    Silk protein of the larval caddisfly, Sienopsyche marmorata (trichoptera: stenopsychidae)(trichoptera: stenopsychidae)

    Stenopsyche marmorata is one of the common caddisfly species inrivers and streamsin Japan which is distributed in East Palaearctic Region・ The larvae spin Silk fiber between stones on the bottom of Bowing water, the silk proteinofS・ marmorata would be one of the best sources of natural biopolymersthat could be used as materials for aquatic industrial purposes・ In order to analyze the basic characteristics towards further applications, we measured physical properties, thermal behaviorsand molecular confわrmation of the silk protein of larval S marmorata・ The wet weight of the larval body andthe silkgland of SOrata Were 543・5土5419and 44・7土17・9 mg, respectively, The water content of the liquid silk protein in the silkglands of the living larvae was 73.4土2.2%. The elongation atthe breaking point of the solid silk proteingland in dry state was 2・2%, whilethat of sampleinwet state was・ 6012%, suggestlngthe silk protein glands become soft and stretchable in wet conditions. FT-IR spectra of the silk protein film showed a major absorption band at 1650 cm-1 (Amide I band), which is attributed to the random coil molecular conformation.     Cytology in Hairy Cell Leukemia (HCL)

    Hairy cell leukemia (HCL) is a chronic lympho-proliferative disease. The disease is clinically characterized by anemia, granulocytopenia, thrombocytopenia, monocytopenia, splenomegaly. This leukemia cells have peculiar shape with hairlike projection, and express mature B-cell antigens such as, CD19,CD20 and Ig heavy chain on their surface. Nuclear and cytoplasm area ratio was low. Cell biological observation and chromosome analysis were performed in 12 patients. Translocation at 14q32 was found in 2 patients and loss of chromosome 6 was found in one patient. These cytogenetical results suggest taht HCL is different disease from chronic lymphocytic leukemia

    ホヤ初期発生におけるRNA結合タンパク質の役割に関する研究

    ホヤ卵では、細胞分化や形態形成に関わる細胞質因子の存在が明確に示されており、それらの卵内における局在場所も同定されている。近年の分子生物学の発達により、細胞質決定因子には母性mRNAが中心的な役割を果たしていることが明らかにされてきた。しかし、その母性mRNAが決定因子として働くには、その保護、局在化、翻訳調節などを司るRNA結合タンパク質の役割が必要不可欠である。そこで本研究では、カタユウレイボヤ卵より、様々なmRNA結合タンパク質を単離し、その発見解析と機能解析を行い、決定因子となる母性mRNAの解明と、それらが決定因子として働く分子メカニズムを明らかにすることを目的とした。今回、解析を行ったおもなRNA結合タンパク質は、以下4つである。(1)CiYB1;コールドショックドメインを持つY-boxタンパク質であり、in vitroでのRNA結合能が示された。CiYB1タンパク質は、受精卵ではマイオプラズムに、卵割期では胚後極のCABに局在していた。CABには、胚の前部の形成に重要な母性Ci-pem mRNAも局在しており、in vitroでのCiYB1タンパク質とCi-pem mRNAの結合も確かめられたことから、CiYB1によるCi-pem mRNAの翻訳抑制と胚の前側形成との関与が強く示唆された。(2)CiGRP1; glycine-rich RNA-binding proteinと高い相同性があり、そのmRNAとタンパク質は尾芽胚期の脳胞および間充織細胞で強く発現していた。既知の多くのglycine-rich RNA-binding proteinと異なり、CiGRP1は温度ショックによって誘導されないことを示し、神経系と間充織細胞の分化への関与を強く示唆した。RNAの強制発現による機能解析では、不十分ながら細胞分裂、細胞増殖との関連も示唆された。(3)CiRGG1;3つのRRMと2つのRGG-boxを持つnucleorin様の構造をしたRNA結合タンパク質であった。このmRNAは、2細胞期に一過的に胚の前方へ偏ることがわかり、2細胞期に胚前期側への細胞質の流れが存在することを示した。(4)CiMsi; Musashiタンパク質のホモログであり、mRNAは、尾芽胚期で神経系細胞と間充織細胞で発現していた。RNAの強制発現による機能解析では、頭部から尾部へかけての形態異常が起こり、尾基部付近の細胞分化に関する機能が示唆された。以上のように、カタユウレイボヤ卵から、様々なタイプのRNA結合タンパク質を単離し、その詳細な発現を解析するとともに、その機能に関する知見を得た。決定因子と直接関わる可能性が明確に示されたものはCiYB1のみであったが、多くのRNA結合タンパク質が卵内にあらかじめ蓄えられていることが明らかになった。今回の結果は、ホヤ卵におけるRNA結合タンパク質と母性mRNAが関与する様々な発生メカニズムを研究する上で重要な知見を与えるものとなった。また、本研究はホヤにおけるRNA結合タンパク質を多角的に解析した初めての研究である。甲南大学平成11年度(1999年度

    距骨壊死に対するアルミナセラミック性人工距骨置換術のSAFE-Qを用いた臨床成績

    Background: The purpose of this retrospective review was to evaluate the clinical outcomes of patients who underwent total talar replacement for talar necrosis using the SAFE-Q score, which was hypothesized to improve postoperatively. Methods: The study included 24 ankles of 22 patients who underwent total talar replacement from 2012 to 2018 and were evaluated using SAFE-Q preoperatively and postoperatively. Statistical analysis was performed using the mean values of the SAFE-Q and JSSF scale scores, and the range of ankle motion was compared before and 3 years after the surgery using the Wilcoxon signed-rank test. Results: The SAFE-Q scores improved postoperatively in the all the subcategories. “Pain and Pain-Related” changed from a mean value of 42.2±23.9 points preoperatively to a mean value of 84.6±12.6 points postoperatively (p<.01); “Physical Functioning and Daily Living” changed from 36.3±25.2 points to 73.4±20.5 points (p<.01); “Social Functioning” changed from 34.1±34.8 points to 81.0±25.3 points (p<.01); “Shoe-Related” changed from 41.3±28.9 points to 75.4±22.3 points (p<.01); “General Health and Well-Being” changed from 36.7±32.1 to 76.9±29.3 points (p<.01). Conclusion: Twenty-four osteonecrotic tali of 22 patients treated with alumina ceramic total talar replacement achieved good clinical results, as evaluated using the JSSF ankle/hindfoot score and SAFE-Q. Alumina ceramic total talar replacement is the mainstream treatment for talar osteonecrosis.博士(医学)・甲第856号・令和4年12月22

    Microarray-based global mapping of integration sites for the retrotransposon, intracisternal A-particle, in the mouse genome

    Mammalian genomes contain numerous evolutionary harbored mobile elements, a part of which are still active and may cause genomic instability. Their movement and positional diversity occasionally result in phenotypic changes and variation by causing altered expression or disruption of neighboring host genes. Here, we describe a novel microarray-based method by which dispersed genomic locations of a type of retrotransposon in a mammalian genome can be identified. Using this method, we mapped the DNA elements for a mouse retrotransposon, intracisternal A-particle (IAP), within genomes of C3H/He and C57BL/6J inbred mouse strains; consequently we detected hundreds of probable IAP cDNA–integrated genomic regions, in which a considerable number of strain-specific putative insertions were included. In addition, by comparing genomic DNAs from radiation-induced myeloid leukemia cells and its reference normal tissue, we detected three genomic regions around which an IAP element was integrated. These results demonstrate the first successful genome-wide mapping of a retrotransposon type in a mammalian genome

    Nuclear abnormalities in aspirated thyroid cells and chromosome aberrations in lymphocytes of residents near the Semipalatinsk Nuclear Test Site

    Chromosomal studies in peripheral lymphocytes from 63 residents near the Semipalatinsk nuclear test site, at ages of 52–63 years old, were performed in 2001–2002. A higher rate of chromosome aberrations was observed in the two contaminated villages, Dolon and Sarjal, compared with the control village, Kokpekti. Moreover, a relationship of frequency of cells with radiation induced chromosome aberrations and the previously estimated exposure dose was observed. Furthermore, apparent nuclear abnormalities (ANA) of thyroid follicular cells were studied in 30 out of 63 residents, who were examined for chromosome aberrations. A higher rate of ANA was also found in the residents in the exposed villages compared with those in the control village. These results suggest radiation effects both on the chromosomes in peripheral lymphocytes and on the follicular cells in the thyroid
