    Pharmacological effects of active saponins from Panax ginseng Meyer

    Purpose: To investigate the pharmacological effects of the active saponins isolated from Panax ginseng Meyer (P. ginseng) via extraction, heat treatment, and enzyme conversion. Methods: The effects of active saponins on rat blood were determined using a multichannel analyzer. The population doubling time (PDT) of mesenchymal stem cells (MSCs) and human-derived leukocyte cancer cells (A549) was determined by cell counting. b-galactosidase was measured in human toothderived stem cells (HTS) using a β-galactosidase ELISA kit. Results: Intraperitoneal administration of active saponins resulted in 30.09 % increase in red blood cell count and 55.55 % decrease in blood triglyceride concentrations. The stimulatory effect of active saponins (10 ng/mL) on cellular differentiation was determined based on PDT of MSCs, which decreased by 33.82 % compared to control. A 22.29 % increase in PDT of A549 cells demonstrated the suppressive effects of active saponins on cancer cell growth. Active saponins (10 ng/mL) also decreased intracellular β-galactosidase concentration by 20.42 % in HTS cells. Conclusion: Administration of active saponins to rats extends the lifespan, promotes differentiation in MSCs, suppresses A549 cell differentiation, and reduces TG and b-galactosidase associated with aging in HTS. Thus, active saponins have potentially beneficial effects in humans

    Comparative analysis of FBS containing media and serum free chemically defined media, CellCor for adipose derived stem cells production

    Background: As a result of the aging society, the average OECD life expectancy has grown to about 80 years, yet the average health life still remains at only 65 years, leaving more than 15 years of life in an uncertain health state. Regenerative medicine is a new concept of medicine that combines cells and biomaterials to restore the functions of aged or damaged tissues or organs. It is also a good treatment for chronic diseases and incurable diseases, receiving attention as a new paradigm for treating diseases. Problems: As the market for regenerative medicine grows, mass production of consistent quality cells is required. Media is the most important thing in mass production of consistent quality cells. However, the fetal bovine serum (FBS) containing media that is currently wide used has many problems, such as unidentified viral infection, immunogenicity, lot variations, unstable supply, and ethical issues. To solve these problems and make rapid progress in regenerative medicine, a high-performance serum free chemically defined media (CDM) is needed. Solution: CellCor is a serum free CDM that provides excellent performance, safety, economy and consistency in stem cell production. CellCor allows higher-speed cell production rate than current FBS containing culture media (Figure 1). Compared to the FBS containing media, CellCor is able to maintain stem cell markers, higher population homogeneity, genetic stability, and excellent differentiation potency even at later passage.     Reflection phase microscopy using spatio-temporal coherence of light

    Many disease states are associated with cellular biomechanical changes as markers. Label-free phase microscopes are used to quantify thermally driven interface fluctuations, which allow the deduction of important cellular rheological properties. Here, the spatio-temporal coherence of light was used to implement a high-speed reflection phase microscope with superior depth selectivity and higher phase sensitivity. Nanometric scale motion of cytoplasmic structures can be visualized with fine details and three-dimensional resolution. Specifically, the spontaneous fluctuation occurring on the nuclear membrane of a living cell was observed at video rate. By converting the reflection phase into displacement, the sensitivity in quantifying nuclear membrane fluctuation was found to be about one nanometer. A reflection phase microscope can potentially elucidate biomechanical mechanisms of pathological and physiological processes.Korea Health Industry Development Institute.     High-fidelity 3D Human Digitization from Single 2K Resolution Images

    High-quality 3D human body reconstruction requires high-fidelity and large-scale training data and appropriate network design that effectively exploits the high-resolution input images. To tackle these problems, we propose a simple yet effective 3D human digitization method called 2K2K, which constructs a large-scale 2K human dataset and infers 3D human models from 2K resolution images. The proposed method separately recovers the global shape of a human and its details. The low-resolution depth network predicts the global structure from a low-resolution image, and the part-wise image-to-normal network predicts the details of the 3D human body structure. The high-resolution depth network merges the global 3D shape and the detailed structures to infer the high-resolution front and back side depth maps. Finally, an off-the-shelf mesh generator reconstructs the full 3D human model, which are available at https://github.com/SangHunHan92/2K2K. In addition, we also provide 2,050 3D human models, including texture maps, 3D joints, and SMPL parameters for research purposes.     Impact of Chronic Cough on Health-Related Quality of Life in the Korean Adult General Population: The Korean National Health and Nutrition Examination Survey 2010–2016

    PurposeChronic cough is a prevalent condition in the community and may pose considerable impairment to quality of life (QoL). However, its disease burden remains largely undefined in the general population. The present study investigated the relationship between chronic cough and health-related QoL in a Korean nationwide population database, with an emphasis on clinical conditions which may confound the impact of cough.MethodsThis study analyzed cross-sectional datasets of adults (aged ≥ 40 years) in the Korean National Health and Nutrition Examination Survey 2010–2016. Health-related QoL was assessed using the 3-level EuroQoL 5-dimension component (EQ-5D-3L) index score. The presence of chronic cough and other conditions were defined using structured questionnaires.ResultsThe prevalence of chronic cough was 3.48% ± 0.17% among adults aged ≥ 40 years. The overall EQ-5D-3L index score was significantly lower in subjects with than without chronic cough (0.79 ± 0.01 vs. 0.86 ± 0.00, P < 0.001). In subgroup analyses by age and sex, chronic cough had a notably large impact on QoL in women aged ≥ 65 years (vs. those without chronic cough: 0.55 ± 0.04 vs. 0.70 ± 0.01, P < 0.001), although the mean difference in the scores exceeded the minimally important difference score of 0.05 in all subgroups. In multivariate analyses, chronic cough was significantly associated with QoL, independent of confounders including depression, arthritis, asthma, and chronic obstructive pulmonary disease. In dimension analyses, chronic cough was more associated with anxiety/depression, pain/discomfort, and usual activities than with self-care or mobility in the EQ-5D.ConclusionsThe present study demonstrated significant associations between chronic cough and health-related QoL in a nationwide large general adult population aged ≥ 40 years, which were independent of clinical confounders. The impact of chronic cough was greater in women aged ≥ 65 years. These findings indicate a considerable burden of chronic cough in the general population and warrant further investigations to assess the disease burden of chronic cough in a global scale

    Pig-to-Nonhuman Primate (NHP) Naked Islet Xenotransplantation

    Islet transplantation is an established therapy for selected type 1 diabetes (T1D) patients with severe hypoglycemic unawareness and glycemic liability despite of insulin treatment. However, the donor organ is limited. Porcine islets are the best alternative source to overcome this limitation, and pig-to-nonhuman primate (NHP) naked islet xenotransplantation studies are being performed worldwide. Several studies including our own have presented successful proof-of-concept results based on immunosuppression regimen including the anti-CD154 monoclonal antibody. Particularly, long-term control of diabetes by adult porcine islet transplantation has been demonstrated in five consecutive monkeys, and the longest survival was ~1000 days after transplantation. Currently, pig-to-NHP islet xenotransplantation based on clinically applicable immunosuppression regimen is being pursued. In this chapter, we will describe all the procedures of pig-to-NHP naked islet xenotransplantation: (1) the porcine islet isolation from designated pathogen-free (DPF) miniature pigs, (2) diabetes induction in monkeys, (3) transplantation procedure via the portal vein, (4) immune monitoring comprising humoral and cellular immunity after porcine islet transplantation, and finally (5) liver biopsy and subsequent immunohistochemical procedure in detail

    Delayed Treatment of Zygomatic Tetrapod Fracture

    Since maxillofacial injury is frequently accompanied by other diseases, its evaluation and treatment are open delayed. When the evaluation is delayed, the surgical treatment can be difficult or impossible. A 21-yr-old man presented with right facial swelling and deformity after injury. We planned immediate surgical repair for his right tetrapod fracture, but the operation was delayed for two months due to severe hyperthyroidism. During the operation, we reducted and fixed the deviated bone after refracture of the zygomatic arch with an osteotome to achieve mobility. The facial deformity and difficulty in mouth opening were improved after the operation. Even in the presence of accompanying fractures, early evaluation and proper management can prevent complications and achieve acceptable cosmetic outcomes in maxillofacial trauma patients. In patients with malunion of fracture sites, fixation after refracture using an osteotome can be a good treatment option for obtaining good mobility
