    Intestinal DNA concentration and protein synthesis in response to low quality diets in two strains of Leghorn layers

    Performance, protein synthesis and mucosal DNA in small intestine of Leghorn hens may be affected by low quality feedstuff. An experiment was conducted in completely randomized design (CRD) in 2 × 2 factorial arrangement. Main factors included diets containing 20 and 40 % barley and black and blue strains of leghorn. Layer performance was recorded weekly up to eight weeks. Egg production (%), egg mass and egg number were significantly higher in black strain leghorn on 20% barley diet than blue strain with 40% barley diet (P<0.05). Treatments blue × 40% barley and black × 20% barley showed lowest and highest egg production percentage, egg mass and egg number, respectively (P<0.05). The lowest protein synthesis in small intestine was shown in blue × 40% barley treatment group. Small intestine mucosal DNA quantity in blue strain was significantly higher than that in black strain (P<0.05). In contrast, protein synthesis in blue strain was lower than that in black strain (P<0.05). It was concluded that lower performance in blue × 40% treatment group could be related to lower protein synthesis in small intestine in blue strain, and this may be due to the high barley (40%) contained in diet.Key words: Performance, protein synthesis, mucosal DNA, small intestine

    Abstract An experiment was conducted to investigate the effects of hydroalcoholic extract of Withania coagulans (WC) fruit and 1.25-dihydroxycholecalciferol (1.25-(OH)2 D3) on bone mineralization, mechanical and histological properties of male broiler chickens at 21 and 42 d of age. A total of six hundred male day-old Ross 308 broiler chickens were randomly distributed according to a completely randomized experimental design in a 2×3×2 factorial arrangement with 12 treatments of five replicates of 10 birds each. Treatments consisted of two basal diets (positive control with adequate Ca level and negative control with 30% less Ca), three levels of WC (0, 100, or 200 mg/kg diet), and two levels of 1.25-(OH)2 D3 (0 or 0.5 µg/kg diet). Birds were housed in floor pens. The diets were fed ad libitum from one to 42 days of age. On day 21 and 42, one bird per replicate was sacrificed and its tibiae were removed. Both Ca and P retention increased when dietary Ca level was reduced (p<0.001). The addition of 200 mg WC/kg to positive control diet increased Ca retention (p<0.01). Except for tibia diameter, no significant main effects of experimental treatments were observed on tibia physical characteristics or on bone mineralization. The diet with 30% Ca reduction decreased tibia diameter at 42 days of age (p<0.05). The dietary addition of 1.25-(OH)2 D3 increased tibia fracture energy, width of tibia mineralized zone, and serum Ca at 42 days of age (p<0.05). At 21 days of age, supplementation of 100 mg WC/kg increased cortical thickness (p<0.05). At 42 days of age, supplementation of 100 mg WC/kg increased tibia shear force (p<0.05) and fracture energy (p<0.01). The results of this experiment showed that supplementation of 100 mg/kg hydroalcoholic extract of WC fruit increased tibia cortical thickness, shear force, and fracture energy

    Tablet Splitting of a Narrow Therapeutic Index Drug: A Case with Levothyroxine Sodium

    Levothyroxine is a narrow therapeutic index, and to avoid adverse effect associated with under or excessive dosage, the dose response is carefully titrated. The tablets are marketed with a score providing an option to split. However, there are no systematic studies evaluating the effect of splitting on dose accuracy, and current study was undertaken to evaluate effects of splitting and potential causes for uniformity failures by measuring assay and content uniformity in whole and split tablets. Stability was evaluated by assaying drug for a period of 8 weeks. Effect of formulation factors on splittability was evaluated by a systematic investigation of formulation factors by preparing levothyroxine tablets in house by varying the type of excipients (binder, diluent, disintegrant, glidant) or by varying the processing factors (granulating liquid, mixing type, compression pressure). The tablets were analyzed using novel analytical tool such as near infrared chemical imaging to visualize the distribution of levothyroxine. Assay was not significantly different for whole versus split tablets irrespective of method of splitting (hand or splitter), and splitting also had no measurable impact on the stability. Split tablets either by hand or splitter showed higher rate of content uniformity failures as compared to whole tablets. Tablet splitter produced more fragmentation and, hence, more content uniformity and friability failures. Chemical imaging data revealed that the distribution of levothyroxine was heterogeneous and was dependent on type of binder and the process used in the manufacture of tablets. Splitting such tablets could prove detrimental if sub- or super-potency becomes an issue
