    Action mechanism and structural requirements of the antimicrobial peptides, gaegurins

    AbstractGaegurins (GGNs) are a family of cationic, α-helical, antimicrobial peptides that were isolated from a Korean frog, Glandirana emeljanovi (formerly classified as Rana rugosa) and represent one of the structurally well-characterized groups. Among six gaegurins, gaegurin 4 (renamed herein esculentin-2EM), gaegurin 5 (brevinin-1EMa), and gaegurin 6 (brevinin-1EMb) have been investigated comprehensively in terms of structure–activity relationships. In this paper, we first suggest renaming of gaegurins according to a recently raised rule of systematic nomenclature. Then, the current understanding of gaegurins is reviewed by summarizing their structure–activity relationships. In particular competing arguments on gaegurins are synthetically inspected. Finally their action mechanism and structural requirements will be discussed

    What you don't see can hurt you : awareness cues to profile indirect competitors

    The awareness-motivation-capability (AMC) framework instructs firms to be aware of rivals, yet it offers limited guidance on how to profile those who are not yet rivals but stand to become so. Because rivals are embedded in dyads, triads, tetrads, etc., amultilevel view can unearth awareness cues that specify a hostility profile and make the awareness construct prescient. Studying thousands of competitive encounters over 10 years, we show that, at the firm and dyad levels, diversification and asymmetric pressure (differential exposure to competitive pressure) are reliable cues predicting competitive encounters. At the network level, convergence drives triadic encounters (competition with a rival's rival), and the degree of separation among indirect competitors defines the outer bounds of the hostility profile. Specifically, direct rivals and second- and third-degree indirect competitors merit awareness-more distal players do not. Together, the awareness cues and hostility profile delineate the conceptual bound within which awareness is prescient and beyond which it is misplaced. Challenging several assumptions, our study shows that an arena view assists in predicting cross-industry competition; applying firm, dyad, and network levels of analysis is advisable to better foresee competition; and indirect competitors are "profilable," allowing us to "see" rivals even before they strike

    Validation of the Dimensions of Anger Reactions Scale (the DAR-5) in non-clinical South Korean adults

    Background Posttraumatic anger is a commonly reported emotion among people who have experienced traumatic events. The current study aimed to demonstrate the reliability and validity of the South Korean version of the DAR-5 (DAR-5-K). The DAR-5 is a single scale with 5 items which measures posttraumatic anger. The DAR-5 is composed of five items that measure anger frequency, intensity, duration, aggression, and its interference with social relations. Methods Data were collected from 814 South Korean adults who had experienced traumatic events and participated in the study and analyzed via the combination of exploratory factor analysis (n = 405) and confirmatory factor analysis (n = 409). Results Results supported the one-factor structure, as reported in previous validation studies. The scale demonstrated robust internal reliability and concurrent validity with measures of posttraumatic stress disorder (PTSD) symptoms, depression, anxiety, and self-esteem. The DAR-5 cut-off score of 12 that was established in the original validation study successfully differentiated high from low scorers with regard to PTSD symptoms, depression, anxiety, and self-esteem. Conclusion The results confirm that the DAR-5-K is a brief and psychometrically robust measure of anger that can be used to examine South Korean adults who have experienced traumatic events

    Effects of Particle Size and Cement Replacement of LCD Glass Powder in Concrete

    The high quality liquid crystal display (LCD) processing waste glass (LPWG) generated from the manufacturing process of Korea’s LCD industries, having the world’s highest technological level and production, was finely ground into particles smaller than cement particles (higher fineness than OPC) to verify their applicability and performance as a replacement for cement. For a concrete mix having a W/B ratio of 0.44, cement was replaced with LPWG glass powder (LGP) at ratios of 5, 10, 15, and 20% (LGP12) and 5 and 10% (LGP5) according to the particle size to prepare test cylinder specimens, which were tested with respect to air contents, slump in fresh concrete, and compressive strength and splitting tensile strength of hardened concrete. The microstructure of the concrete specimens was analyzed through Scanning Electron Microscopy (SEM), Energy Dispersive X-ray (EDX), and a Mercury Intrusion Porosimetry (MIP). Replacement of cement with LGP for cement could effectively decrease the quantity of cement used due to the excellent performance of LGP. It may positively contribute to the sustainable development of the cement industry as well as waste recycling and environment conservation on a national scale

    Increased fucosyl glycoconjugate by Mycoplasma hyopneumoniae enhances adherences of Pasteurella multocida type A in the ciliated epithelial cells of the respiratory tract

    BACKGROUND: The objective of this study was to elucidate the pathogenic mechanisms of how Mycoplasma hyopneumoniae enhances secondary Pasteurella multocida type A infection which leads to porcine enzootic pneumonia in infected pigs. Sixteen pigs were experimentally infected with M. hyopneumoniae and then euthanized at 7, 14, 21 and 28 days post inoculation. In situ hybridization for M. hyopneumoniae DNA and Ulex europaeus agglutinin-I (UEA-I) lectin histochemistry for fucosyl glycoconjugate, was performed in serial lung sections to determine alteration of fucosyl glycoconjugate in M. hyopneumoniae-infected bronchial and bronchiolar epithelium. Bacterial overlay assay was performed to determine the affinity of P. multocida type A with L-fucose. RESULTS: The luminal surface of bronchial and bronchiolar epithelial cells that were stained with UEA-I always showed hybridization signals for M. hyopneumoniae but it was negative in the unaffected parts of the lung from M. hyopneumoniae-infected pigs and in lung from negative control pigs. Colocalization of M. hyopneumoniae and UEA-I was especially prominent in the luminal surface of bronchial and bronchiolar epithelial cells in serial section of lung. The mean number of M. hyopneumoniae-positive cells correlated with the mean number of UEA-I-positive cells in lungs from infected pigs throughout the experiment. All eight P. multocida type A isolates from naturally occurring enzootic pneumonia, bound strongly at levels of 2 μg and 5 μg of L-fucose. CONCLUSIONS: The results of the present study demonstrate that M. hyopneumoniae increases the L-fucose composition to enhance adherence of P. multocida type A to the bronchial and bronchiolar epithelial cells

    Synergistic effect of acetyl xylan esterase on xylanase reaction originated from Ochrovirga pacifica

    Acetyl xylan esterase plays an important role in complete enzymatic hydrolysis of lignocellulosic materials into fermentable sugars. It hydrolyzes ester linkages of acetic acid in xylan polysaccharide and supports to enhance the activity of xylanase. This study was conducted to recognize and overexpress the acetyl xylan esterase gene found from Ochrovirga pacifica strain S85 which was isolated from Chuuk state, Micronesia. The genome sequence was analyzed with genome sequencer-FLX and acetyl xylan esterase gene (Axe) was detected. The gene had an open reading frame of 864 bp encoding a polypeptide of 287 amino acids. Theoretical molecular mass and isoelectric point (pI) were 32 kDa and 5.9, respectively. The deduced amino acid sequence of the Axe showed 35.1% similarity with both endo-1,4-β-xylanase B from Robiginitalea biformata HTCC2501. The mature protein displayed the catalytic residues classically found in enzymes belonged to GH16 family. Axe was cloned into pET11a vector and recombinant protein was expressed in E. coli BL21 (DE3), purified by nickel affinity chromatography and its purity was visualized on SDS-PAGE. Commercial xylanase activity was tested after treatment of recombinant acetyl xylan esterase (rAXE) to birchwood xylan substrate. The xylanase activity of rAXE treated sample was about 2 times higher than xylanase only treated sample. Please click Additional Files below to see the full abstract
