    AN EXPLORATION OF ORGANIZATIONAL BUYING BEHAVIOR IN THE PUBLIC SECTOR

    This dissertation begins with a comprehensive examination of the current state of research regarding organizational buying behavior. Through this review we identify a significant gap in our existing knowledge regarding organizational buying behavior in the public sector. Due to the high level of impact that government purchasing has on the economy, and the nuances that differentiate public from private sector purchasing practices, I further explore organizational buying behavior in the public sector to make the following contributions. First, I highlight the common practice in business-to-government (B2G) and business-to-business (B2B) transactions where buyers limit suppliers’ access to them during the buying process. This research terms these buyers “barricaded buyers.” Despite their prominence in practice, research related to barricaded buyers remains virtually non-existent. Therefore, the present research draws on insights gleaned from eight case studies over a period of approximately eighteen months to shed light on this important topic. Second, this dissertation advances a conceptual framework highlighting competitive actions a focal supplier can take to improve its selection likelihood when selling to barricaded buyers. The framework identifies novel ways suppliers can gain advantage by reducing competitive intensity in the pre-barricade phase (e.g., by peacocking) and by enhancing their RFP response quality in the post-barricade phase (e.g., by offering consummate solutions). Importantly, the framework invokes the notion of strategic information disclosure whereby a focal supplier may gain advantage by knowing when to convey what types of information in barricaded buying environments

    Forest Stand Preference of Sirex Nigricornis, and Sirex Noctilio Hazard in the Southeastern United States

    The Eurasian wood wasp, Sirex noctilio, is considered a secondary pest in its native range; however, it has caused significant economic damage when introduced to pine plantations in the Southern Hemisphere. Sirex noctilio was recently introduced to the northeastern U.S., which has raised concerns about its potential impact on Southeastern pine plantations. This research was conducted to understand how silvicultural management affects populations of a native wood wasp, Sirex nigricornis, a wood wasp with similar ecosystem functions as S. noctilio. Sirex nigricornis abundance was higher in un-managed pine plantations than in managed plantations, mixed, and old growth forests. Additionally, geospatial models were built displaying S. noctilio hazard for the Southeastern U.S. based on oviposition host preference assays and historical outbreak information. Sirex noctilio hazard models will inform land managers about areas of greatest concern under various scenarios and should be used to decrease susceptibility of pine forests to this pest

    Model-guided design of ligand-regulated RNAi for programmable control of gene expression

    Progress in constructing biological networks will rely on the development of more advanced components that can be predictably modified to yield optimal system performance. We have engineered an RNA-based platform, which we call an shRNA switch, that provides for integrated ligand control of RNA interference (RNAi) by modular coupling of an aptamer, competing strand, and small hairpin (sh) RNA stem into a single component that links ligand concentration and target gene expression levels. A combined experimental and mathematical modelling approach identified multiple tuning strategies and moves towards a predictable framework for the forward design of shRNA switches. The utility of our platform is highlighted by the demonstration of fine-tuning, multi-input control, and model-guided design of shRNA switches with an optimized dynamic range. Thus, shRNA switches can serve as an advanced component for the construction of complex biological systems and offer a controlled means of activating RNAi in disease therapeutics

    Synthetic control of a fitness tradeoff in yeast nitrogen metabolism

    Background: Microbial communities are involved in many processes relevant to industrial and medical biotechnology, such as the formation of biofilms, lignocellulosic degradation, and hydrogen production. The manipulation of synthetic and natural microbial communities and their underlying ecological parameters, such as fitness, evolvability, and variation, is an increasingly important area of research for synthetic biology. Results: Here, we explored how synthetic control of an endogenous circuit can be used to regulate a tradeoff between fitness in resource abundant and resource limited environments in a population of Saccharomyces cerevisiae. We found that noise in the expression of a key enzyme in ammonia assimilation, Gdh1p, mediated a tradeoff between growth in low nitrogen environments and stress resistance in high ammonia environments. We implemented synthetic control of an endogenous Gdh1p regulatory network to construct an engineered strain in which the fitness of the population was tunable in response to an exogenously-added small molecule across a range of ammonia environments. Conclusion: The ability to tune fitness and biological tradeoffs will be important components of future efforts to engineer microbial communities

    Video-Poker Play in the Laboratory: The Effect of Information and Monetary Value on Rates of Play

    Previous research has found that participants will risk more credits across a video-poker session when they are required to play the optimal cards than when they have complete control over the game, a finding that would seem at least partially inconsistent with the illusion of control (Langer, 1975). Forty-two participants were recruited to play video poker in two sessions, one in which the game informed them of the optimal cards to play and one in which it did not. The session length for some participants was limited by time and for other participants by the number of hands played. Some of the participants played the game for money whereas others did not. When sessions were limited by time, the previous research results were replicated. However, when the sessions were limited by the number of hands played, no differences in gambling were observed between the two sessions. These results indicate that the effect of the game-provide information is to increase the speed at which people play, not to alter their gambling (i.e., betting). Implications of this finding are discussed

    Geophysical characterisation of the Onewhero and Kellyville volcanic complexes, South Auckland Volcanic Field

    The Quaternary-aged (1.59 – 0.51 Ma) intraplate, monogenetic, basaltic South Auckland Volcanic Field consists of at least 82 volcanic centres that span an area of 300 km². This study focusses on the volcanic and sedimentary histories of the Onewhero and Kellyville maars, interpreted from additional geological observations, which build upon previous geological studies, and gravimetric and magnetic surveys. New geological observations of the Onewhero maar reveal the presence of diatomaceous sediment, and at least one lava flow, that probably originated from the nearby Klondyke cone to the south, and which occupies a significant area of the crater floor. A geological investigation of the Kellyville maar reveals a crater that has been partially filled by a thick accumulation of basalt and scoria and overlain by Karapiro Formation sediments and diatomite. Available borehole stratigraphy suggests that the Glass Hill basalt cone present in the Kellyville maar was an early-stage volcanic product rather than a late-stage product as previously thought. Gravimetric surveys in the Onewhero and Kellyville maars revealed contrasting crater fill deposits. Raw gravity data was acquired in the field, corrected for drift and then reduced to a Bouguer anomaly. The Onewhero maar is characterised by an anomaly of -5.8 mGal that has a concave profile. The post-eruption geological body present at Onewhero is two-dimensionally modelled with a density of 1.3 g/cm³, and has a maximum thickness of 100 m in the northwest of the crater. The Kellyville maar is characterised by an anomaly of +2.5 mGal that has a convex profile. The dominant geological body present in the crater is a lava-lake deposit that is two-dimensionally modelled with a density of 2.9 g/cm³, and has a maximum thickness of 60 m. The total magnetic field strength of the Onewhero and Kellyville maars was recorded with a proton magnetometer and the values were mapped in ArcMap. In the Onewhero maar, two subdued anomalies were outliers. A positive magnetic anomaly (213 nT) with a broad crescent shape was identified in the middle of the crater and interpreted to be either an extrusive feeder dyke or an accumulation of highly magnetic volcanic sediment deposited in a topographic trough. A circular negative anomaly (-1057 nT) in the northwest of the crater is an accumulation of lake sediment with a low magnetic susceptibility. In the Kellyville maar, two contrasting anomalies were observed. The strongly positive anomaly (1275 nT) corresponds to the geographical position of Glass Hill, a basaltic cone. A strong negative anomaly (-3315 nT) in the middle of the crater corresponds to a known deposit of diatomite. Overall, the structure of the Onewhero maar, as inferred by this investigation, is similar to many other maars globally and its sedimentary fill is remarkably similar to the Dottingen maar in Germany. The Kellyville maar has a more unique structure but can be compared to the Domain maar in the nearby Auckland Volcanic Field

    A summary of water-quality and salt marsh monitoring, Humboldt Bay, California

    This report summarizes data-collection activities associated with the U.S. Geological Survey Humboldt Bay Water-Quality and Salt Marsh Monitoring Project. This work was undertaken to gain a comprehensive understanding ofwater-quality conditions, salt marsh accretion processes, marsh-edge erosion, and soil-carbon storage in Humboldt Bay, California. Multiparameter sondes recorded water temperature, specific conductance, and turbidity at a 15-minute timestep at two U.S. Geological Survey water-quality stations: Mad River Slough near Arcata, California (U.S. Geological Survey station 405219124085601) and (2) Hookton Slough near Loleta, California (U.S. Geological Survey station 404038124131801). At each station, discrete water samples were collected to develop surrogate regression models that were used to compute a continuous time seriesof suspended-sediment concentration from continuously measured turbidity. Data loggers recorded water depth at a 6-minute timestep in the primary tidal channels (Mad River Slough and Hookton Slough) in two adjacent marshes (Mad River marsh and Hookton marsh). The marsh monitoring network included five study marshes. Three marshes (Mad River, Manila, and Jacoby) are in the northern embayment of Humboldt Bay and two marshes (White and Hookton) are in the southern embayment. Surface deposition and elevation change were measured using deep rod surface elevation tables and feldspar marker horizons. Sediment characteristics and soil-carbon storage were measured using a total of 10 shallow cores, distributed across 5 study marshes, collected using an Eijkelkamp peat sampler. Rates of marsh edge erosion (2010–19) were quantified in four marshes (Mad River, Manila, Jacoby, and White) by estimating changes in the areal extent of the vegetated marsh plain using repeat aerial imagery and light detection and ranging (LiDAR)-derived elevation data. During the monitoring period (2016–19), the mean suspended-sediment concentration computed for Hookton Slough (50±20 milligrams per liter [mg/L]) was higher than Mad River Slough (18±7 mg/L). Uncertainty in mean suspended-sediment concentration values is reported using a 90-percent confidence interval. Across the five study marshes, elevation change (+1.8±0.6 millimeters per year[mm/yr]) and surface deposition (+2.5±0.5 mm/yr) were lower than published values of local sea-level rise (4.9±0.8 mm/yr), and mean carbon density was 0.029±0.005 grams of carbon per cubic centimeter. From 2010 to 2019, marsh edge erosion and soil carbon loss were greatest in low-elevation marshes with the marsh edge characterized by a gentle transition from mudflat to vegetated marsh (herein, ramped edge morphology) and larger wind-wave exposure. Jacoby Creek marsh experienced the greatest edge erosion. In total, marsh edge erosion was responsible for 62.3 metric tons of estuarine soil carbon storage loss across four study marshes. Salt marshes are an important component of coastal carbon, which is frequently referred to as “blue carbon.” The monitoring data presented in this report provide fundamental information needed to manage blue carbon stocks, assess marsh vulnerability, inform sea-level rise adaptation planning, and build coastal resiliency to climate change

    The GALEX View of "Boyajian's Star" (KIC 8462852)

    The enigmatic star KIC 8462852, informally known as "Boyajian's Star", has exhibited unexplained variability from both short timescale (days) dimming events, and years-long fading in the Kepler mission. No single physical mechanism has successfully explained these observations to date. Here we investigate the ultraviolet variability of KIC 8462852 on a range of timescales using data from the GALEX mission that occurred contemporaneously with the Kepler mission. The wide wavelength baseline between the Kepler and GALEX data provides a unique constraint on the nature of the variability. Using 1600 seconds of photon-counting data from four GALEX visits spread over 70 days in 2011, we find no coherent NUV variability in the system on 10-100 second or months timescales. Comparing the integrated flux from these 2011 visits to the 2012 NUV flux published in the GALEX-CAUSE Kepler survey, we find a 3% decrease in brightness for KIC 8462852. We find this level of variability is significant, but not necessarily unusual for stars of similar spectral type in the GALEX data. This decrease coincides with the secular optical fading reported by Montet & Simon (2016). We find the multi-wavelength variability is somewhat inconsistent with typical interstellar dust absorption, but instead favors a RV_V = 5.0 ±\pm 0.9 reddening law potentially from circumstellar dust.Comment: 8 pages, 4 figures, ApJ Accepte
