    Preparative Synthesis of dTDP-L-Rhamnose Through Combined Enzymatic Pathways

    dTDP-L-rhamnose, an important precursor of O-antigen, was prepared on a large scale from dTMP by executing an one-pot reaction in which six enzymes are involved. Two enzymes, dTDP-4-keto-6-deoxy-D-glucose 3,5-epimerase and dTDP-4-keto-rhamnose reductase, responsible for the conversion of dTDP-4-keto-6-deoxy- D-glucose to dTDP-L-rhamnose, were isolated from their putative sequences in the genome of Mesorhizobium loti, functionally expressed in Escherichia coli, and their enzymatic activities were identified. The two enzymes were combined with an enzymatic process for dTDP-4- keto-6-deoxy-D-glucose involving TMP kinase, acetate kinase, dTDP-glucose synthase, and dTDP-glucose 4,6- dehydratase, which allowed us to achieve a preparative scale synthesis of dTDP-L-rhamnose using dTMP and glucose-1-phosphate as starting materials. About 82% yield of dTDP-L-rhamnose was obtained based on initial dTMP concentration at 20 mM dTMP, 1 mM ATP, 10 mM NADH, 60 mM acetyl phosphate, and 80 mM glucose-1- phosphate. From the reaction with 20 ml volume, approximately 180 mg of dTDP-L-rhamnose was obtained in an overall yield of 60% after two-step purification, that is, anion exchange chromatography and gel filtration for desalting.     Dye-sensitized solar cells (DSSCs) have been extensively evolved for the past two decades in order to improve their cell performance. From the commercialization point of view, the overall solar to electrical energy conversion efficiency should compete with other solar cells. But, due to structural restrictions of DSSC using the liquid electrolyte and a space requirement between two electrodes, the direct tandem construction of DSSCs by stacking of repeating units is highly limited. In this feature article, important research trials to overcome these barriers and a recent research trend to improve the light harvesting strategies mainly panchromatic engineering, various tandem approaches such as parallel tandem, series tandem, p-n tandem etc., have been briefly reviewed.close271

    Monitoring transfected cells without selection agents by using the dual-cassette expression EGFP vectors

    Conventional methods of selecting gene transfected cells by toxic agents may yield ambiguous results. It is difficult to determine whether cell death is due to selection agents or gene transfection, owing to the substantial overlap of the time-courses for both effects. Therefore, to determine transfection-induced cell toxicity, the mammalian expression vector pEGFP-N1 (CLONTECH Lab., Palo Alto, CA, USA) has been modified to the dual-cassette expression vectors named pEGFP-Ks by the relocation of its EGFP expression cassette. We have precisely monitored the cells transfected with this vector on our custom culture dishes, thereby bypassing the need for selection agent or fluorescent cell sorting. This is a useful method to screen genes encoding potential toxic or useful proteins without performing undesirable selection agent and also can be used to monitor the transfected cells for various purposes, either the inhibition or proliferation of mammalian cells for applications in biotechnology

    Measurement of the Background Activities of a 100Mo-enriched powder sample for AMoRE crystal material using a single high purity germanium detector

    The Advanced Molybdenum-based Rare process Experiment (AMoRE) searches for neutrino-less double-beta (0{\nu}\b{eta}\b{eta}) decay of 100Mo in enriched molybdate crystals. The AMoRE crystals must have low levels of radioactive contamination to achieve low background signals with energies near the Q-value of the 100Mo 0{\nu}\b{eta}\b{eta} decay. To produce low-activity crystals, radioactive contaminants in the raw materials used to form the crystals must be controlled and quantified. 100EnrMoO3 powder, which is enriched in the 100Mo isotope, is of particular interest as it is the source of 100Mo in the crystals. A high-purity germanium detector having 100% relative efficiency, named CC1, is being operated in the Yangyang underground laboratory. Using CC1, we collected a gamma spectrum from a 1.6-kg 100EnrMoO3 powder sample enriched to 96.4% in 100Mo. Activities were analyzed for the isotopes 228Ac, 228Th, 226Ra, and 40K. They are long-lived naturally occurring isotopes that can produce background signals in the region of interest for AMoRE. Activities of both 228Ac and 228Th were < 1.0 mBq/kg at 90% confidence level (C.L.). The activity of 226Ra was measured to be 5.1 \pm 0.4 (stat) \pm 2.2 (syst) mBq/kg.     Innovative Wireless Power Receiver for Inductive Coupling and Magnetic Resonance Applications

    This chapter presents a wireless power receiver for inductive coupling and magnetic resonance applications. The active rectifier with shared delay-locked loop (DLL) is proposed to achieve the high efficiency for different operation frequencies. In the DC–DC converter, the phase-locked loop is adopted for the constant switching frequency in the process, voltage, and temperature variation to solve the efficiency reduction problem, which results in the heat problem. An automatic mode switching between pulse width modulation and pulse frequency modulation is also adopted for the high efficiency over the wide output power. This chip is implemented using 0.18 μm BCD technology with an active area of 5.0 mm × 3.5 mm. The maximum efficiency of the active rectifier is 92%, and the maximum efficiency of the DC–DC converter is 92% when the load current is 700 mA
