    Most of the VP1 Unique Region of B19 Parvovirus Is on the Capsid Surface

    AbstractB19 parvovirus is pathogenic in man and a vaccine is desirable. In convalescence after acute infection, the dominant humoral immune response is directed to the minor capsid protein called VP1, which differs from the major capsid protein by an additional NH2-terminal 227 amino acids. We have previously shown that this unique region contains multiple linear neutralizing epitopes. We produced seven recombinant B19 capsids that contained progressively truncated VP1 unique region sequences, each fused to a Flag peptide (AspTyrLysAspAspAspAspLys) at the NH2-terminus. Capsids containing normal VP2 and truncated Flag-VP1 proteins and, in some cases, only truncated Flag-VP1 chimeric proteins, were analyzed by ELISA, affinity chromatography, and electron microscopy using anti-Flag monoclonal antibody. All regions examined showed binding to anti-Flag antibody in multiple assays, indicating that most of the VP1 unique region is external to the capsid and accessible to antibody binding. These results have implications for the design of a B19 parvovirus vaccine and the use of empty capsids for presentation of heterologous protein antigens

    On-The-Fly Observing System of the Nobeyama 45-m and ASTE 10-m Telescopes

    We have developed spectral line On-The-Fly (OTF) observing mode for the Nobeyama Radio Observatory 45-m and the Atacama Submillimeter Telescope Experiment 10-m telescopes. Sets of digital autocorrelation spectrometers are available for OTF with heterodyne receivers mounted on the telescopes, including the focal-plane 5 x 5 array receiver, BEARS, on the 45-m. During OTF observations, the antenna is continuously driven to cover the mapped region rapidly, resulting in high observing efficiency and accuracy. Pointing of the antenna and readouts from the spectrometer are recorded as fast as 0.1 second. In this paper we report improvements made on software and instruments, requirements and optimization of observing parameters, data reduction process, and verification of the system. It is confirmed that, using optimal parameters, the OTF is about twice as efficient as conventional position-switch observing method.Comment: 11 pages, 13 figures, accepted for publication in PAS

    The Hepcidin-25/Ferritin Ratio Is Increased in University Rugby Players with Lower Fat Mass

    Hepcidin-25 is suggested as a surrogate iron status marker in athletes who show exercise-induced anemia; however, the implications of hepcidin concentration in this population remain poorly understood. This study aimed to investigate the relationship between hepcidin and body fat levels in rugby football players. We included 40 male university rugby football players (RUG) and 40 non-athlete controls. All participants underwent an anthropometric analysis and blood testing that included both hepcidin-25 and ferritin levels. The hepcidin-25 level was slightly (11.6%, p = 0.50) higher, and the ferritin level was significantly (35.9%, p < 0.05) lower, in the RUG group than in controls. The hepcidin-25 to-ferritin ratio was significantly higher (62.5%, p < 0.05) in the RUG group. While significant U-shaped correlations were observed between the body fat and ferritin levels in both groups, the correlations between the hepcidin levels and fat mass index were significantly higher in the RUG group (RUG: r = 0.79, controls: r = 0.45). Notably, the RUG with the lower fat mass index group had a higher hepcidin-25 level, lower ferritin level, and then significantly higher hepcidin-25/ferritin ratio. The hepcidin-25/ferritin ratio may serve as a biomarker for iron status in RUG, especially RUG with lower fat mass

    Influence of Intrauterine Inflammation, Delivery, and Postnatal Feeding on the Temporal Changes of Serum Alpha 1 Acid Glycoprotein Levels in Extremely-Low-Birth-Weight Infants

    Infection remains the primary cause of death in extremely-low-birth-weight infants (ELBWIs). Alpha 1 acid glycoprotein (α1AG), an acute-phase protein, has been shown to be elevated in sporadic cases of septic ELBWIs prior to abnormal clinical signs. To delineate the roles of inflammation, delivery, and feeding in postnatal α1AG changes in ELBWIs, 75 ELBWIs of 26.5 ± 2.2 weeks of gestation born between May 2011 and August 2017 were retrospectively studied. The dependence of α1AG levels obtained on days 0–5 on the clinical variables was examined by incorporating interactions with age, followed by estimations of regression coefficients between clinical variables and α1AG levels at the early and late postnatal ages, defined by their standard deviation. Chorioamnionitis (p p = 0.045), vaginal delivery (p = 0.025), enteral feeding (p = 0.022), and probiotics (p = 0.005) were associated with early α1AG elevations. Hypertensive disorder of pregnancy (p p = 0.001) were associated with late α1AG elevation; premature rupture of membranes (p p = 0.021), body weight z-scores (p p = 0.045) were associated with late α1AG reduction. Postnatal α1AG changes in ELBWIs were associated with variables representative of age, growth, delivery, inflammation, and enteral feeding, potentially reflecting the process of sensitization to extrinsic microbes in utero, at birth, and thereafter

    A Lubricant-Sandwiched Coating with Long-Term Stable Anticorrosion Performance

    Lubricant-infused surface(s) (LIS) bioinspired by the <i>Nepenthes</i> pitcher plant are receiving enormous attention owing to their excellent hydrophobicity as well as their self-healing ability. Thus, they have been applied as anticorrosion coatings. However, the loss of lubricant mediated by vapor or other liquids deteriorates their functions. Herein, we introduce a lubricant-inserted (sandwiched) microporous triple-layered surface (LIMITS) that prevents the sudden loss of lubricant. The sandwiched lubricant gradually self-secretes toward the surface, resulting in long-term stability even under water. The LIMITS prevented the corrosion of the Fe plate for at least 45 days, which is much superior to a conventional LIS coating. This work opens an avenue for the application of slippery coating materials that are stable under water and will also promote the development of anticorrosion coating in various industries

    Neutron Diagnostics in the Large Helical Device

    The deuterium operation of the large helical device (LHD) began in March 7, 2017, after long-term preparation and commissioning of apparatuses necessary for execution of the deuterium experiment. A comprehensive set of neutron diagnostics was developed and installed onto LHD through numerous efforts in preparation. Neutron diagnostics play an essential role in both neutron yield management for the radiation safety and extension of energetic-particle physics study in LHD. Neutron flux monitor (NFM) characterized by fast-response and wide dynamic range capabilities is successfully working. Total neutron emission rate reached 3.3 × 10 15 (n/s) in the first deuterium campaign of LHD. The highest neutron emission rate was recorded in inward shifted configuration. Neutron yield evaluated by neutron activation system agrees with neutron yield measured with NFM. Performance of vertical neutron camera was demonstrated. Neutron emission profile was inwardly shifted in the inwardly shifted configuration, whereas it was outwardly shifted in the outwardly configuration. Secondary deuterium-tritium neutrons produced by triton burnup in LHD deuterium plasmas were detected for the first time in stellarator/heliotron devices in the world.
