    To determine the binding-site of a combinatorially-selected peptide possessing a fluoroprobe, a novel cysteine reactive small photo-crosslinker that can be excited by a conventional long-wavelength ultraviolet handlamp (365 nm) was synthesized via Suzuki coupling with three steps. The crosslinker is rationally designed, not only as a bioisostere of the fluoroprobe, but as a caged-fluorophore, and the photo-crosslinked target protein became fluorescent with a large Stokes-shift. By introducing the crosslinker to a designated sulfhydryl (SH) group of a combinatorially-selected peptide, the protein-binding site of the targeted peptide was deduced by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE)/fluorescence imaging followed by matrix-assisted laser desorption ionization-time of flight tandem mass spectrometry (MALDI-TOF-MS/MS) analysis

    Development of Continuous Radon Bronchial Dosimeter

    The deposition of radon decay products in each respiratory region can increase the lung cancer risk. Therefore, it is necessary to calculate the deposition of radon decay products in respiratory regions. Nowadays, the deposition in each respiratory region is able to be imitated using wire screens in many studies. Those studies employed grab sampling, that is, sampling for short period less than several minutes. In natural environment, because the concentration of radon decay products changes with time, it is necessary to measure continuously. In this study continuous dosimeter (CD) that imitates deposition of radon decay products in respiratory regions by using wire screens was developed for the purpose of estimation of the deposition in respiratory regions in various environments. The ratio of activities deposited in the simulated N and TB regions was calculated from results of measurement by CD-R, CD-N and CD-TB continuously, so that after aerosols were injected into the radon chamber, the ratio of N and TB depositions decreased, and, in addition, N / (N+TB) deposition decreased. These results indicated that CD-R, CD-N and CD-TB could measure the change of deposition ratio with time

    Continuous measurements of bronchial exposure induced by radon decay products during inharation

    The deposition of radon decay products is not equal in each of the respiratory regions and as the presence of radon has been linked with an increase in lung cancer risk, it is important to calculate the deposition of radon decay products in each of the respiratory regions. Recently, many studies on the deposition of radon in respiratory regions have been simulated using wire screens. The systems and equipment used in those studies are not suitable for field measurements as their dimensions are relatively massive, nor can they measure continuously. We developed a continuous bronchial dosimeter (CBD) which is suitable for field measurements. It was designed with specifications that allow it to be remain compact. The CBD simulates the deposition of radon decay products in the different respiratory regions by the use of a combination of wire screens. Deposition in the simulated regions of the lung can be continuously estimated in various environments. The ratio of activities deposited in a simulated nasal cavity (N) and tracheobronchial (TB) regions was calculated from the results of simultaneous measurements using CBD-R (reference), CBD-N (nasal), and CBD-TB (tracheobronchial) measurement units. After aerosols were injected into the radon chamber, the ratio of N and TB depositions decreased. This results indicate that the CBD gave a good response to changes in the environment. It was found that the ratio of N and TB deposition also varied with time in each actual environment

    Relationship between Dementia Severity and Behavioral and Psychological Symptoms of Dementia in Dementia with Lewy Bodies and Alzheimer's Disease Patients

    Background/Aims: Behavioral and psychological symptoms of dementia (BPSD) are common in the clinical manifestation of dementia. Although most patients with dementia exhibit some BPSD during the course of the illness, the association of BPSD with the stage of dementia remains unclear. It was the aim of this study to evaluate the impact of severity of dementia on the expression of BPSD in patients with dementia with Lewy bodies (DLB) and Alzheimer's disease (AD). Methods: Ninety-seven patients with DLB and 393 patients with AD were recruited from 8 dementia clinics across Japan. BPSD were assessed by the Neuropsychiatric Inventory (NPI). A relationship between BPSD and dementia stage classified by the Clinical Dementia Rating (CDR) in each type of dementia was assessed. Results: No significant difference was seen in NPI total score across CDR staging in the DLB group. On the other hand, the NPI total score significantly increased with dementia stage in the AD group. Conclusion: The relationship of dementia stage with the expression of BPSD was different according to the type of dementia. BPSD and dementia stage were correlated in AD subjects, in whom psychiatric symptoms increase as the disease progresses, but not in DLB subjects
