HDL cholesterol is known to be inversely correlated with cardiovascular disease due to its diverse antiatherogenic functions. SR-BI mediates the selective uptake of HDL-C. SR-BI knockout diminishes but does not completely block the transport of HDL; other receptors may be involved. Ectopic ATP synthase β-chain in hepatocytes has been previously characterized as an apoA-I receptor, triggering HDL internalization. This study was undertaken to identify the overexpression of ectopic ATP synthase β-chain on DIL-HDL uptake in primary hepatocytes in vitro and on plasma HDL levels in SR-BI knockout mice. Human ATP synthase β-chain cDNA was delivered to the mouse liver by adenovirus and GFP adenovirus as control. The adenovirus-mediated overexpression of β-chain was identified at both mRNA and protein levels on mice liver and validated by its increasing of DiL-HDL uptake in primary hepatocytes. In response to hepatic overexpression of β-chain, plasma HDL-C levels and cholesterol were reduced in SR-BI knockout mice, compared with the control. The present data suggest that ATP synthase β-chain can serve as the endocytic receptor of HDL, and its overexpression can reduce plasma HDL-C

Objective. This study aimed to determine the effects of melatonin on insulin resistance in obese patients with acanthosis nigricans (AN). Methods. A total of 17 obese patients with acanthosis nigricans were recruited in a 12-week pilot open trial. Insulin sensitivity, glucose metabolism, inflammatory factors, and other biochemical parameters before and after the administration of melatonin were measured. Results. After 12 weeks of treatment with melatonin (3 mg/day), homeostasis model assessment insulin resistance index (HOMA-IR) (8.99 ± 5.10 versus 7.77 ± 5.21, p<0.05) and fasting insulin (37.09 5 ± 20.26 μU/ml versus 32.10 ± 20.29 μU/ml, p<0.05) were significantly decreased. Matsuda index (2.82 ± 1.54 versus 3.74 ± 2.02, p<0.05) was significantly increased. There were also statistically significant declines in the AN scores of the neck and axilla, body weight, body mass index, body fat, visceral index, neck circumference, waist circumference, and inflammatory markers. Conclusions. It was concluded that melatonin could improve cutaneous symptoms in obese patients with acanthosis nigricans by improving insulin sensitivity and inflammatory status. This trial is registered with ClinicalTrials.gov NCT02604095

Objective. We aimed to investigate the relationship between FGF21 and obesity-related acanthosis nigricans (AN). Methods. 40 obese patients without AN (OB group), 40 obese patients with AN (AN group), and 40 healthy volunteers (control group, CON) were included in this study. Weight, BMI, lipid profile, FFA, UA, and CRP were measured in all participants. Oral glucose tolerance tests (OGTT) were performed and serum glucose and plasma insulin were measured. Serum FGF21 was measured by ELISA. Results. Compared with OB group, AN group had higher levels of fasting insulin and homeostasis model of assessment for insulin resistance (HOMA-IR) (P<0.05), but lower serum levels of blood glucose. The difference of FGF21 among three groups was significant and AN group showed the highest serum level of FGF21 (P<0.05). Serum FGF21 was most positively correlated with fasting insulin and HOMA-IR. Multiple logistic analysis showed that FGF21 was the independent risk factor for AN (OR 4.550; 95% CI 1.054–19.635; P=0.042). Conclusion. AN patients had more serious hyperinsulinemia but better serum levels of blood glucose than OB. Increased FGF21 is associated with AN in obese patients and may be considered as compensatory response to the decreased insulin sensitivity

Effects of Periodic Tidal Elevations on the Air-Sea Momentum and Turbulent Heat Fluxes in the East China Sea

Using bulk formulas, two-year platform (fastened to the seabed) hourly observations from 2016 to 2017 in the East China Sea (121.6° E, 32.4° N) are used to investigate the role of the tide-induced surface elevation in changing the fixed observational height and modifying the momentum and air-sea turbulent heat fluxes. The semidiurnal tide-dominated elevation anomalies ranging from −3.6 to 3.9 m change the fixed platform observational height. This change causes hourly differences in the wind stress and latent and sensible heat fluxes between estimates with and without considering surface elevation, with values ranging from −1.5 × 10−3 Nm−2, −10.2 Wm−2, and −3.6 Wm−2 to 2.2 × 10−3 Nm−2, 8.4 Wm−2, and 4.6 Wm−2, respectively. More significant differences occur during spring tides. The differences show weak dependence on the temperature, indicating weak seasonal variations. The mean (maximum) difference percentage relative to the mean magnitude is approximately 3.5% (7%), 1.5% (3%), and 1.5% (3%) for the wind stress and latent and sensible heat fluxes, respectively. The boundary layer stability (BLS) can convert from near-neutral conditions to stable and unstable states in response to tide-induced changes in the observational height, with a probability of occurrence of 2%. Wind anomalies play dominant roles in determining the hourly anomalies of the latent heat flux, regardless of the state of the BLS. Extreme cases, including the cold air outbreak in 2016, tropical cyclones Meranti in 2016, and Ampil in 2018, are also examined. This study will facilitate future observation-reanalysis comparisons in the studied coastal region where ocean–atmosphere-land interactive processes are significant
