    Evaluation of low density array technology for quantitative parallel measurement of multiple genes in human tissue

    BACKGROUND: Low density arrays (LDAs) have recently been introduced as a novel approach to gene expression profiling. Based on real time quantitative RT-PCR (QRT-PCR), these arrays enable a more focused and sensitive approach to the study of gene expression than gene chips, while offering higher throughput than more established approaches to QRT-PCR. We have now evaluated LDAs as a means of determining the expression of multiple genes simultaneously in human tissues and cells. RESULTS: Comparisons between LDAs reveal low variability, with correlation coefficients close to 1. By performing 2-fold and 10-fold serial dilutions of cDNA samples in the LDAs we determined a clear linear relationship between the gene expression data points over 5 orders of magnitude. We also showed that it is possible to use LDAs to accurately and quantitatively detect 2-fold changes in target copy number as well as measuring genes that are expressed with low and high copy numbers in the range of 1 × 10(2 )– 1 × 10(6 )copies. Furthermore, the data generated by the LDA from a cell based pharmacological study were comparable to data generated by conventional QRT-PCR. CONCLUSION: LDAs represent a valuable new approach for sensitive and quantitative gene expression profiling

    Running impact forces: from half a leg to holistic understanding – comment on Nigg et al.

    Running impact forces have immediate relevance for the muscle tuning paradigm proposed here and broader relevance for overuse injuries, shoe design and running performance. Here, we consider their mechanical basis. Several studies demonstrate that the vertical ground reaction force-time (vGRFT) impulse, from touchdown to toe-off, corresponds to the instantaneous accelerations of the body’s entire mass (Mb) divided into two or more portions. The simplest, a two-mass partitioning of the body (lower-limb, M1=0.08•Mb; remaining mass, M2=0.92•Mb) can account for the full vGRFT waveform under virtually all constant-speed, level-running conditions. Model validation data indicate that: 1) the non-contacting mass, M2, often accounts for one-third or more of the early “impact” portion of the vGRFT, and 2) extracting a valid impact impulse from measured force waveforms requires only lower-limb motion data and the fixed body mass fraction of 0.08 for M1

    Structure and dynamics of a subglacial discharge plume in a Greenlandic fjord

These plumes impact submarine melting, calving, and fjord circulation. Observations of plume properties and dynamics are challenging due to their proximity to the calving edge of glaciers. Therefore, to date information on these plumes has been largely derived from models. Here we present temperature, salinity, and velocity data collected in a plume that surfaced at the edge of Saqqarliup Sermia, a midsized Greenlandic glacier. The plume is associated with a narrow core of rising waters approximately 20 m in diameter at the ice edge that spreads to a 200 m by 300 m plume pool as it reaches the surface, before descending to its equilibrium depth. Volume flux estimates indicate that the plume is primarily driven by subglacial discharge and that this has been diluted in a ratio of 1:10 by the time the plume reaches the surface. While highly uncertain, meltwater fluxes are likely 2 orders of magnitude smaller than the subglacial discharge flux.     Functional Status and Out-of-Hospital Outcomes in Different Types of Vascular Surgery Patients

    Background: We aimed to determine the correlation between the functional status at discharge in non-cardiac vascular surgery patients and the out-of-hospital mortality. Methods: We performed a retrospective cohort study including adult non-cardiac vascular surgery patients (open, endovascular and venous procedures) surviving hospitalization in Boston, Massachusetts, USA. The exposure of interest was functional status determined by a licensed physical therapist at hospital discharge and rated based on qualitative categories adapted from the Functional Independence Measure. The primary outcome was all cause 90 day mortality after hospital discharge. The secondary outcome was readmission within 30days. Adjusted odds ratios were estimated by multivariable logistic regression models. Results: This cohort included 2318 patients (male 51%; mean age 61 +/- 17.7). After evaluation by a physiotherapist, 425 patients scored the lowest functional status, 631 scored moderately low, 681 moderately high and 581 scored the highest functional status. The lowest functional status was associated with a 3.41-fold increased adjusted odds for 90-day mortality (95%CI, 1.70- 6.84) compared to patients with the highest functional status. When excluding venous intervention patients, the adjusted odds ratio was 6.76 (95%CI, 2.53-18.12) for the 90-day mortality post discharge. The adjusted odds for readmission within 30-days was 1.5-fold increase in patients with the lowest functional status (95%CI, 1.04-2.20). Conclusions: In vascular surgery patients surviving hospitalization, functional status is strongly associated with out-of-hospital mortality and readmission rate. Future trials could provide evidence if improvement of functional status could prevent adverse outcomes in the postoperative setting
