    Dengue Virus Infects Primary Human Hair Follicle Dermal Papilla Cells

    During the epidemic of the dengue virus (DENV) infection in Taiwan in 2014 and 2015, we observed an abnormally high frequency of increased scalp hair shedding in infected individuals that could not be explained by telogen effluvium. In this study, the mechanism of hair loss caused by DENV was explored. Human hair follicle dermal papilla cells (HFDPCs) are essential for hair follicle morphogenesis and cycling. Thus, we established an in vitro DENV infection model in HFDPCs. On immunofluorescence analysis, HFDPCs that were susceptible to DENV infection responded to type I interferon (IFN) treatment, and the cells showed antibody-dependent enhancement (ADE) effect. The expression of the pro-inflammatory cytokines, interleukin 6 (IL-6), and tumor necrosis factor-alpha (TNF-α), revealed an inflammatory response in DENV-infected HFDPCs. In particular, DENV infection impaired cell viability, and it activated caspase-associated cell death signaling in HFDPCs. In conclusion, our data indicate that direct infection with DENV causes inflammation and cell death in HFDPCs, which is involved in the mechanisms of hair loss after DENV infection. The knowledge of DENV infection in an immune-privileged tissue, such as hair follicles, may suggest their use for further studies on post-dengue fatigue syndrome (PDFS)

    Perceptions, Behavioral Expectations, and Implementation Timing for Response Actions in a Hurricane Emergency

    This study examined the perceived attributes, behavioral expectations, and expected implementation timing of 11 organizational emergency response actions for hurricane emergencies. The perceived attributes of the hurricane response actions were characterized by two hazard-related attributes (effectiveness for person protection and property protection) and five resource-related attributes (financial costs, required knowledge/skill, required equipment, required time/effort, and required cooperation). A total of 155 introductory psychology students responded to a hypothetical scenario involving an approaching Category 4 hurricane. The data collected in this study explain previous findings of untimely protective action decision making. Specifically, these data reveal distinctly different patterns for the expected implementation of preparatory actions and evacuation recommendations. Participants used the hazard-related and resource-related attributes to differentiate among the response actions and the expected timing of implementation. Moreover, participants’ behavioral expectations and expected implementation timing for the response actions were most strongly correlated with those actions’ effectiveness for person protection. Finally, participants reported evacuation implementation times that were consistent with a phased evacuation strategy in which risk areas are evacuated in order of their proximity to the coast. However, the late initiation of evacuation in risk areas closest to the coast could lead to very late evacuation of risk areas farther inland

    Evolving Patterns of Metastasis in Renal Cell Carcinoma: Do We Need to Perform Routine Bone Imaging?

    Advance diagnostic and treatment modalities have improved outcomes for renal cell carcinoma (RCC) patients, but the prognosis for those with metastatic disease (mRCC) remains poor. As given metastatic distribution is critical in guiding treatment decisions for mRCC patients, we evaluated evolving metastatic patterns to assess if our current practice standards effectively address patient needs. A systematic literature review was performed to identify all publicly available prospective clinical trials in metastatic renal cell carcinoma (mRCC) from 1990 to 2018. A total of 16,899 mRCC patients from 127 qualified phase I–III clinical trials with metastatic site documentations were included for analysis for incidence of metastases to lung, liver, bone, and lymph nodes (LNs) over time. Studies were categorized into three treatment eras based on the timing of regulatory approval: Cytokine Era (1990-2004), vascular endothelial growth factor/tyrosine kinase inhibitor (TKI) Era (2005-2016), and immune checkpoint inhibitor/TKI Era (ICI-TKI, 2017-2018) and also classified as first-line only (FLO) or second-line and beyond (SLB). Overall, an increase in the incidence of bone and LNs metastases in FLO and SLB, and lung metastases in FLO, was seen over the three treatment eras. Generally, the burden of disease is higher in SLB when compared with FLO. Importantly, in the ICI-TKI era, the incidences of bone metastasis are 28% in FLO and 29% in SLB settings. The disease burden in patients with mRCC has increased steadily over the past three decades. Given the unexpectedly high rate of bone metastasis, routine dedicated bone imaging should be considered in all patients with mRCC

    Association of early childhood constipation with the risk of autism spectrum disorder in Taiwan: Real-world evidence from a nationwide population-based cohort study

    BackgroundAutism spectrum disorder (ASD) is a neurodevelopmental problem that presents with limited interests, repetitive behaviors, and deficits in reciprocal communication and social interactions. Mounting evidence indicates that an imbalanced gut microbiota contributes to autism via the gut-brain axis. Constipation may result in alteration of the gut microbiota. The clinical influence of constipation on ASD has not been fully researched. Thus, in this study we aimed to evaluate whether early childhood constipation influenced the risk of developing ASD using a nationwide population-based cohort study.MethodsWe identified 12,935 constipated children aged 3 years or younger from the National Health Insurance Research Database (NHIRD) in Taiwan from 1997 to 2013. Non-constipated children were also selected from the database and propensity score matching of age, gender, and underlying comorbidities was conducted with a ratio of 1:1. Kaplan–Meier analysis was applied to determine different levels of constipation severity and cumulative incidence of autism. Subgroup analysis was also applied in this study.ResultsThe incidence rate of ASD was 12.36 per 100,000 person-months in the constipation group, which was higher than the rate of 7.84 per 100,000 person-months noted in the non-constipation controls. Constipated children had a significantly higher risk of autism when compared to the non-constipation group (crude relative risk = 1.458, 95% CI = 1.116–1.904; adjusted hazard ratio = 1.445, 95% CI = 1.095–1.907).Moreover, among constipated children, a higher number of laxative prescriptions, male gender, constipation during infancy, and atopic dermatitis were significantly associated with higher risks of ASD when compared to the non-constipation group.ConclusionConstipation in early childhood was correlated with a significantly increased risk of ASD. Clinicians should pay attention to the possibility of ASD in constipated children. Further research is necessary to study the possible pathophysiological mechanisms of this association

    Field Evaluation of Four Low-cost PM Sensors and Design, Development and Field Evaluation of A Wearable PM Exposure Monitoring System

    To mitigate the significant biases/errors in research studying the associations between PM and health, which are introduced by the coarse/inadequate assessments of PM exposure from conventional PM monitoring paradigm, a personalized monitoring system consisting of a low-cost wearable PM device is proposed. However, due to the absence of a unifying evaluation protocol for low-cost PM sensors, the evaluation results/performance specifications from existing studies/datasheets are of limited reference values when attempting to determine the best candidate for the proposed system. In this regard, the authors appeal to the research community to develop a standardized evaluation protocol for low-cost PM sensors/devices, and a unifying attempt is established in this manuscript by adopting the definitive terminology from international documents and the evaluation metrics regarded as best practices. Collocated on the rooftop of the HKUST Supersite, four empirically selected PM sensors were compared against each other and calibrated against two reference monitors. They were then evaluated against the reference following the protocol. The PlanTower PMS-A003 sensor was selected for the wearable device as it outperformed the others in terms of affordability, portability, detection capability, data quality, as well as humidity and condensation insusceptibility. An automated approach was proposed to identify and remove the condensation associated abnormal measurements. The proposed device has better affordability and portability as well as similar usability and data accessibility compared to those existing devices recognized. The first 10 devices were also evaluated and calibrated at the Supersite. Additional 120 units were manufactured and delivered to the subjects to acquire their daily PM2.5 exposures for investigating the association with subclinical atherosclerosis

    Hydrogen isotope exchange experiments in high entropy alloy WMoTaNbV

    Plasma–facing components in future fusion reactors must endure high temperatures as well as high fluxes and fluences of high energy particles. Currently tungsten has been chosen as the primary plasma-facing material due to its good thermal conductivity, low erosion rate and low fuel retention. Materials with even better properties are still being investigated to be used in reactor regions with demanding plasma conditions. High entropy alloys (HEA) are a new class of metallic alloys and their exploitation in fusion applications has not been widely studied. In this work, the hydrogen isotope exchange effect in an equiatomic HEA containing W, Mo, Ta, Nb, and V was studied. Deuterium was implanted into HEA samples with 30 keV/D energy and the HEA and reference samples were annealed in H2 atmosphere and in vacuum at various temperatures up to 400 °C, respectively. The near-surface D concentration profiles were measured with ERDA and the isotope exchange was observed to remove over 90 % of the trapped deuterium from the implantation region at temperatures above 200 °C. TDS was used to measure retention deeper in the bulk in which the reduction of trapped deuterium was significantly lower. High total retention of H was found in the bulk after H2 atmosphere annealing which indicates permeation and deep trapping of H in the material.Plasma-facing components in future fusion reactors must endure high temperatures as well as high fluxes and fluences of high energy particles.
