    A comprehensive review of the applications of bacteriophage-derived endolysins for foodborne bacterial pathogens and food safety: recent advances, challenges, and future perspective

    Foodborne diseases are caused by food contaminated by pathogenic bacteria such as Escherichia coli, Salmonella, Staphylococcus aureus, Listeria monocytogenes, Campylobacter, and Clostridium, a critical threat to human health. As a novel antibacterial agent against foodborne pathogens, endolysins are peptidoglycan hydrolases encoded by bacteriophages that lyse bacterial cells by targeting their cell wall, notably in Gram-positive bacteria due to their naturally exposed peptidoglycan layer. These lytic enzymes have gained scientists’ interest in recent years due to their selectivity, mode of action, engineering potential, and lack of resistance mechanisms. The use of endolysins for food safety has undergone significant improvements, which are summarized and discussed in this review. Endolysins can remove bacterial biofilms of foodborne pathogens and their cell wall-binding domain can be employed as a tool for quick detection of foodborne pathogens. We explained the applications of endolysin for eliminating pathogenic bacteria in livestock and various food matrices, as well as the limitations and challenges in use as a dietary supplement. We also highlight the novel techniques of the development of engineering endolysin for targeting Gram-negative bacterial pathogens. In conclusion, endolysin is safe and effective against foodborne pathogens and has no adverse effect on human cells and beneficial microbiota. As a result, endolysin could be employed as a functional bio-preservative agent to improve food stability and safety and maintain the natural taste of food quality

    Surgical Outcome of Otogenic Brain Abscess in Chronic Suppurative Otitis Media

    Objective: To determine the surgical out come of otogenic brain abscess in chronic suppurative otitis media.Material and Methods: This descriptive study was conducted at the departments of ENT, Head, Neck Surgery and Neurosurgery Postgraduate Medical Institute (PGMI) Lady Reading Hospital Peshawar from June 2004 to May 2009 with a total duration of five years. The diagnostic criteria were detailed history, thorough Neuro-otological examination and radiological investigations. These patients were admitted into ENT Department and they were put on quadruple therapy. CT scans were performed on emergency basis and as the patients were diagnosed as having otogenic brain abscess urgent evacuation of abscess was done by neurosurgeon. All the patients with established brain abscess were included in this study while those patients having small multiple abscesses, early cereberitis and non otogenic brain abscesses were excluded from study. A well informed consent was taken from patient explaining the procedure, its risks, benefits and associated complications. The abscesses were primarily treated by neurosurgeons and the ears were later on treated by ENT surgeons. There was neither intra operative nor postoperative complication.Results: This study included 22 patients in the age range of 12-51 years with mean age 25 years. Sixteen were male and six were female, with female to male ratio 1 : 2.6. These patients presented with history of discharging ear, headache, vomiting and disorientation. The signs of disease in these patients were granulation tissues/ cholesteatoma in ear and papilloedema. CT scans were performed in all 22 cases (100%) while MRI in 10 cases (45.45%). There were single brain abscess in 14 cases (63.63%) while 8 cases (44.45%) having multiple brain abscesses. Mainly abscess were found in temporal region followed by cerebellar. The patients with chronic ear disease were treated by radical mastoidectomy and modified radical mastoidectomy after treating the brain abscess by Neurosurgeon. The Neurosurgical procedures performed were capsulotmies in 13 cases (59.09%), Bur hole aspiration in 7 cases (31.81%) and Trans mastoid excision in 2 cases (9.09%). These patients were followed for 3 monthly having CT scan with contrast with bone window.Conclusion: We concluded from our study that otogenic brain abscess is a life threatening complication, so early identification with help of CT scan and prompt surgical intervention with appropriate treatment of middle ear (primary disease) is essential for better outcom

    Phage-Encoded Endolysins

    Due to the global emergence of antibiotic resistance, there has been an increase in research surrounding endolysins as an alternative therapeutic. Endolysins are phage-encoded enzymes, utilized by mature phage virions to hydrolyze the cell wall from within. There is significant evidence that proves the ability of endolysins to degrade the peptidoglycan externally without the assistance of phage. Thus, their incorporation in therapeutic strategies has opened new options for therapeutic application against bacterial infections in the human and veterinary sectors, as well as within the agricultural and biotechnology sectors. While endolysins show promising results within the laboratory, it is important to document their resistance, safety, and immunogenicity for in-vivo application. This review aims to provide new insights into the synergy between endolysins and antibiotics, as well as the formulation of endolysins. Thus, it provides crucial information for clinical trials involving endolysins

    Epidemiology of coronaviruses, genetics, vaccines, and scenario of current pandemic of coronavirus diseases 2019 (COVID-19): a fuzzy set approach

    Human coronaviruses (HCoVs) are associated with a range of respiratory complications. In the last two decades, three major outbreaks have been reported due to HCoVs including the current pandemic. In December 2019, a newly emerged virus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was first reported in Wuhan city, China. This paper presents a detailed review of the literature and discusses the uncertain spread of coronavirus disease 2019 (COVID-19) using fuzzy set as classical set theory logic to measure uncertainty and vagueness of COVID-19 in China. Our findings show that both infection and death rate touched the peak (normal fuzzy sets) and have shown a decline. The graphs are not convex, which shows that there remains much uncertainty in the spread of COVID-19. Effective vaccines are clearly needed to control and prevent the COVID-19 pandemic

    Background: Cryptorchidism, undescended testes, is a pathological condition that is due to failure of descent of testes in the scrotum. This study was aimed to determine the diagnostic accuracy of diffusion-weighted magnetic resonance imaging (DW-MRI) in localization of undescended testes taking laparoscopic findings as the gold standard. Methods: A cross-sectional study was conducted in the radiology department of a tertiary care hospital from September 27, 2018 to September 26, 2019. A total of 416 patients were included. Abdomen-pelvic and scrotal ultrasound were performed. Preoperative abdominal and pelvic DW-MRI was performed with a 1.5-T MRI system using a body coil. All study patients underwent laparoscopic exploration. Intra-abdominal atrophic testes were treated with laparoscopic orchiectomy and orchiectomy samples were taken for histopathologic examination. DW-MRI findings were correlated with laparoscopic findings. A 2 x 2 table was used to calculate the sensitivity, specificity, positive predictive value, negative predictive value and diagnostic accuracy of DW-MRI taking laparoscopy as the gold standard. Results: The mean age was 17.08 ± 7.99 years and the mean BMI was 19.36 ± 4.96 kg/m2. In our study, 34.1% of cases were diagnosed as undescended testes localized by DWI-MRI and 51% of cases were diagnosed as undescended testes localized via laparoscopy. Sensitivity, specificity, PPV, NPV and accuracy were 65.1%, 98%, 97.2%, 73% and 81.3% respectively. Conclusion: DW-MRI improves the detection of undescended testes. DW-MRI can be a recommended imaging tool to increase the preoperative diagnostic accuracy of MRI in localizing nonpalpable undescended testes

    Starch-grafted polyacrylic acid copolymer with acrylamide: an advanced adsorbent for Victoria green B dye removal and environmental remediation

    A new adsorbent, starch grafted polyacrylic acid copolymer with polyacrylamide (SG@AA-co-AM), was synthesized using free radical polymerization techniques. Proton nuclear magnetic resonance (1H NMR), Fourier-transform infrared spectrophotometry (FTIR), X-ray diffraction analysis (XRD), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), particle size distribution (PSD), and fluorescent electron microscopy (FEM) were employed to elucidate the structure, crystalline nature, thermal stability, and surface morphology of SG@AA-co-AM. SG@AA-co-AM was utilized as an adsorbent for the removal of Victoria green B (VGB) dye from wastewater. SG@AA-co-AM exhibited a removal percentage (% R) of 97.6% towards VGB under optimized conditions: a contact time of 30 min, temperature of 25 °C, adsorbent dose of 20 mg, pH of 8, concentration of dye solution of 20 ppm, and volume of dye solution of 30 mL. The point of zero charge (PZC) for SG@AA-co-AM was determined to be 5.2. Nonlinear pseudo-second-order (PSO) and Langmuir adsorption isotherm models best fitted to the experimental data, with regression coefficients (R2) of 0.95 and 0.99, respectively. The results confirmed the chemi-sorption and monolayer adsorption of VGB onto SG@AA-co-AM. Thermodynamic studies revealed that the adsorption of VGB onto SG@AA-co-AM is endothermic and spontaneous. Furthermore, the regeneration experiment showed a decline of 3.9% after five cycles, confirming the economical and reusable nature of SG@AA-co-AM.<br/

    The applications of animal models in phage therapy: An update

    The rapid increase in antibiotic resistance presents a dire situation necessitating the need for alternative therapeutic agents. Among the current alternative therapies, phage therapy (PT) is promising. This review extensively summarizes preclinical PT approaches in various in-vivo models. PT has been evaluated in several recent clinical trials. However, there are still several unanswered concerns due to a lack of appropriate regulation and pharmacokinetic data regarding the application of phages in human therapeutic procedures. In this review, we also presented the current state of PT and considered how animal models can be used to adapt these therapies for humans. The development of realistic solutions to circumvent these constraints is critical for advancing this technology
