    MFG-E8 induced differences in proteomic profiles in mouse C2C12 cells and its effect on PI3K/Akt and ERK signal pathways

    Milk fat globule-EGF factor 8 (MFG-E8) is one of the major proteins in milk fat globule membrane. In this study, mouse-derived C2C12 myoblast cells were served as an experimentally tractable model system for investigating the molecular basis of skeletal muscle cell specification and development. To examine the biochemical adaptations associated with myocytes formation comprehensively, a liquid chromatography coupled with tandem mass spectrometry label-free semi-quantitative  approach was used to analyse the myogenic C2C12 proliferation program. Over 1987  proteins were identified in C2C12 cells. The MFG-E8 (200 mg/mL) and MFG-E8 (500 26 mg/mL) with significant differences were compared based on the relative abundance. The result profiles of regulation of MFG-E8 to the expression of proteins in C2C12 cells revealed that differential waves of expression of proteins linked to intracellular signaling, transcription, cytoarchitecture, adhesion, metabolism, and muscle contraction across during the C2C12 cell proliferation process. Based on the analysis of  KEGG and STRING database, further to verification the expression of PI3K and ERK phosphorylation levels by Western blot. This study found that the data of proteomic was complementary to recent MFG-E8 studies of protein expression patterns in developing myotubes and provided a holistic framework for understanding how diverse biochemical processes are coordinated at the cellular level during skeletal muscle development

    Identification of polycentric cities in China based on NPP-VIIRS nighttime light data

    Nighttime light data play an important role in the research on cities, while the urban centers over a large spatial scale are still far from clearly understood. Aiming at the current challenges in monitoring the spatial structure of cities using nighttime light data, this paper proposes a new method for identifying urban centers for massive cities at the large spatial scale based on the brightness information captured by the Suomi National Polar-Orbiting Partnership’s Visible Infrared Imaging Radiometer Suite (NPP-VIIRS) sensor. Based on the method for extracting the peak point based on digital elevation model (DEM) data in terrain analysis, the maximum neighborhood and di_erence algorithms were applied to the NPP-VIIRS data to extract the pixels with the peak nighttime light intensity to identify the potential locations of urban centers. The results show 7239 urban centers in 2200 cities in China in 2017, with an average of 3.3 urban centers per city. Approximately 68% of the cities had significant polycentric structures. The developed method in this paper is useful for identifying the urban centers and can provide the reference to the city planning and construction

    Effects of Low Temperature at Booting Stage on Sucrose Metabolism and Endogenous Hormone Contents in Winter Wheat Spikelet

    Low spring temperatures often occur during the winter wheat booting stage, when the young ears are very sensitive to cold. In this study, we used two wheat varieties differing in cold sensitivity (sensitive variety Yangmai 18 and tolerant variety Yannong 19) to examine the effect of low temperature on wheat grain number at booting stage. Low temperature stress was simulated in an artificial climate chamber at 4°C for 60 h in 2016 and at 2, 0, or −2°C for 24 h in morphological assays, showing that the development of wheat spikelets was inhibited and floret growth was delayed following low temperature stress. However, an increase in the sucrose content of young panicles was also observed, and the activity of enzymes involved in sucrose metabolism was dynamically altered. Sucrose phosphate synthase activity was enhanced, and sucrose synthase activity significantly increased after treatment at 4 and 2°C, respectively. However, activities of sucrose synthase and invertase decreased with a reduction in temperature. Gene expression assays further revealed downregulation of TaSuS1 expression and upregulation of TaSuS2, while expression of CWINV was inhibited. Moreover, phytohormone content assays showed an increase in the content of abscisic acid in young wheat ears, but a decrease in the content of auxin and gibberellins. The grain number per spike and 1000-grain weight also showed a downward trend following low temperature stress. Overall, these findings suggest that low temperature at booting induces abscisic acid accumulation in winter wheat, altering the activity of the enzymes involved in sucrose metabolism, which leads to an accumulation of sucrose in the young ears, thereby having a negative effect on wheat production

    Intra-Familial Phenotypic Heterogeneity and Telomere Abnormality in von Hippel- Lindau Disease: Implications for Personalized Surveillance Plan and Pathogenesis of VHL-Associated Tumors

    von Hippel-Lindau (VHL) disease is a hereditary cancer syndrome with poor survival. The current recommendations have proposed uniform surveillance strategies for all patients, neglecting the obvious phenotypic varieties. In this study, we aim to confirm the phenotypic heterogeneity in VHL disease and the underlying mechanism. A total of 151 parent-child pairs were enrolled for genetic anticipation analysis, and 77 sibling pairs for birth order effect analysis. Four statistical methods were used to compare the onset age of patients among different generations and different birth orders. The results showed that the average onset age was 18.9 years earlier in children than in their parents, which was statistically significant in all of the four statistical methods. Furthermore, the first-born siblings were affected 8.3 years later than the other ones among the maternal patients. Telomere shortening was confirmed to be associated with genetic anticipation in VHL families, while it failed to explain the birth order effect. Moreover, no significant difference was observed for overall survival between parents and children (p = 0.834) and between first-born patients and the other siblings (p = 0.390). This study provides definitive evidence and possible mechanisms of intra-familial phenotypic heterogeneity in VHL families, which is helpful to the update of surveillance guidelines

    Estimating and Interpreting Fine-Scale Gridded Population Using Random Forest Regression and Multisource Data

    Gridded population results at a fine resolution are important for optimizing the allocation of resources and researching population migration. For example, the data are crucial for epidemic control and natural disaster relief. In this study, the random forest model was applied to multisource data to estimate the population distribution in impervious areas at a 30 m spatial resolution in Chongqing, Southwest China. The community population data from the Chinese government were used to validate the estimation accuracy. Compared with the other regression techniques, the random forest regression method produced more accurate results (R2 = 0.7469, RMSE = 2785.04 and p < 0.01). The points of interest (POIs) data played a more important role in the population estimation than the nighttime light images and natural topographical data, particularly in urban settings. Our results support the wide application of our method in mapping densely populated cities in China and other countries with similar characteristics
