    Is T. Vulgaris effective in preventing human breast adenocarcinoma cell metastasis?

    BACKGROUND AND AIM: The genus Thymus, belonging to thefamily Lamiaceae, has multiple biological and pharmacologicalproperties, including antimicrobial, antiseptic, antiviral and antifungal activities. This study aimed to determine the cytotoxic effectand metastatic potential of methanol/water phase extract preparedfrom Thymus vulgaris (TV) in human breast adenocarcinoma cellline (BT-474) by combining it with the chemotherapeutic agentpaclitaxel (PAC).MATERIALS and METHODS: The IC25 and IC50 concentrationsof TV and PAC in BT-474 cells were determined by the modifiedMTT method we developed in our laboratory.Cell migration asan indicator of metastatic potential; with in vitro wound healingmethod, colony formation potential was evaluated with SpheroidModel. The evaluation of the data was made with SPSS program.Conformity of the variables to the normal distribution ANOVA wasused to compare the groups.RESULTS: In wound healing assays, TV50 dose was found to bestatistically significant in “cell migration rate” measurements compared with treatment groups.P&lt;0,05. When the spheroidal formation in the 10-day 3D agarose gel is taken into consideration, there isno significant statistical difference between the PAC 50 and TS50groups, while in the combined group; spheroid formations largerthan the reference value were statistically significantly lower. Asignificant reduction in spheroidal formation was observed in combined (TV25+PAC25) and TV50 doses compared to the controlgroup. P&lt;0.05CONCLUSION: These findings are due to the fact that TV therapyin breast adenocarcinoma cells; especially when combined withPAC, it reveals that it may be effective in preventing tumor cellsfrom becoming spheroidal and tumor cell migration.Keywords: metastasis, migration, invasion, thymus vulgaris</p

The disease spreads locally and systemically in a very short time. It is resistant to almost all available cytostatics. There is an urgent need for chemicals with an effective, low toxicity profile. Since Docetaxel (DX) is partially effective in the current routine, it is used to prolong survival in patients with ATK. In this study, the antimetastatic property of tetrahydrocurcumin (THC), the active metabolite of Curcuma Longa, was compared with docetaxel and the presence of synergism in the combination of the two substances wasinvestigated in vitro. Materials-Methods: IC25 and IC50 concentrations of THC and (DX) in CAL62 and 8505C ATC cells were determined by the modified MTT method we developed in our laboratory. These 2 concentrations were compared with the control in the treatment groups. Cell motility as an indicator of metastatic potential was evaluated by in vitro wound healing method. Free oxygen radicals were evaluated by TAS and TOS analyses.Results: It was observed that cell adhesions decreased in 8505 cell lines starting from the 2nd hour in the treatment groups, more so in the groups combined with THC and curcumin. It was found that the combined dose prevented migration and invasion in CAL62 cells in the first hours (24) statistically, and the 72-hour IC25 dose did not have a statistically significant difference with the DX50 dose. There was no difference in Total Oxidant level between the combination and control groups. It was observed that the most effective dose that significantly reduced the oxidative level was DX50. Conclusions: THC in ATK cells has an antimetastatic potential close to Docetaxel; It was observed that docetaxel decreased oxidative stress.Keywords: Anaplastic thyroid cancer (ATC), Docetaxel (DX), tetrahydrocurcumin (THC), Total Antioksidan Status (TAS), Total Antioksidan Status (TOS) Amaç: Anaplastik tiroid kanseri (ATK) ortalama sağkalımbirkaç ayla sınırlı olduğu, bilinen en agresiv kanser türlerinden birisidir. Hastalık oldukça kısa sürede lokal ve sistemik yayılım yapmaktadır. Mevcut sitostatiklerin neredeyse tamamına dirençlidir. Etkin, düşük toksisite profiline sahip kimyasallara acil ihtiyaç bildirlmektedir. Güncel rutinde Docetaksel (DX) kısmen etkinlik gösterdiğinden, ATK lı olgularda sağkalımı uzatmak amacıyla kullanılmaktadır. Bu çalışmada Curcuma Longanın aktif metaboliti olan tetrahidrokurkuminin(THC) antimetastatik özelliği docetaksel ile karşılaştırmak ve iki madde kombinasyonunda sinerjizmin varlığı in vitro incelenmiştir.Gereç-Yöntem: CAL62 ve 8505C ATC hücrelerinde THC ve (DX) ait IC25 ve IC50 konsantrasyonları, laboratuvarımızda geliştirdiğimiz modifiye MTT yöntemi ile belirlendi. Bu 2 konsantrasyon, tedavi gruplarında kontrol ile karşılaştırıldı. Metastatik potansiyelin bir göstergesi olarak hücre motilitesi invitro yara iyileşme yöntemi ile değerlendirildi. Hücresel sitotoksiste TAS ve TOS analizleri ile değerlendirildi.Bulgular:8505 hücre hattında tedavi gruplarında 2. Saatten itibaren THC ve kurkuminle kombin gruplarda daha fazla olmak üzere hücre adezyonlarının azaldığı gözlendi.CAL62 hücrelerinde kombine dozun ilk saatlerde (24) migrasyonu ve invazyonu istatistiksel anlamlı engellediği, 72 sa lik IC25 dozunun DX50ndozu ile anlamlı istatistiksel farka sahip olmadığı bulgularına ulaşıldı.Total Oksidan seviyede kombinasyon ve kontrol grupları arasında farki izlenmedi.     Anti-Cancer Potential of Curcumin in Anaplastic Thyroid Cancer

    BACKGROUND AND AIM: Anaplastic Thyroid Cancer (ATK);It is one of the aggressive malignancies whose potential molecularmechanisms have not been elucidated and is the 9th among common cancers. It is resistant to existing cytostatic agents.To supportthe current therapy, the antimetastatic property of Tetrahydrocurcumin (THC) was compared with DX and in-vitro studies wereconducted to demonstrate synergism in combination use.MATERIALS and METHODS: IC25 and IC50 concentrations ofTHC and DX in CAL62 and 8505C cell lines were determined bythe modified MTT method we developed. Cell motility, indicative of metastatic potential; In-vitro wound healing method and 3Dcolony formation potential were evaluated with Agarose Gel Spheroid Model. The effects of treatments on oxidation on cells wereevaluated by TAS and TOS analyzes. SPSS program was used inthe evaluation. Compliance with normal distribution (Kolmogorov-Smirnov/Shapiro-Wilk tests), comparison of groups was evaluated with ANOVA, Significance, Holm-Sidak test.RESULTS: In migration analysis; It was observed that the THC50dose decreased the migration significantly compared to the DX50and DX+THC (Combined) groups (P&lt;0.05). DX+THC; It was observed that it reduced spheroid formation the most, and there was aclose effect between this dose effect and the THC25 dose in termsof spheroid formation and numbers (P&lt;0.05). There was no statistically significant difference in Total Oxidant level between thecombination and control.CONCLUSION: We predict that THC may be effective in metastasis and tumor growth at low doses in ATK cells.
