females. CONCLUSION: We failed to replicate the previously reported association of Pro4Thr of the VMAT1 gene with schizophrenia. However, we obtained evidence for a possible role of the Thr98Ser in giving susceptibility to schizophrenia in women

    There have been suicides at the Hansen’s disease sanatoria in the past. What is the level of suffering that makes a person think of suicide? Through the analysis of a Hansen’s disease survivor’s life, we clarified that the nature of sufferings is spiritual pain that is brought by the “loss of autonomy and independence for carving out my life.” The patient was stuck in a negative cycle for about 20 years: All my dreams and hopes cut off → Despair → I want to die but cannot die easily → Lethargy∙Desperation∙Nihilistic → Momentary pleasure by gambling → I do not feel like living → I ask myself why am I alive → Self-criticism → Repeat again. Recovery from spiritual pain was achieved with time and by reflection. The bellwethers led to a turning point that helped to change the flow of his life. He also started using time effectively. Spiritual well-being of aging survivors has a trilaminar structure, comprising the following: (1) establishing the existential foundation of a sustained self; (2) living in the moment while contemplating the future; and (3) dovetailing within the society and transcending space and time. It indicates that they can achieve the developmental tasks related to old age, personal integration containing spirituality, and the wisdom and strength to get over difficulties.     Lateral current injection GaInAsP/InP laser on semi-insulating substrate for membrane-based photonic circuits

    金沢大学理工研究域電子情報学系A room-temperature pulsed operation was demonstrated using lateral current injection-type lasers composed of a 400-nm-thick GaInAsP core layer with compressively strained 5 quantum wells. A threshold current of 105 mA and corresponding density of 1.3 kA/cm2 (260 A/cm2 per well) were obtained with the stripe width of 5.4 μm and the cavity length of 1.47 mm. A fundamental transverse mode operation was obtained with the narrower stripe device of 2.0 μm and the cavity length of 805 μm, while the threshold current and corresponding density were 49 mA and 3.0 kA/cm 2, respectively. © 2009 Optical Society of America

    Pharmacologic characterization of TBP1901, a prodrug form of aglycone curcumin, and CRISPR-Cas9 screen for therapeutic targets of aglycone curcumin

    プロドラッグ型クルクミン注射製剤の抗腫瘍効果及び治療標的の包括的な解析 --安全性の高い抗がん薬としての開発に期待--. 京都大学プレスリリース. 2022-10-21.Curcumin (aglycone curcumin) has antitumor properties in a variety of malignancies via the alteration of multiple cancer-related biological pathways; however, its clinical application has been hampered due to its poor bioavailability. To overcome this limitation, we have developed a synthesized curcumin β-D-glucuronide sodium salt (TBP1901), a prodrug form of aglycone curcumin. In this study, we aimed to clarify the pharmacologic characteristics of TBP1901. In β-glucuronidase (GUSB)-proficient mice, both curcumin β-D-glucuronide and its active metabolite, aglycone curcumin, were detected in the blood after TBP1901 injection, whereas only curcumin β-D-glucuronide was detected in GUSB-impaired mice, suggesting that GUSB plays a pivotal role in the conversion of TBP1901 into aglycone curcumin in vivo. TBP1901 itself had minimal antitumor effects in vitro, whereas it demonstrated significant antitumor effects in vivo. Genome-wide clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 screen disclosed the genes associated with NF-κB signaling pathway and mitochondria were among the highest hit. In vitro, aglycone curcumin inhibited NF-kappa B signaling pathways whereas it caused production of reactive oxygen species (ROS). ROS scavenger, N-acetyl-L-cysteine, partially reversed antitumor effects of aglycone curcumin.
