    Cell Type-Dependent RNA Recombination Frequency in the Japanese Encephalitis Virus

    Japanese encephalitis virus (JEV) is one of approximately 70 flaviviruses, frequently causing symptoms involving the central nervous system. Mutations of its genomic RNA frequently occur during viral replication, which is believed to be a force contributing to viral evolution. Nevertheless, accumulating evidences show that some JEV strains may have actually arisen from RNA recombination between genetically different populations of the virus. We have demonstrated that RNA recombination in JEV occurs unequally in different cell types. In the present study, viral RNA fragments transfected into as well as viral RNAs synthesized in mosquito cells were shown not to be stable, especially in the early phase of infection possibly via cleavage by exoribonuclease. Such cleaved small RNA fragments may be further degraded through an RNA interference pathway triggered by viral double-stranded RNA during replication in mosquito cells, resulting in a lower frequency of RNA recombination in mosquito cells compared to that which occurs in mammalian cells. In fact, adjustment of viral RNA to an appropriately lower level in mosquito cells prevents overgrowth of the virus and is beneficial for cells to survive the infection.     New Calibration Method Using Low Cost MEM IMUs to Verify the Performance of UAV-Borne MMS Payloads

    Spatial information plays a critical role in remote sensing and mapping applications such as environment surveying and disaster monitoring. An Unmanned Aerial Vehicle (UAV)-borne mobile mapping system (MMS) can accomplish rapid spatial information acquisition under limited sky conditions with better mobility and flexibility than other means. This study proposes a long endurance Direct Geo-referencing (DG)-based fixed-wing UAV photogrammetric platform and two DG modules that each use different commercial Micro-Electro Mechanical Systems’ (MEMS) tactical grade Inertial Measurement Units (IMUs). Furthermore, this study develops a novel kinematic calibration method which includes lever arms, boresight angles and camera shutter delay to improve positioning accuracy. The new calibration method is then compared with the traditional calibration approach. The results show that the accuracy of the DG can be significantly improved by flying at a lower altitude using the new higher specification hardware. The new proposed method improves the accuracy of DG by about 20%. The preliminary results show that two-dimensional (2D) horizontal DG positioning accuracy is around 5.8 m at a flight height of 300 m using the newly designed tactical grade integrated Positioning and Orientation System (POS).     Counting Crowds in Bad Weather

    Crowd counting has recently attracted significant attention in the field of computer vision due to its wide applications to image understanding. Numerous methods have been proposed and achieved state-of-the-art performance for real-world tasks. However, existing approaches do not perform well under adverse weather such as haze, rain, and snow since the visual appearances of crowds in such scenes are drastically different from those images in clear weather of typical datasets. In this paper, we propose a method for robust crowd counting in adverse weather scenarios. Instead of using a two-stage approach that involves image restoration and crowd counting modules, our model learns effective features and adaptive queries to account for large appearance variations. With these weather queries, the proposed model can learn the weather information according to the degradation of the input image and optimize with the crowd counting module simultaneously. Experimental results show that the proposed algorithm is effective in counting crowds under different weather types on benchmark datasets.     The Design a TDCP-Smoothed GNSS/Odometer Integration Scheme with Vehicular-Motion Constraint and Robust Regression

    Global navigation satellite system (GNSS) is widely regarded as the primary positioning solution for intelligent transport system (ITS) applications. However, its performance could degrade, due to signal outages and faulty-signal contamination, including multipath and non-line-of-sight reception. Considering the limitation of the performance and computation loads in mass-produced automotive products, this research investigates the methods for enhancing GNSS-based solutions without significantly increasing the cost for vehicular navigation system. In this study, the measurement technique of the odometer in modern vehicle designs is selected to integrate the GNSS information, without using an inertial navigation system. Three techniques are implemented to improve positioning accuracy; (a) Time-differenced carrier phase (TDCP) based filter: A state-augmented extended Kalman filter is designed to incorporate TDCP measurements for maximizing the effectiveness of phase-smoothing; (b) odometer-aided constraints: The aiding measurement from odometer utilizing forward speed with the lateral constraint enhances the state estimation; the information based on vehicular motion, comprising the zero-velocity constraint, fault detection and exclusion, and dead reckoning, maintains the stability of the positioning solution; (c) robust regression: A weighted-least-square based robust regression as a measurement-quality assessment is applied to adjust the weightings of the measurements adaptively. Experimental results in a GNSS-challenging environment indicate that, based on the single-point-positioning mode with an automotive-grade receiver, the combination of the proposed methods presented a root-mean-square error of 2.51 m, 3.63 m, 1.63 m, and 1.95 m for the horizontal, vertical, forward, and lateral directions, with improvements of 35.1%, 49.6%, 45.3%, and 21.1%, respectively. The statistical analysis exhibits 97.3% state estimation result in the horizontal direction for the percentage of epochs that had errors of less than 5 m, presenting that after the intervention of proposed methods, the positioning performance can fulfill the requirements for road level applications.     High-Frequency Sea Level Variations Observed by GPS Buoys Using Precise Point Positioning Technique

    In this study, sea level variation observed by a 1-Hz Global Positioning System (GPS) buoy system is verified by comparing with tide gauge records and is decomposed to reveal high-frequency signals that cannot be detected from 6-minute tide gauge records. Compared to tide gauges traditionally used to monitor sea level changes and affected by land motion, GPS buoys provide high-frequency geocentric measurements of sea level variations. Data from five GPS buoy campaigns near a tide gauge at Anping, Tainan, Taiwan, were processed using the Precise Point Positioning (PPP) technique with four different satellite orbit products from the International GNSS Service (IGS). The GPS buoy data were also processed by a differential GPS (DGPS) method that needs an additional GPS receiver as a reference station and the accuracy of the solution depends on the baseline length. The computation shows the average Root Mean Square Error (RMSE) difference of the GPS buoy using DGPS and tide gauge records is around 3 - 5 cm. When using the aforementioned IGS orbit products for the buoy derived by PPP, its average RMSE differences are 5 - 8 cm, 8 - 13 cm, decimeter level, and decimeter-meter level, respectively, so the accuracy of the solution derived by PPP highly depends on the accuracy of IGS orbit products. Therefore, the result indicates that the accuracy of a GPS buoy using PPP has the potential to measure the sea surface variations to several cm.     Study of Alternative GPS Network Meteorological Sensors in Taiwan: Case Studies of the Plum Rains and Typhoon Sinlaku

    Plum rains and typhoons are important weather systems in the Taiwan region. They can cause huge economic losses, but they are also considered as important water resources as they strike Taiwan annually and fill the reservoirs around the island. There are many meteorological sensors available for investigating the characteristics of weather and climate systems. Recently, the use of GPS as an alternative meteorological sensor has become popular due to the catastrophic impact of global climate change. GPS provides meteorological parameters mainly from the atmosphere. Precise Point Positioning (PPP) is a proven algorithm that has attracted attention in GPS related studies. This study uses GPS measurements collected at more than fifty reference stations of the e-GPS network in Taiwan. The first data set was collected from June 1st 2008 to June 7th 2008, which corresponds to the middle of the plum rain season in Taiwan. The second data set was collected from September 11th to September 17th 2008 during the landfall of typhoon Sinlaku. The data processing strategy is to process the measurements collected at the reference stations of the e-GPS network using the PPP technique to estimate the zenith tropospheric delay (ZTD) values of the sites; thus, the correlations between the ZTD values and the variation of rainfall during the plum rains and typhoon are analyzed. In addition, several characteristics of the meteorological events are identified using spatial and temporal analyses of the ZTD values estimated with the GPS network PPP technique

    Intelligent Sensor Positioning and Orientation Through Constructive Neural Network-Embedded INS/GPS Integration Algorithms

    Mobile mapping systems have been widely applied for acquiring spatial information in applications such as spatial information systems and 3D city models. Nowadays the most common technologies used for positioning and orientation of a mobile mapping system include a Global Positioning System (GPS) as the major positioning sensor and an Inertial Navigation System (INS) as the major orientation sensor. In the classical approach, the limitations of the Kalman Filter (KF) method and the overall price of multi-sensor systems have limited the popularization of most land-based mobile mapping applications. Although intelligent sensor positioning and orientation schemes consisting of Multi-layer Feed-forward Neural Networks (MFNNs), one of the most famous Artificial Neural Networks (ANNs), and KF/smoothers, have been proposed in order to enhance the performance of low cost Micro Electro Mechanical System (MEMS) INS/GPS integrated systems, the automation of the MFNN applied has not proven as easy as initially expected. Therefore, this study not only addresses the problems of insufficient automation in the conventional methodology that has been applied in MFNN-KF/smoother algorithms for INS/GPS integrated systems proposed in previous studies, but also exploits and analyzes the idea of developing alternative intelligent sensor positioning and orientation schemes that integrate various sensors in more automatic ways. The proposed schemes are implemented using one of the most famous constructive neural networks—the Cascade Correlation Neural Network (CCNNs)—to overcome the limitations of conventional techniques based on KF/smoother algorithms as well as previously developed MFNN-smoother schemes. The CCNNs applied also have the advantage of a more flexible topology compared to MFNNs.     An Artificial Neural Network Embedded Position and Orientation Determination Algorithm for Low Cost MEMS INS/GPS Integrated Sensors

    Digital mobile mapping, which integrates digital imaging with direct geo-referencing, has developed rapidly over the past fifteen years. Direct geo-referencing is the determination of the time-variable position and orientation parameters for a mobile digital imager. The most common technologies used for this purpose today are satellite positioning using Global Positioning System (GPS) and Inertial Navigation System (INS) using an Inertial Measurement Unit (IMU). They are usually integrated in such a way that the GPS receiver is the main position sensor, while the IMU is the main orientation sensor. The Kalman Filter (KF) is considered as the optimal estimation tool for real-time INS/GPS integrated kinematic position and orientation determination. An intelligent hybrid scheme consisting of an Artificial Neural Network (ANN) and KF has been proposed to overcome the limitations of KF and to improve the performance of the INS/GPS integrated system in previous studies. However, the accuracy requirements of general mobile mapping applications can’t be achieved easily, even by the use of the ANN-KF scheme. Therefore, this study proposes an intelligent position and orientation determination scheme that embeds ANN with conventional Rauch-Tung-Striebel (RTS) smoother to improve the overall accuracy of a MEMS INS/GPS integrated system in post-mission mode. By combining the Micro Electro Mechanical Systems (MEMS) INS/GPS integrated system and the intelligent ANN-RTS smoother scheme proposed in this study, a cheaper but still reasonably accurate position and orientation determination scheme can be anticipated

    LACMA: Language-Aligning Contrastive Learning with Meta-Actions for Embodied Instruction Following

    End-to-end Transformers have demonstrated an impressive success rate for Embodied Instruction Following when the environment has been seen in training. However, they tend to struggle when deployed in an unseen environment. This lack of generalizability is due to the agent's insensitivity to subtle changes in natural language instructions. To mitigate this issue, we propose explicitly aligning the agent's hidden states with the instructions via contrastive learning. Nevertheless, the semantic gap between high-level language instructions and the agent's low-level action space remains an obstacle. Therefore, we further introduce a novel concept of meta-actions to bridge the gap. Meta-actions are ubiquitous action patterns that can be parsed from the original action sequence. These patterns represent higher-level semantics that are intuitively aligned closer to the instructions. When meta-actions are applied as additional training signals, the agent generalizes better to unseen environments. Compared to a strong multi-modal Transformer baseline, we achieve a significant 4.5% absolute gain in success rate in unseen environments of ALFRED Embodied Instruction Following. Additional analysis shows that the contrastive objective and meta-actions are complementary in achieving the best results, and the resulting agent better aligns its states with corresponding instructions, making it more suitable for real-world embodied agents.
