    α-Linolenic acid supplementation in BioXcell® extender can improve the quality of post-cooling and frozen-thawed bovine sperm

    The present study was conducted to determine the effects of supplementing α-linolenic acid (ALA) into BioXcell® extender on post-cooling, post-thawed bovine spermatozoa and post thawed fatty acid composition. Twenty-four semen samples were collected from three bulls using an electro-ejaculator. Fresh semen samples were evaluated for general motility using computer assisted semen analyzer (CASA) whereas morphology and viability with eosin–nigrosin stain. Semen samples extended into BioXcell® were divided into five groups to which 0, 3, 5, 10 and 15 ng/ml of ALA were added, respectively. The treated samples were incubated at 37 °C for 15 min for ALA uptake by sperm cells before being cooled for 2 h at 5 °C. After evaluation, the cooled samples were packed into 0.25 ml straws and frozen in liquid nitrogen for 24 h before thawing and evaluation for semen quality. Evaluation of cooled and frozen-thawed semen showed that the percentages of all the sperm parameters improved with 5 ng/ml ALA supplement. ALA was higher in all treated groups than control groups than control group. In conclusion, 5 ng/ml ALA supplemented into BioXcell® extender improved the cooled and frozen-thawed quality of bull spermatozoa

    Nontraditional Feedstuffs as an Alternative in Poultry Feed

    Soybean meal and yellow corn are conventional feedstuffs used as the main ingredients in poultry feeds due to their high nutrients availability. On the other hand, these two feedstuffs are high in demand by other animals (soybean meal) and humans (yellow corn). By the year 2050, the world’s population is expected to increase up to 9.1 billion. Global consumption of poultry products, such as meat or eggs is increasing predominantly in developing countries. Consequently, the global demand for poultry feedstuffs would increase. The availability of feedstuffs for poultry nutrition nowadays is becoming more competitive. Thus, food security, especially in the developing countries, would be threatened. Currently, efforts are being made to use alternative feedstuffs to substitute portion of soybean meal and yellow corn in poultry diets. This chapter discusses the alternative feedstuffs that can be incorporated in poultry feeds. In addition, the nutritive content and availability are examined as well as how to improve the nutritive quality of such nontraditional feedstuffs

    Pre-emptive multimodal analgesia with tramadol and ketamine–lidocaine infusion for suppression of central sensitization in a dog model of ovariohysterectomy

    Objectives: The effects of pre-emptive infusion of ketamine-lidocaine with tramadol on the suppression of central sensitization were investigated in a dog ovariohysterectomy model. Patients and methods: Twelve dogs were randomly assigned to two groups: ketamine-lidocaine-tramadol (KLT) and tramadol (T) groups. Both groups received intravenous tramadol 4 mg/kg body weight as premedication. Immediately after induction, the KLT group received ketamine and lidocaine at 0.5 and 2 mg/kg loading dose, followed by continuous rate infusion of 50 and 100 µg/kg/min, respectively, for 2 hours. Dogs in T group received saline bolus and continuous rate infusion at equi-volume. Intraoperatively, hemodynamic responses to surgical stimulation were recorded, whereas postoperative pain was evaluated using an algometer and short form of the Glasgow composite measure pain scale. Results: Intraoperatively, hemodynamic responses to surgical stimulation were obtunded to a greater degree in KLT compared to T group. Postoperatively, the pain scores increased only for the first hour in KLT group, compared to 12 hours in T group. Mechanical thresholds at the abdomen decreased postoperatively between 12 and 60 hours in KLT group versus the entire 72 hours in T group. Thresholds at tibia and radius in both groups increased in the immediate 1 hour postoperatively, but decreased thereafter. Significant decrement of thresholds from baseline were detected in the tibia at 24, 42, and 60 hours in KLT group compared to 24-72 hours in T group, and in the radius between 36 and 48 hours in T group, but none in KLT group. Conclusion: Addition of pre-emptive ketamine-lidocaine infusion to single intravenous dose of tramadol enhanced attenuation of central sensitization and improved intra- and postoperative analgesia

    Effect of feeding fermented and non-fermented palm kernel cake on the performance of broiler chickens: a review

    Palm kernel cake (PKC) is a by-product of oil extraction from palm fruits and has been included in poultry diets as an alternative to soybean meal and yellow corn. Due to its high content of fibre, coarse texture and gritty appearance, the use of PKC in poultry nutrition is limited. In order to increase the nutritive value of PKC, there is a tendency nowadays to create solid state fermentation (SSF) by using cellulolytic microbes. This paper reviews the impact of feeding fermented and non-fermented PKC on the performance of broiler chickens. Recent studies have reported that SSF by cellulolytic microorganisms improved the nutritive value of PKC. The nutrient digestibility has been increased significantly in PKC fermented using Paenibacillus polymyxya ATCC 842 or Weisella confusa SR-17b. The availability of valine, histidine, methionine and arginine was 70.42%, 71.50%, 71.92% and 81.15%, respectively, in PKC fermented using P. polymyxa ATCC 842. The digestibility of crude protein (CP) increased by 61.83% and 59.90% in PKC fermented using P. polymyxya ATCC 842 or W. confusa SR-17b, respectively. In addition, body weight gain (BWG) and feed conversion ratio (FCR) improved significantly in broilers fed 15% fermented PKC compared to those fed 15% non-fermented PKC (2000.43 g versus 1823.23 g and 1.75 versus 1.91, respectively). The intestinal Enterobacteriaceae decreased (4.03 CFU/g) and lactic acid bacteria increased (5.56 CFU/g) in birds fed 15% PKC fermented by P. polymyxa ATCC 842. Therefore, fermented PKC can be included in a broiler diet up to 15%, replacing part of soybean and yellow corn in the diet, leading to a decrease in the overall cost of poultry feeding

    A comparison of bleeding efficiency, microbiological quality and lipid oxidation in goats subjected to conscious halal slaughter and slaughter following minimal anesthesia

    The study assessed the effect of conscious halal slaughter and slaughter following minimal anesthesia on bleeding efficiency of goats and keeping quality of goat meat. Ten Boer cross bucks were divided into two groups and subjected to either halal slaughter without stunning (HS) or minimal anesthesia prior to slaughter (AS). The blood lost during exsanguination was measured. Residual blood was further quantified by determination of hemoglobin and myoglobin content in longissimus lumborum muscle. Storage stability of the meat was evaluated by microbiological analysis and lipid oxidation. Blood loss at exsanguination, residual hemoglobin and lipid oxidation were not significantly different (p>0.05) between HS and AS. Lactic acid bacteria was the only microbe that was significantly elevated after 24h of storage at 4°C in the AS group. In conclusion, slaughtering goats under minimal anesthesia or fully conscious did not affect bleeding efficiency and keeping quality of goat meat

    Effect of caponization on muscle, liver and adipose tissue fatty acid composition, lipid peroxidation, and cholesterol in breast muscle of Cobb 500 broilers

    Capon is the surgical removal of testes from roosters which promotes the accumulation of intramuscular fat and affects fatty acid composition of poultry meat. We report effect of caponization on muscle, liver and adipose tissue fatty acid composition, lipid peroxidation and cholesterol content in breast muscle of broilers. Groups of 30 male 1-d old commercial Cobb 500 broilers were caponized at 21 days of age. Controls were 30 intact birds. Each treatment group consisted of 6 replicates (cages) with 5 birds in each cage. Broilers were fed commercial feed and slaughtered at 40 days. Major fatty acids in breast muscle and adipose tissue were oleic (C18:1, n-9), palmitic (C16:0) and linoleic (C18:2, n-6) and stearic (C18:0). Caponization did not affect fatty acid composition of muscle, liver or adipose tissues or lipid peroxidation of meat. However, we did find lower (P < 0.018) cholesterol (0.66 ± 0.02mg/ml) in breast muscle from caponized birds (n=9) compared with 9 intact controls (0.77 ± 0.04mg/ml). Our findings suggest that caponization does not affect free fatty acid composition or lipid peroxidation but may reduce cholesterol in tissues of broilers

    Effects of slaughter positions on catecholamine, blood biochemical and electroencephalogram changes in cattle restrained using a modified Mark IV box

    The proper slaughter positioning of animals is among the most crucial factors in animal welfare. The lateral position in Halal slaughter is a technique used around the world by Muslims, with a few practicing the upright position. The literature on the effects of slaughter in upright versus lateral positions on pain and stress is scarce. Thus, this study was designed to evaluate the effects of slaughter positions on blood biochemical parameters, plasma catecholamines, and electroencephalographic (EEG) responses. Twenty Brahman crossbred steers were subjected to slaughter in either lateral recumbency (LP) (n = 10) or an upright position (UP) (n = 10). There was a significant increase in adrenaline (p < 0.0001) and noradrenaline (p < 0.05) at T2 compared to T1 in the animals of both groups. A significant difference (p < 0.0001) was observed in the median frequency (MF) and total power (Ptot) of EEG, parameters for pain and stress, between the animals slaughtered in the upright and the lateral position. However, MF and delta waves were significantly higher (p < 0.05) after slaughter in the UP group than in the LP group. The results demonstrate a lesser amount of stress and pain responses among the LP group

    Electroencephalographic responses to neck cut and exsanguination in minimally anaesthetized goats

    Conscious animals typically experience sensory (nociception) and emotional pain, whereas unconscious animals that were minimally anesthetized would experience minimal emotional pain. To determine whether ‘silencing’ the emotional component through a minimally anesthetized model would minimize stress response, and thus improve animal welfare, this study aimed at comparing changes in electroencephalographic (EEG) activities associated with possible noxious stimuli following neck-cut slaughter in conscious non-anesthetized versus minimally anaesthetized Boer cross-bred goats. Ten bucks were randomly assigned to two groups of five animals each, and subjected to neck-cut slaughter when fully conscious (HS) or under minimal anaesthesia (AS) and exsanguinated. The anaesthesia was induced with propofol (5 mg/kg) administered to effect by rapid injection into a cephalic vein and maintained with halothane in 100 % oxygen. Changes in the root mean square (RMS) for each of alpha, beta, delta and theta waves, median frequency (F50) and total power of the EEG (Ptot) were compared in each group before and after neck cut and between groups following treatments. Electroencephalographic parameters did not differ between goats that were fully conscious or slaughtered under minimal anaesthesia. These findings showed that the noxious stimuli from neck cut were present in both conscious and minimally anaesthetized goats. Most importantly, the presence of emotional pain and nociception did not affect the extent of electroencephalographic responses significantly compared with animals that were experiencing nociception only

    Postnatal ex vivo rat model for longitudinal bone growth investigations

    Background: Chondrocytes in the growth plate (GP) undergo increases in volume during different cascades of cell differentiation during longitudinal bone growth. The volume increase is reported to be the most significant variable in understanding the mechanism of long bone growth. Methods: Forty‐five postnatal Sprague‐Dawley rat pups, 7‐15 days old were divided into nine age groups (P7‐P15). Five pups were allocated to each group. The rats were sacrificed and tibia and metatarsal bones were harvested. Bone lengths were measured after 0, 24, 48, and 72 hours of ex vivo incubation. Histology of bones was carried out, and GP lengths and chondrocyte densities were determined. Results: There were significant differences in bone length among the age groups after 0 and 72 hours of incubation. Histological sectioning was possible in metatarsal bone from all age groups, and in tibia from 7‐to 13‐day‐old rats. No significant differences in tibia and metatarsal GP lengths were seen among different age groups at 0 and 72 hours of incubation. Significant differences in chondrocyte densities along the epiphyseal GP of the bones between 0 and 72 hours of incubation were observed in most of the age groups. Conclusion: Ex vivo growth of tibia and metatarsal bones of rats aged 7‐15 days old is possible, with percentage growth rates of 23.87 ± 0.80% and 40.38 ± 0.95%measured in tibia and metatarsal bone, respectively. Histological sectioning of bones was carried out without the need for decalcification in P7‐P13 tibia and P7‐P15metatarsal bone. Increases in chondrocyte density along the GP influence overall bone elongation

    Changes of microbial spoilage, lipid-protein oxidation and physicochemical properties during post mortem refrigerated storage of goat meat

    Examined was the effect of post mortem refrigerated storage on microbial spoilage, lipid-protein oxidation and physicochemical traits of goat meat. Seven Boer bucks were slaughtered, eviscerated and aged for 24 h. The Longissimus lumborum (LL) and Semitendinosus (ST) muscles were excised and subjected to 13 days post mortem refrigerated storage. The pH, lipid and protein oxidation, tenderness, color and drip loss were determined in LL while microbiological analysis was performed on ST. Bacterial counts generally increased with increasing aging time and the limit for fresh meat was reached at day 14 post mortem. Significant differences were observed in malondialdehyde (MDA) content at day 7 of storage. The thiol concentration significantly reduced as aging time increased. The band intensities of myosin heavy chain (MHC) and troponin-T significantly decreased as storage progressed, while actin remained relatively stable. After 14 days of aging, tenderness showed significant improvement while muscle pH and drip loss reduced with increase in storage time. Samples aged for 14 days had higher lightness (P < 0.05) and lower (P < 0.05) yellowness and redness.
