    Isolation and sequencing of swine carbonic anhydrase VI, an enzyme expressed in the swine kidney

    BACKGROUND: Carbonic anhydrase VI (CA-VI) is produced by the salivary gland and is secreted into the saliva. Although CA-VI is found in the epithelial cells of distal straight tubule of swine kidneys, the exact function of CA-VI in the kidneys remains unclear. RESULTS: CA-VI was located in the epithelial cells of distal straight tubule of swine kidneys. A full-length cDNA clone of CA-VI was generated from the swine parotid gland by reverse transcription polymerase chain reaction, using degenerate primers designed based on conserved regions of the same locus in human and bovine tissues. The cDNA sequence was 1348 base pairs long and was predicted to encode a 317 amino acid polypeptide with a putative signal peptide of 17 amino acids. The deduced amino acid sequence of mature CA-VI was most similar (77.4%) to that of human CA-VI. CA-VI expression was confirmed in both normal and nephritic kidneys, as well as parotid. As the primers used in this study spanned two exons, the influence of genomic DNA was not detected. The expression of CA-VI was demonstrated in both normal and nephritic kidneys, and mRNA of CA-VI in the normal kidneys which was the normalised to an endogenous β–actin was 0.098 ± 0.047, while it was significantly lower in the diseased kidneys (0.012 ± 0.007). The level of CA-VI mRNA in normal kidneys was 19-fold lower than that of the parotid gland (1.887). CONCLUSIONS: The localisation of CA-VI indicates that it may play a specialised role in the kidney

    間伐材利用の地域連携デザインの研究(その1)

    学長特別研究費木の文化の可能性と展望人・もの・社会のより良いあり方や、人と自然との関わりについての探求 天竜地域はじめ国内の林業地域では、間伐材利用方法の開発が、地域・産業振興に係る共通した重要課題とされている。そうしたなか、2004年の浜名湖花博に向けて、本学教員・学生も参加のもと、地元間伐材を利用する木製ベンチのデザイン制作事業が、主にユニバーサルデザインの視点から、進められてきた。 本研究では、その事業に対して木の文化・産業の視点も加えて、一過性のベンチ制作にとどまらず、間伐材利用をコアとした産学などの新たな地域連携システムのモデル化を図っていくことを目的とした。 なお、本研究でのケーススタディーの対象に選定した「浜名湖花博ユニバーサルデザインベンチのデザイン制作」プロジェクトは、2004年に浜松市郊外で開催の浜名湖花博に向けて、(財)静岡国際園芸博覧会協会、天竜市森林組合及び本学教員・学生グループの連携のもと実施されてきたものであり、実験的性格を有する地域連携デザインプロジェクトといえるものである

    Immunostaining of thymidylate synthase and p53 for predicting chemoresistance to S-1/cisplatin in gastric cancer

    High expression of thymidylate synthase (TS) and inactivation of p53 are allegedly associated with chemoresistance. The authors evaluated TS and p53 expression in gastric cancer treated with neoadjuvant S-1/cisplatin chemotherapy. Paraffin sections of pretreatment biopsy and surgical specimens from 41 gastric cancers were immunostained for TS and p53 protein after appropriate antigen retrieval. Fifty-one cases without neoadjuvant chemotherapy were also studied. In the pretreatment biopsies, high expression of TS was seen in 8% of the histologic responders, in 28% of the nonresponders and in 31% of the controls. High expression of p53 was observed in 56% of the nonresponders, but in 8% of the responders and in 29% of the controls (P<0.01 and P<0.05, respectively). The TS- and/or p53-high phenotype was seen in 76% of the nonresponders and in 54% of the controls, but in 8% of the responders (P<0.0001 and P<0.005, respectively). The data of the surgical specimens were consistent with those of the pretreatment biopsies. These results suggest that immunostaining for TS and p53 protein is useful for pretreatment selection of gastric cancer patients unresponsive to S-1/cisplatin chemotherapy

    Synthesis and Antimicrobial Evaluation of Side-Chain Derivatives based on Eurotiumide A

    Side-chain derivatives of eurotiumide A, a dihydroisochroman-type natural product, have been synthesized and their antimicrobial activities described. Sixteen derivatives were synthesized from a key intermediate of the total synthesis of eurotiumide A, and their antimicrobial activities against two Gram-positive bacteria, methicillin-susceptible and methicillin-resistant Staphylococcus aureus (MSSA and MRSA), and a Gram-negative bacterium, Porphyromonas gingivalis, were evaluated. The results showed that derivatives having an iodine atom on their aromatic ring instead of the prenyl moiety displayed better antimicrobial activity than eurotiumide A against MSSA and P. gingivalis. Moreover, we discovered that a derivative with an isopentyl side chain, which is a hydrogenated product of eurotiumide A, is the strongest antimicrobial agent against all three strains, including MRSA

    Development of the ARICA-2 Satellite Using Spresense as an Onboard Computer

    Gamma-ray bursts (GRBs) are transient astronomical phenomena that emit enormous amounts of energy in electromagnetic waves, mainly in the gamma-ray range, for several seconds to tens of seconds. GRB observations are challenging because of the difficulty in predicting the location and time of occurrence and its extremely short duration. Therefore, it is necessary to notify about the discovery in space and to conduct follow-up observations by researchers. The AGU Remote Innovative CubeSat Alert system-2 (ARICA-2) has been developed to demonstrate a new alert system using commercial satellite network services. ARICA-2 uses SONY’s Spresense as its onboard computer (OBC). We manufactured the special board to attach two Spresenses as a redundancy of the OBC system.     Power System Development of the AGU Remote Innovative CubeSat Alert System -2 – ARICA-2

    We present the power system development of the 2U CubeSat, AGU Remote Innovative CubeSat Alert system -2(ARICA-2). The main goal of the ARICA-2 project is to demonstrate the real-time alert system of transient astronomical sources using commercial satellite network devices. 1U CubeSat ARICA was launched in November 2021. However, we have not been able to send and receive the data at this point. Therefore, we started developing 2U CubeSat ARICA-2, which is an improved version of ARICA, in April 2022. One of the possible causes of the communication problem of ARICA is the power system, such as a negative power budget or a failure in the installation of the inhibit switches. ARICA-2 is upsized from 1U to 2U to ensure a sufficient power generation and is equipped with improved inhibit switches. The calculation of power consumption and simulation of power supply on orbit have been finished. We confirmed the performance of our Electric Power System (EPS) and the health of the installed batteries. We are currently in the EM development phase with the goal of launching in Japanese fiscal year 2024
