    Effects of the tat and nef gene products of human immunodeficiency virus type 1 (HIV-1) on transcription controlled by the HIV-1 long terminal repeat and on cell growth in macrophages

    The RAW264 murine macrophage cell line was used as a model to examine the role of the tat and nef gene products in the transcription regulation of the human immunodeficiency virus type 1 (HIV-1) long terminal repeat (LTR) in macrophages. Contrary to claims that the activity of the HIV-1 LTR responds poorly in rodent cells to trans activation by the viral tat gene product, cotransfection of RAW264 cells with a tat expression plasmid in transient transfection assays caused a > 20-fold increase in reporter gene expression that was inhibited by mutations in the TAR region. RAW264 cells stably transfected with the tat plasmid displayed similarly elevated HIV-1 LTR-driven reporter gene activity. By contrast to previous reports indicating a negative role for nef in HIV transcription, cotransfection of RAW264 cells with a nef expression plasmid trans activated the HIV-1 LTR driving either a chloramphenicol acetyltransferase or a luciferase reporter gene. The action of nef was specific to the LTR, as expression of nef had no effect on the activity of the simian virus 40, c-fms, urokinase plasminogen activator, or type 5 acid phosphatase promoter. trans-activating activity was also manifested by a frameshift mutant expressing only the first 35 amino acids of the protein. The effects of nef were multiplicative with those of tat gene product and occurred even in the presence of bacterial lipopolysaccharide, which itself activated LTR-directed transcription. Examination of the effects of selected mutations in the LTR revealed that neither the kappa B sites in the direct repeat enhancer nor the TAR region was required as a cis-acting element in nef action. The action of nef was not species restricted; it was able to trans activate in the human monocyte-like cell line Mono Mac 6. The presence of a nef expression cassette in a neomycin phosphotransferase gene expression plasmid greatly reduced the number of G418-resistant colonies generated in stable transfection of RAW264 cells, and many of the colonies that were formed exhibited very slow growth. The frameshift mutant was also active in reducing colony generation. Given the absence of any effect of the frameshift mutation on nef function, its actions on macrophage growth and HIV transcription are discussed in terms of the role of the N-terminal 30 amino acids and of stable secondary structures in the mRNA

    Multicolor Dye-based Flow Structure Visualization for seal-whisker geometry characterized by computer vision

    Pinniped vibrissae possess a unique and complex three-dimensional topography, which has beneficial fluid flow characteristics such as substantial reductions in drag, lift, and vortex induced vibration. To understand and leverage these effects, the downstream vortex dynamics must be studied. Dye visualization is a traditional qualitative method of capturing these downstream effects, specifically in comparative biological investigations where complex equipment can be prohibitive. High-fidelity numerical simulations or experimental particle image velocimetry (PIV) are commonplace for quantitative high-resolution flow measurements, but are computationally expensive, require costly equipment, and can have limited measurement windows. This study establishes a method for extracting quantitative data from standard dye visualization experiments on seal whisker geometries by leveraging novel but intuitive computer vision techniques, which maintain simplicity and an advantageous large experimental viewing window while automating the extraction of vortex frequency, position, and advection. Results are compared to direct numerical simulation (DNS) data for comparable geometries. Power spectra and Strouhal numbers show consistent behavior between methods for a Reynolds number of 500, with minima at the canonical geometry wavelength of 3.43 and a peak frequency of 0.2 for a Reynolds number of 250. The vortex tracking reveals a clear increase in velocity from roll-up to 3.5 whisker diameters downstream, with a strong overlap with the DNS data but shows steady results beyond the limited DNS window. This investigation provides insight into a valuable bio-inspired engineering model while advancing an analytical methodology that can readily be applied to a broad range of comparative biological studies

    Water quality changes during the first meter of managed aquifer recharge

    The capacity of an artificial recharge field to alter organic matter and the bacterial flora of surface water was assessed by following changes in bacterial communities and composition of natural organic matter (NOM) over the first meter of infiltration depth. The sampling strategy applied in this study ensured that water samples consisted only of infiltrated water, excluding natural groundwater. Water was sampled at 50 and 100 cm below the surface of an infiltration basin divided into two halves; one side was dried and frozen and one was infiltrating water during the winter period prior to the sampling period. Bacterial cell counts, proportions of intact cells and community fingerprints were determined by flow cytometry, and NOM was characterized using total organic carbon (TOC), UV254 nm-absorbance (UVA) and fluorescence spectroscopy. Around 40% of the NOM was removed after only 50 cm. Protein-like components were reduced to a larger extent (45-50%) than the humic-like components (25%), suggesting removal of mostly biodegradable fractions of NOM. After only 50 cm of infiltration, about 99% total cell count (TCC) was removed. The flow cytometric data revealed that the bacterial communities collected after infiltration from the basin area that had been dried and frozen were more similar to those in the raw water. This suggests that drying and freezing the basin negatively impacted its treatment capacity. The results from this study highlight the importance of a well-developed biofilm and unsaturated zone for artificial recharge

    Does Gender Impact Intensity of Care Provided to Older Medical Intensive Care Unit Patients?

    Introduction. Women receive less aggressive critical care than men based on prior studies. No documented studies evaluate whether men and women are treated equally in the medical intensive care unit (MICU). The Therapeutic Intervention Scoring System-28 (TISS-28) has been used to examine gender differences in mixed ICU studies. However, it has not been used to evaluate equivalence of care in older MICU patients. We hypothesize that given nonsignificant, baseline health differences between genders at MICU admission, the level of care provided would be equivalent. Methods. Prospective cohort of 309 patients ≥60 years old in the MICU of an urban university teaching hospital. Explanatory variables were demographic data and baseline measures. Primary outcomes were TISS-28 scores and MICU interventions. We compare TISS-28 scores by gender using a statistical test of equivalence. Results. Women were older and had more chronic respiratory failure at MICU admission. Using equivalence limits of ±15% on gender-based scores of TISS-28, MICU interventions were equivalent. Supplementary analysis showed no statistically significant association between gender and mortality. Conclusions. In contrast with other reports from the cardiac critical care literature, as measured by the TISS-28, gender-based care delivered to older MICU patients in this cohort was equivalent

    Bringing trauma home : reflections on interviewing survivors of trauma while working from home

    In response to the COVID-19 pandemic, working from home became the new normal for many professionals. While this was beneficial in managing the rapidly spreading virus, it had varying impacts on the mental health of those previously not accustomed to remote work. This paper provides a critical reflection of the researcher’s experience of conducting interviews with survivors of trauma while working from home. The research aimed to understand the experiences of significant others supporting patients with severe burn injury in the Intensive Care Unit (ICU). As an experienced ICU nurse, the researcher has well developed personal coping strategies for dealing with complex trauma and in working with significant others of patients with severe burn injury in hospital settings. Due to the pandemic, data collection moved from face-to-face in the hospital, as originally intended, to phone or videoconference interviews. 17 participants were recruited, with all participants given the option of videoconference (n = 3) or telephone interviews (n = 14). Interviews had an average length of 55 minutes. This paper discusses the strategies adopted to cope with the sharing of significant others’ experiences of trauma while in the home environment. Careful consideration was needed for the researcher, the participants and those within the homes of both researcher and participant, in terms of psychological safety, ethical considerations and rapport building

    De novo post-diagnosis statin use, breast cancer-specific and overall mortality in women with stage I-III breast cancer.

    BACKGROUND: Prior evidence suggests a role for statins in the management of cancer. However, the benefit of statin use in the adjuvant setting remains uncertain. This study investigates associations between statin use initiated after a breast cancer diagnosis and mortality. METHODS: Women with stage I-III breast cancer were identified from the National Cancer Registry of Ireland (N=4243). Post-diagnostic statin initiators were identified from pharmacy claims data (N=837). Multivariate models were used to estimate hazard ratios (HRs) and 95% confidence intervals (CIs) for associations between de novo statin use and mortality. RESULTS: The median duration of statin use was 6.7 years. No association was found between post-diagnostic statin use and breast cancer-specific (HR 0.88, 95% CI 0.66, 1.17) or all-cause mortality (HR 1.00, 95% CI 0.82, 1.21). CONCLUSIONS: The results from our study suggest that initiating statin use after a diagnosis of stage I-III breast cancer is not associated with a reduction in breast cancer-specific mortality

    Lack of a Y-Chromosomal Complement in the Majority of Gestational Trophoblastic Neoplasms

    Gestational trophoblastic neoplasms (GTNs) are a rare group of neoplastic diseases composed of choriocarcinomas, placental site trophoblastic tumors (PSTTs) and epithelioid trophoblastic tumors (ETTs). Since these tumors are derivatives of fetal trophoblastic tissue, approximately 50% of GTN cases are expected to originate from a male conceptus and carry a Y-chromosomal complement according to a balanced sex ratio. To investigate this hypothesis, we carried out a comprehensive analysis by genotyping a relatively large sample size of 51 GTN cases using three independent sex chromosome genetic markers; Amelogenin, Protein Kinase and Zinc Finger have X and Y homologues that are distinguishable by their PCR product size. We found that all cases contained the X-chromosomal complement while only five (10%) of 51 tumors harbored the Y-chromosomal complement. Specifically, Y-chromosomal signals were detected in one (5%) of 19 choriocarcinomas, one (7%) of 15 PSTTs and three (18%) of 17 ETTs. The histopathological features of those with a Y-chromosome were similar to those without. Our results demonstrate the presence of a Y-chromosomal complement in GTNs, albeit a low 10% of cases. This shortfall of Y-chromosomal complements in GTNs may reinforce the notion that the majority of GTNs are derived from previous molar gestations
