    The Protective Effect of Resveratrol on Cisplatin Induced Damage in Rat Liver

    Aim: One of the underlying causes of cisplatin-induced hepatotoxicity is oxidative stress. We assessed the effect of an antioxidant, resveratrol, on cisplatin-induced damage in the rat liver. Methods: The project-starting date was designed as 01.10.2020 and the project-ending date was planned as 01.04.2021. Three groups were created with 30 female Wistar-Albino rats: In group 1 (control group), 1 mL of 0.9% NaCl (saline) was administered intraperitoneally for 3 days. In group 2 (cisplatin group), 7.5 mg/kg intraperitoneal cisplatin was given for 3 days. In group 3 (cisplatin + resveratrol group), 7.5 mg/kg cisplatin and 10 mg/kg resveratrol were given via the intraperitoneal route. The livers were surgically extirpated in all the groups. In both blood and tissues, malondialdehyde (MDA) levels and activities of catalase (CAT) and superoxide dismutase (SOD) were measured. Also, toxicity markers such as hepatocyte damage (cellular changes), inflammation, hemorrhage, congestion, fibrosis, disorganization of the hepatic cords, and necrosis were assessed by examining the preparations prepared from hepatic tissue with light microscopy and immunohistochemistry. Results: Histopathological tissue damage was significantly higher in group 2 than in other groups (p 0.03). MDA levels were significantly higher and the activities of SOD and CAT were lower in group 2 than in the other groups (p=0.04 and p=0.01, respectively). Conclusion: According to our short-term findings, resveratrol might be an effective molecule for preventing the harmful effects of cisplatin in the rat liver. © 2022, Galenos Publishing House.     The effect of astaxanthin on amiodarone induced cardiac tissue damage in rat

    Purpose: The aim of this study was to evaluate the effect of astaxanthin on amiodarone induced cardiac tissue damage.Materials and Methods: 3 groups were formed using 30 Wistar albino rats. In group 1 (control group) (n=10), neither any drugs were given nor anything was performed. In group 2 (amiodarone group) (n=10), 100 mg/kg amiodarone was given for 7 days. In group 3 (amiodarone+astaxanthin group) (n=10), 100 mg/kg amiodarone and 25 mg/kg astaxanthin were given for 7 days. Hearts were surgically extirpated in all groups. Blood malondialdehyde (MDA) levels and activities of catalase (CAT) and superoxide dismutase (SOD) were measured. Also, toxicity markers such as edema, hemorrhage, fibrosis, inflammatory cell infiltration were assessed by examining the slides prepared from cardiac tissue with microscopy.Results: The MDA levels were significantly higher and the activities of SOD, and CAT were lower in group 2 than group 3. Tissue damage was significantly higher in group 2 than group 3.Conclusion: According to our short term findings, astaxanthin reversed the toxicity of amiodarone on cardiac tissue. In the light of these promising results, we suggest that astaxanthin usage should be thought to protect the cardiac damage due to amiodarone

Also, toxicity markers such as vascular congestion, hemorrhage, tubule degeneration and glomerular damage were assessed by examining the slides prepared from kidney tissue with microscopy.Results: Tissue damage was significantly higher in group 2 than other groups. The MDA levels were significantly higher and the activities of SOD, and CAT were lower in group 2 than other groups.Conclusion: According to our short term findings, resveratrol might be an effective molecule to prevent the harmful effect of cisplatin in rat kidney

    The Efficacy of Decreased Ovarian Reserve on in Vitro Fertilization Outcome

    Purpose: In this study, we aimed to compare the effects of microdose and antagonist regimens on IVF-ICSI outcome. Materials and Methods: The study was conducted at the Maltepe University Medical Faculty Training and Research Hospital IVF Clinic during the period of 2018 and 2022. The files of the patients were reviewed retrospectively and age, duration of infertility, basal FSH level, body mass index, antral follicle number (AFC), antimullerian hormon (AMH) level, retrieved oocyte number, metaphase 2 oocyte number, fertilization and pregnancy rates were evaluated. The patients in Group 1 (n=124) were treated by using gonadotropin releasing hormone (GnRH) microdose protocol and in Group 2 (n=136) were treated by using antagonist protocol. Results: There was no significant difference between the groups in terms of the characteristics of the patients such as age, infertility duration, basal FSH level body mass index, AFC and AMH level (p>0.05). The retrieved oocyte number, metaphase 2 oocyte number, fertilization rates and pregnancy rates were also similar (p>0.05). Conclusion: Our results demonstrated that microdose and antagonist regimens have similar effect in poor reponder women on IVF-ICSI outcome

    Evaluation of the Relationship Between Endometrial Thickness and IVF Success

    Purpose: The aim of this study was to evaluate the relationship between endometrial thickness measured on the day of embryo transfer and the results of in vitro fertilization-intracytoplasmic sperm injection (IVF-ICSI). Materials and Methods: 740 women who applied to Maltepe University Medical Faculty Training and Research Hospital IVF Clinic due to infertility were included in the study. The patients' age, infertility duration, basal follicle stimulating hormone (FSH) level, basal estradiol (E2) level and body mass index, fertilization and pregnancy rates were evaluated. Endometrial thickness was measured by transvaginal USG (TV USG) on the day of embryo transfer. A total of 4 groups were created. Group 1 included patients with endometrial thickness less than 7 mm. Group 2 included patients with endometrial thickness between 7-11 mm. Group 3 included patients with endometrial thickness between 11-14 mm and group 4 patients with endometrial thickness greater than 14 mm. Results: There was no significant difference between the groups in terms of age, infertility duration, basal FSH level, basal E2 level, anti mullerian hormon (AMH) and body mass index. The clinical pregnancy rates in group 1 were significantly lower than the other groups (p<0.05). There was no significant difference between the groups in terms of the number of oocytes retrieved or fertilization rates. Conclusion: Having an endometrial thickness of less than 7 mm on the day of transfer significantly reduces pregnancy rates

    Effect of thermal and mechanical load cycling on dentin bond strength of a self-etch resin luting cement

    Amaç: Bu çalışmanın amacı, termal ve mekanik yükleme döngülerinin, bir self – etch rezin yapıştırma simanının dentine bağlanma dayanımını in vitro şartlar altında değerlendirmektir.Gereç ve Yöntemler: Çürüksüz, 18 adet mandibular üçüncü molar diş üzerinde inlay kaviteleri hazırlandı. Restorasyonlar, feldspatik porselen kullanılarak hazırlandı (Vita VM9). Dentine bağlantı bir dual- cure, self- etch rezin yapıştırma simanı (Clearfil Esthetic Cement) kullanılarak sağlandı. Sonrasında dişler rastgele üç gruba ayrıldı: Grup I: Kontrol Grubu (termal veya mekanik siklus yok). Grup II: Termal siklus (5.000 termal siklus). Grup III: Mekanik yükleme siklusu (1.200.000 siklus). Herbir dişten (n:12) uzun eksen doğrultusunda kesiler yapılarak ikişer adet I-şeklinde kesitler elde edildi ve toplamda otuz altı adet örneğe, 1 mm/dk hızla çekme kuvvetleri uygulanarak kırılma sırasındaki maksimum kuvvet ölçüldü. Kırık hattı, ayrılma tipini tanımlamak amacı ile stereomikroskop (Olympus, SZ-PT) ile incelendi. Her bir simantasyon grubundan bir diş SEM (435 VP; Leo SEM Products) analizleri yapılmak üzere hazırlanarak arayüzeyler SEM altında incelendi.Bulgular: One-way ANOVA ile gruplar arasında anlamlı farklılıklar olduğu ortaya konuldu (p&lt;0.05). Tukey’s HSD analizi, Grup III’te görülen ortalama MTBS değerinin Grup I ve Grup II’den anlamlı derecede düşük olduğunu ortaya koydu (p&lt;0.05). Grup I ve Grup II arasında istatistiksel olarak anlamlı fark görülmemiştir (p&gt;0.05).Sonuç: Bu çalışmanın sonuçlarına bakılarak, tüm gruplar için gözlenen asıl ayrılma tipinin interfasiyal (adeziv) tip olduğu söylenebilir. Ayrıca, sıcaklık değişimleri önemli derece etkili değil iken, mekanik yüklemenin bağlanma stabilitesi üzerine etkili olduğu görülmektedir

    Regio- and stereo-chemical ring-opening reactions of the 2,3-epoxy alcohol derivative with nucleophiles: Explanation of the structures and C-2 selectivity supported by theoretical computations

    The ring-opening reactions of (1aS,2S,6bR)-5-ethyl-2-hydroxyhexahydro-4H-oxireno[2,3-e]isoindole-4,6(5H)-dione were investigated under very mild and nonchelated conditions. C-2 selective ring-opening products were obtained with nucleophilic additions such as Cl-, Br- and N-3(-). The exact configuration of (3aS,4R,5R,6S,7aS)-5-chloro-2-ethyl-4,6-dihydroxyhexahydro-1H-isoindole-1,3(2H)-dione was determined by X-Ray diffraction analysis which was obtained from the reaction of epoxy alcohol with HCl . On the other hand, theoretical computations were carried out to explain the regioselectivity in the ring opening reaction of epoxy alcohols. The results showed that the ring-opening reaction of both epoxy alcohols proceeds in a kinetically controlled manner and regioselectivity occurs depending on the transition state.     Seramik restorasyonların mikrosızıntısı üzerine yüksek yoğunluklu LED işık kaynağı mesafesinin değerlendirilmesi

    Amaç: Rezin simanla yapıştırılmış IPS e.max Press slot kavite restorasyonlarının; iki farklı yüksek yoğunluklu LED ışık kaynağı ile ve 0 ve 9 mm uzaklıktan mikrosızıntılarının değerlendirilmesidir. Gereç ve Yöntemler: Bu çalışmada 48 adet çekilmiş insan premoları kullanılmıştır. Dişlerin aproksimal yüzeylerinde inley preparasyon seti (Komet, Germany) kullanılarak slot kaviteler oluşturulmuştur. IPS e.max Press (Ivoclar Vivadent AG, Liechtenstein) seramik inley restorasyonlar hazırlanmıştır. İki farklı LED ışık kaynağı yüksek güç yoğunluklu LED (HPIL; Demi Ultra) ve yüksek yoğunluklu LED (HIL; Valo Cordless) dual cure rezin simanı polimerize etmek için kullanılmıştır (NX3 Nexus, Kerr). Işık kaynağının ucu ile restorasyon arasındaki mesafe 0 ve 9 mm olarak ayarlanıp ve plastik halka kullanılarak kontrol edilmiştir. Dişler rastgele olarak 4 gruba ayrılmıştır. Grup 1: HPIL ışık kaynağı ile restorasyon arasında mesafe yok, Grup 2: HIL ışık kaynağı ile restorasyon arasında mesafe yok, Grup 3: HPIL şık kaynağı ile restorasyon arasında 9 mm mesafe bulunmakta, Grup 4: HIL ışık kaynağı ile restorasyon arasında 9 mm mesafe bulunmaktadır. Polimerizasyon işleminden sonra, örnekler 5 ve 55 ˚C arasında, her bir haznede 30 s kalacak şekilde 5000 termal siklusa tabi tutulmuştur. Örnekler tırnak cilasi ile kaplanmış, % 0.5’lik bazik fuksinde 24 saat bekletilmiş, dişler ikiye bölünmüş, stereomikroskop altında incelenmiş ve gingival marjindeki mikrosızıntı skorlanmıştır. Kruskal-Wallis ve Mann-Whitney U-tests kullanılarak istatistiksel analiz yapılmıştır. Bulgular: Çalışmanın sonucunda grup 1, 2 ve 3 arasında istatiksel bir fark gözlenmezken (p &gt; 0.05), grup 4’ün mikrosızıntı değeri diğer tüm gruplardan daha yüksek bir değer göstermiştir (p &lt; 0.05). Sonuçlar: Restorasyon yüzeyinden, HIL ışık kaynağının mesafesinin artması diş ve restorasyon arasında mikrosızıntının artmasıyla sonuçlanmıştır
