    Acid ribonuclease activity of hepatoma cells

    Activities of intracellular RNase of the liver cytoplasm, normal liver cells exposed to 3'-Me-DAB and heaptoma cells, have been studied in correlation with the contents of RNA and DNA and morphologic changes of the cells with or without treating RNase. The data showed that in hepatoma cells the intracellular acid RNase activity decreases with the decrease of RNA and unchanged DNA contents and alkaline RNase activity. Morphologic observation proved that hepatoma cells show a small low massed vesicular or vacuolar endoplasmic reticulum having ribosomes. For the exposure to RNase the hepatoma cells proved to be much less resistant comparing to normal liver cells. The former lost the granules and was destroyed in its endoplasmic reticulum, whereas the latter retained ribosomes and ER. From these experiments it has been speculated that acid RNase in the cell may be involved in RNA synthesis and alkaline RNase in RNA decompostition, though the effect of the difference in concentration in the case given RNase experimentally can not be neglected.</p

    In an attempt to eliminate Japanese encephalitis virus in natural surroundings, pigs having maternal antibody were given inoculation of live-attenuated Japanese encephalitis vaccine and injection of Freund's complete adjuvant simultaneously. Titer of hemoagglutination inhibiting antibodies of pigs inoculated with live attenuated vaccine and complete adjuvant, was higher than that inoculated with vaccine alone and its titer persisted.</p

    Regulation of Id2 expression by CCAAT/enhancer binding protein β

    Mice deficient for Id2, a negative regulator of basic helix–loop–helix (bHLH) transcription factors, exhibit a defect in lactation due to impaired lobuloalveolar development during pregnancy, similar to the mice lacking the CCAAT enhancer binding protein (C/EBP) β. Here, we show that Id2 is a direct target of C/EBPβ. Translocation of C/EBPβ into the nucleus, which was achieved by using a system utilizing the fusion protein between C/EBPβ and the ligand-binding domain of the human estrogen receptor (C/EBPβ-ERT), demonstrated the rapid induction of endogenous Id2 expression. In reporter assays, transactivation of the Id2 promoter by C/EBPβ was observed and, among three potential C/EBPβ binding sites found in the 2.3 kb Id2 promoter region, the most proximal element was responsible for the transactivation. Electrophoretic mobility shift assay (EMSA) identified this element as a core sequence to which C/EBPβ binds. Chromatin immunoprecipitation (ChIP) furthermore confirmed the presence of C/EBPβ in the Id2 promoter region. Northern blotting showed that Id2 expression in C/EBPβ-deficient mammary glands was reduced at 10 days post coitus (d.p.c.), compared with that in wild-type mammary glands. Thus, our data demonstrate that Id2 is a direct target of C/EBPβ and provide insight into molecular mechanisms underlying mammary gland development during pregnancy

    The Aqua-Planet Experiment (APE): CONTROL SST Simulation

    Climate simulations by 16 atmospheric general circulation models (AGCMs) are compared on an aqua-planet, a water-covered Earth with prescribed sea surface temperature varying only in latitude. The idealised configuration is designed to expose differences in the circulation simulated by different models. Basic features of the aqua-planet climate are characterised by comparison with Earth. The models display a wide range of behaviour. The balanced component of the tropospheric mean flow, and mid-latitude eddy covariances subject to budget constraints, vary relatively little among the models. In contrast, differences in damping in the dynamical core strongly influence transient eddy amplitudes. Historical uncertainty in modelled lower stratospheric temperatures persists in APE.Aspects of the circulation generated more directly by interactions between the resolved fluid dynamics and parameterized moist processes vary greatly. The tropical Hadley circulation forms either a single or double inter-tropical convergence zone (ITCZ) at the equator, with large variations in mean precipitation. The equatorial wave spectrum shows a wide range of precipitation intensity and propagation characteristics. Kelvin mode-like eastward propagation with remarkably constant phase speed dominates in most models. Westward propagation, less dispersive than the equatorial Rossby modes, dominates in a few models or occurs within an eastward propagating envelope in others. The mean structure of the ITCZ is related to precipitation variability, consistent with previous studies.The aqua-planet global energy balance is unknown but the models produce a surprisingly large range of top of atmosphere global net flux, dominated by differences in shortwave reflection by clouds. A number of newly developed models, not optimised for Earth climate, contribute to this. Possible reasons for differences in the optimised models are discussed.The aqua-planet configuration is intended as one component of an experimental hierarchy used to evaluate AGCMs. This comparison does suggest that the range of model behaviour could be better understood and reduced in conjunction with Earth climate simulations. Controlled experimentation is required to explore individual model behavior and investigate convergence of the aqua-planet climate with increasing resolution

    Activity Measurement of Chitosanase by an Amperometric Biosensor

    An amperometric biosensor for the detection of glucosamine (GlcN) and chitosan oligosaccharides ((GlcN)(n)) was introduced for an activity measurement of chitosanase. By using the biosensor, an increase in the anodic current due to the production of GlcN and (GlcN)(n) by chitosanase was measured in a chitosan solution. The maximum value of the slope of the current increase was proportional to the enzyme concentration up to 1.4 mu g mL(-1).
