    Massive gravitons dark matter scenario revisited

    We reexamine the massive graviton dark matter scenario (MGCDM) which was recently considered as an alternative to dark energy models. When introducing the native and effective equations of state (EoS), it is shown that there is no phantom phase in the evolution toward the far past. Also we show that the past accelerating phase arises from the interaction between massive graviton and cold dark matter.Comment: 13 pages, 6 figure

    Palmitate reduces starvation-induced ER stress by inhibiting ER-phagy in hypothalamic cells

    Palmitate is a saturated fatty acid that is well known to induce endoplasmic reticulum (ER) stress and autophagy. A high-fat diet increases the palmitate level in the hypothalamus, the main region of the brain regulating energy metabolism. Interestingly, hypothalamic palmitate level is also increased under starvation, urging the study to distinguish the effects of elevated hypothalamic palmitate level under different nutrient conditions. Herein, we show that ER-phagy (ER-targeted selective autophagy) is required for progress of ER stress and that palmitate decreases ER stress by inhibiting ER-phagy in hypothalamic cells under starvation. Palmitate inhibited starvation-induced ER-phagy by increasing the level of B-cell lymphoma 2 (Bcl-2) protein, which inhibits autophagy initiation. These findings suggest that, unlike the induction of ER stress under nutrient-rich conditions, palmitate protects hypothalamic cells from starvation-induced stress by inhibiting ER-phagy. © 2021, The Author(s).1

    Dual priming oligonucleotide system for the multiplex detection of respiratory viruses and SNP genotyping of CYP2C19 gene

    Successful PCR starts with proper priming between an oligonucleotide primer and the template DNA. However, the inevitable risk of mismatched priming cannot be avoided in the currently used primer system, even though considerable time and effort are devoted to primer design and optimization of reaction conditions. Here, we report a novel dual priming oligonucleotide (DPO) which contains two separate priming regions joined by a polydeoxyinosine linker. The linker assumes a bubble-like structure which itself is not involved in priming, but rather delineates the boundary between the two parts of the primer. This structure results in two primer segments with distinct annealing properties: a longer 5′-segment that initiates stable priming, and a short 3′-segment that determines target-specific extension. This DPO-based system is a fundamental tool for blocking extension of non-specifically primed templates, and thereby generates consistently high PCR specificity even under less than optimal PCR conditions. The strength and utility of the DPO system are demonstrated here using multiplex PCR and SNP genotyping PCR

    Study on Personal Characteristics and Emotional Affordances through User Activity Analysis

    Design methodology should address psychological and cognitive aspects based on human emotion and feeling as well as physical and functional aspects based on product structure. Donald Norman introduced perceived affordances as characteristics in the appearance of an object that gives clues for its proper operations. We conducted a case study in a public space - building lobby - used by many general people. User activities and behaviors were analyzed in several specific tasks given to twenty participants with similar cultural background in the lobby of a building they have never been to before. Then we identified affordances and building structure elements, called affordance features, critically related to those affordances. While performing the tasks in the same building lobby, user activities can be classified into several different groups for each task based on affordance features involved in their activities. These user activity differences are then compared with their personal cognitive characteristics derived from their perception and judgment preferences using intuitive and factual perception orientation and objective and subjective decision making inclination respectively. For user of less common activities for some tasks, personal cognitive characteristics have been identified. This study could help in affordance-based design so that alternative affordance features are designed considering diverse user cognitive characteristics

    Meteorin regulates mesendoderm development by enhancing nodal expression

    During gastrulation, distinct lineage specification into three germ layers, the mesoderm, endoderm and ectoderm, occurs through an elaborate harmony between signaling molecules along the embryonic proximo-distal and anterior-posterior axes, and Nodal signaling plays a key role in the early embryonic development governing embryonic axis formation, mesoderm and endoderm specification, and left-right asymmetry determination. However, the mechanism by which Nodal expression is regulated is largely unknown. Here, we show that Meteorin regulates Nodal expression and is required for mesendoderm development. It is highly expressed in the inner cell mass of blastocysts and further in the epiblast and extra-embryonic ectoderm during gastrulation. Genetic ablation of the Meteorin gene resulted in early embryonic lethality, presumably due to impaired lineage allocation and subsequent cell accumulation. Embryoid body culture using Meteorin-null embryonic stem (ES) cells showed reduced Nodal expression and concomitant impairment of mesendoderm specification. Meteorin-null embryos displayed reduced levels of Nodal transcripts before the gastrulation stage, and impaired expression of Goosecoid, a definitive endoderm marker, during gastrulation, while the proximo-distal and anterior-posterior axes and primitive streak formation were preserved. Our results show that Meteorin is a novel regulator of Nodal transcription and is required to maintain sufficient Nodal levels for endoderm formation, thereby providing new insights in the regulation of mesendoderm allocation.open1113sciescopu

    Energy and carbon footprint reduction during textile-based product design and manufacturing

    Due to concerns over non-renewable energy consumption and associated emissions, industry has sought methods and technologies to support energy efficiency practices and use of alternative energy during product manufacturing, use, and end-of-life. Efforts have been undertaken to more precisely calculate environmental metrics, such as energy consumption and carbon footprint, to support broader sustainable design activities. The work reported endeavours to integrate sustainability principles into the design of products, manufacturing processes, and relevant supply chain networks to assist decision makers. Two backpacks are evaluated to examine the influence of design choices on energy consumption and carbon footprint. The study system boundary includes raw material extraction, materials processing, manufacturing operations, and transportation for each component. The results show that manufacturing processes dominate transportation-related impacts. The work appears to be the first to apply a comprehensive process-based approach to estimate cradle-to-gate energy consumption and carbon footprint for textile-based product design variants
