    Ph-like ALL is characterized by several chromosomal translocations involving activating cytokine receptor or tyrosine kinase such as CRLF2, ABL1, JAK2, and PDGFRB (Robert K.G et al, 2014). Recent increasing evidences suggest that patients with Ph-like ALL bearing PDGFRB translocation are potentiated to respond to tyrosine kinase inhibitors. Thus, this translocation should be included within the molecular companion diagnostics to facilitate tailor-made cancer therapy

    Pathological study of chronic pulmonary toxicity induced by intratracheally instilled Asian sand dust (Kosa): possible association of fibrosis with the development of granulomatous lesions

    Introduction. Exposure to Asian sand dust (ASD) is associated with enhanced pulmonary morbidity and mor­tality, and the reporting of such cases has rapidly increased in East Asia since 2000. The purpose of the study was to assess chronic lung toxicity induced by ASD. Material and methods. A total of 174 ICR mice were randomly divided into 5 control and 17 exposure groups. Suspensions of low dose (0.2, 0.4 mg) and high dose (3.0 mg) of ASD particles in saline were intratracheally instilled into ICR mice, followed by sacrifice at 24 hours, 1 week, and 1, 2, 3 and 4 months after instillation. Paraffin sections of lung tissues were stained with hematoxylin and eosin and by immunohistochemistry to detect α-smooth muscle actin, collagen III, matrix metalloproteinase-9 (MMP-9), tissue inhibitor of metalloproteinases-1 (TIMP-1), CD3, CD20, immunoglobulin G, interleukin-1β and inducible nitric oxide synthase. Results. A lung histological examination revealed similar patterns in the lesions of the groups treated with high (3.0 mg) or low dose (0.4 mg) of ASD. Acute inflammation was observed 24 h after treatment and subsided after 1 week; persistent granulomatous changes were observed at 2 months, focal lymphocytic infiltration at 3 months, and granuloma formation at 4 months. An increase in the size of granulomatous lesions was observed over time and was accompanied by collagen deposition in the lesions. The cytoplasm of macrophages in inflammatory lesions showed positive immunolabeling for MMP-9 at 24 h, 1 and 2 months after instillation of 3.0 mg of ASD. Positive immunolabeling for TIMP-1 was demonstrated in the cytoplasm of macrophages at 2 and 4 months after instillation of 3.0 mg of ASD. These findings suggest association between the expression of MMP-9 and TIMP-1 with the development of lung granulomatous lesions. Conclusions. These findings suggest that collagen deposition resulting from the altered regulation of extracel­lular matrix is associated with granuloma formation in the lungs of mice treated with ASD

    A construction of 3-dimensional lattice sieve for number field sieve over F_{p^n}

    The security of pairing-based cryptography is based on the hardness of solving the discrete logarithm problem (DLP) over extension field F_{p^n} of characteristic p and degree n. Joux et al. proposed an asymptotically fastest algorithm for solving DLP over F_{p^n} (JLSV06-NFS) as the extension of the number field sieve over prime field F _p (JL03-NFS). The lattice sieve is often used for a large-scaled experiment of solving DLP over F_p by the number field sieve. Franke and Kleinjung proposed a 2-dimensional lattice sieve which efficiently enumerates all the points in a given sieve region of the lattice. However, we have to consider a sieve region of more than 2 dimensions in the lattice sieve of JLSV06-NFS. In this paper, we extend the Franke-Kleinjung method to 3-dimensional sieve region. We construct an appropriate basis using the Hermite normal form, which can enumerate the points in a given sieve region of the 3-dimensional lattice. From our experiment on F_{p^{12}} of 303 bits, we are able to enumerate more than 90\% of the points in a sieve region in the lattice generated by special-q. Moreover, we implement the number field sieve using the proposed 3-dimensional lattice sieve. Our implementation of the JLSV06 over F_{p^6} of 240 bits is about as efficient as that of the current record over F_{p^6} using 3-dimensional line sieve by Zajac

However, using hiPSC-derived NPCs clinically is a challenge because hiPSCs themselves are tumorigenic. LIN28A, ESRG, CNMD and SFRP2 transcripts have been used as a marker of residual hiPSCs for a variety of cell types undergoing clinical trials. In this study, by reanalyzing public databases, we found a baseline expression of LIN28A, ESRG, CNMD and SFRP2 in hiPSC-derived NPCs and several other cell types, suggesting LIN28A, ESRG, CNMD and SFRP2 are not always reliable markers for iPSC detection. As an alternative, we discovered a lncRNA marker gene, MIR302CHG, among many known and unknown iPSC markers, as highly differentially expressed between hiPSCs and NPCs, by RNA sequencing and quantitative RT-PCR (qRT-PCR) analyses. Using MIR302CHG as an hiPSC marker, we constructed two assay methods, a combination of magnetic bead-based enrichment and qRT-PCR and digital droplet PCR alone, to detect a small number of residual hiPSCs in NPC populations. The use of these in vitro assays could contribute to patient safety in treatments using hiPSC-derived cells

    Experimental IgA Nephropathy Induced by Oral Administration of Dextran

    Attempts were made to produce experimental IgA nephropathy by oral administration for 18 weeks of dextran having a molecular weight of 10,000, 70,000, and 500,000 to 6-week old female BALB/c mice. It was observed that in three out of eight cases belonging to the group given dextran having a molecular weight of 70,000 and in three out of seven cases belonging to the group given dextran having a molecular weight of 500,000, deposition of IgA on the mesangium was demonstrated by immunofluorescence direct method. Furthermore, deposition of IgG and IgM was observed in not only the groups given dextran but also in the control group, but no significant difference in deposition pattern could be demonstrated between the groups. C3 was observed only in one case belonging to the group given dextran having a molecular weight of 10,000. The serum IgA value showed a higher value in the cases with IgA deposition than in the control group. Serum C3 value was higher in the IgA deposition cases in the group given dextran having a molecular weight of 500,000 than in the control group. These results suggest that it is possible to produce experimental IgA nephropathy by continual oral administration of carbohydrate antigen of bacterial origin and that bacterial infection is involved in the development of IgA nephropathy

    New Solutions to the Firing Squad Synchronization Problems for Neural and Hyperdag P Systems

    We propose two uniform solutions to an open question: the Firing Squad Synchronization Problem (FSSP), for hyperdag and symmetric neural P systems, with anonymous cells. Our solutions take e_c+5 and 6e_c+7 steps, respectively, where e_c is the eccentricity of the commander cell of the dag or digraph underlying these P systems. The first and fast solution is based on a novel proposal, which dynamically extends P systems with mobile channels. The second solution is substantially longer, but is solely based on classical rules and static channels. In contrast to the previous solutions, which work for tree-based P systems, our solutions synchronize to any subset of the underlying digraph; and do not require membrane polarizations or conditional rules, but require states, as typically used in hyperdag and neural P systems

    Quantification of Cell Migration and Invasion, and Their Association with Periostin in Anaplastic Thyroid Cancer, Using a Real-time Cell Analyzer

    Anaplastic thyroid cancer (ATC) is known to be a highly malignant cancer of the thyroid with a high mortality rate. In a previous study, we used real-time cell analysis (RTCA) to analyze cell migration and invasion of oral squamous cell carcinomas (OSCCs) of the tongue and floor of the mouth. In the present study, we investigated cell migration and invasion of ATC using RTCA, as well as their association with periostin, matrix metalloproteinases (MMPs), and integrins. Experiments were performed on TCO-1 and HTC/C3 cells, which are human ATC cell lines. OSCC cell lines were used for comparison. Using the cell analysis system, cell migration was assessed on fibronectin-coated CIM-Plates, whereas invasion was assessed on fibronectin- and matrigel-coated CIM-Plates. SCC-4 cells exhibited high cell migration and invasion activity compared with other OSCC cell lines. TCO-1 cells exhibited equivalent cell invasion but stronger migration than SCC-4 cells. Although TCO-1 cells had strong invasive activity, they did not express MMP-9, unlike SCC-4 cells. Conversely, periostin expression was high in TCO-1 cells. Therefore, periostin expression appears to be associated with the cell migration and invasion activity of ATC. The RTCA system will be useful for the analysis of the metastatic characteristics of ATC in head and neck cancer

    Virus-Mediated Transient Expression Techniques Enable Functional Genomics Studies and Modulations of Betalain Biosynthesis and Plant Height in Quinoa

    スーパー作物キヌアにおける遺伝子機能の解析技術を開発 --優れた環境適応性や栄養特性の謎を解き、作物開発を加速化--. 京都大学プレスリリース. 2021-03-19.Quinoa (Chenopodium quinoa), native to the Andean region of South America, has been recognized as a potentially important crop in terms of global food and nutrition security since it can thrive in harsh environments and has an excellent nutritional profile. Even though challenges of analyzing the complex and heterogeneous allotetraploid genome of quinoa have recently been overcome, with the whole genome-sequencing of quinoa and the creation of genotyped inbred lines, the lack of technology to analyze gene function in planta is a major limiting factor in quinoa research. Here, we demonstrate that two virus-mediated transient expression techniques, virus-induced gene silencing (VIGS) and virus-mediated overexpression (VOX), can be used in quinoa. We show that apple latent spherical virus (ALSV) can induce gene silencing of quinoa phytoene desaturase (CqPDS1) in a broad range of quinoa inbred lines derived from the northern and southern highland and lowland sub-populations. In addition, we show that ALSV can be used as a VOX vector in roots. Our data also indicate that silencing a quinoa 3, 4-dihydroxyphenylalanine 4, 5-dioxygenase gene (CqDODA1) or a cytochrome P450 enzyme gene (CqCYP76AD1) inhibits betalain production and that knockdown of a reduced-height gene homolog (CqRHT1) causes an overgrowth phenotype in quinoa. Moreover, we show that ALSV can be transmitted to the progeny of quinoa plants. Thus, our findings enable functional genomics in quinoa, ushering in a new era of quinoa research
