    Overview of interpretive modelling of fusion performance in JET DTE2 discharges with TRANSP

    In the paper we present an overview of interpretive modelling of a database of JET-ILW 2021 D-T discharges using the TRANSP code. The main aim is to assess our capability of computationally reproducing the fusion performance of various D-T plasma scenarios using different external heating and D-T mixtures, and to understand the performance driving mechanisms. We find that interpretive simulations confirm a general power-law relationship between increasing external heating power and fusion output, which is supported by absolutely calibrated neutron yield measurements. A comparison of measured and computed D-T neutron rates shows that the calculations' discrepancy depends on the absolute neutron yield. The calculations are found to agree well with measurements for higher performing discharges with external heating power above ∼20 MW\mathrm{MW}, while low-neutron shots display an average discrepancy of around +40% compared to measured neutron yields. A similar trend is found for the ratio between thermal and beam-target fusion, where larger discrepancies are seen in shots with dominant beam-driven performance. We compare the observations to studies of JET-ILW D discharges, to find that on average the fusion performance is well modelled over a range of heating power, although an increased unsystematic deviation for lower-performing shots is observed. The ratio between thermal and beam-induced D-T fusion is found to be increasing weakly with growing external heating power, with a maximum value of \gtrsim1 achieved in a baseline scenario experiment. An evaluation of the fusion power computational uncertainty shows a strong dependence on the plasma scenario type and fusion drive characteristics, varying between ±25% and 35%. D-T fusion alpha simulations show that the ratio between volume-integrated electron and ion heating from alphas is \lesssim10 for the majority of analysed discharges. Alphas are computed to contribute between ∼15% and 40% to the total electron heating in the core of highest performing D-T discharges. An alternative workflow to TRANSP was employed to model JET D-T plasmas with the highest fusion yield and dominant non-thermal fusion component because of the use of fundamental radio-frequency heating of a large minority in the scenario, which is calculated to have provided ∼10% to the total fusion power.This work has been carried out within the framework of the EUROfusion Consortium, funded by the European Union via the Euratom Research and Training Programme (Grant Agreement No. 101052200—EUROfusion).     Ameliorating effects of Mango (Mangifera indica L.) fruit on plasma ethanol level in a mouse model assessed with 1H-NMR based metabolic profiling

    The ameliorating effects of Mango (Mangifera indica L.) flesh and peel samples on plasma ethanol level were investigated using a mouse model. Mango fruit samples remarkably decreased mouse plasma ethanol levels and increased the activities of alcohol dehydrogenase and acetaldehyde dehydrogenase. The 1H-NMR-based metabolomic technique was employed to investigate the differences in metabolic profiles of mango fruits, and mouse plasma samples fed with mango fruit samples. The partial least squares-discriminate analysis of 1H-NMR spectral data of mouse plasma demonstrated that there were clear separations among plasma samples from mice fed with buffer, mango flesh and peel. A loading plot demonstrated that metabolites from mango fruit, such as fructose and aspartate, might stimulate alcohol degradation enzymes. This study suggests that mango flesh and peel could be used as resources for functional foods intended to decrease plasma ethanol level after ethanol uptake

    Small-Molecule Hydrophobic Tagging Induced Degradation of HaloTag Fusion Proteins

    The ability to regulate any protein of interest in living systems with small molecules remains a challenge. We hypothesized that appending a hydrophobic moiety to the surface of a protein would mimic the partially denatured state of the protein, thus engaging the cellular quality control machinery to induce its proteasomal degradation. We designed and synthesized bifunctional small molecules to bind a bacterial dehalogenase (the HaloTag protein) and present a hydrophobic group on its surface. Hydrophobic tagging of the HaloTag protein with an adamantyl moiety induced the degradation of cytosolic, isoprenylated and transmembrane HaloTag fusion proteins in cell culture. We demonstrated the in vivo utility of hydrophobic tagging by degrading proteins expressed in zebrafish embryos and by inhibiting Hras1G12V-driven tumor progression in mice. Therefore, hydrophobic tagging of HaloTag fusion proteins affords small-molecule control over any protein of interest, making it an ideal system for validating potential drug targets in disease models

    We obtain (3+3)- or (3+2)-dimensional global flat embeddings of four uncharged and charged scalar-tensor theories with the parameters B or L in the (2+1)-dimensions, which are the non-trivially modified versions of the Banados-Teitelboim-Zanelli (BTZ) black holes.     Epidemiological characteristics of ovarian cancer in Korea

    Objective: This study was conducted to examine recent trends in ovarian cancer incidence and mortality and secular trends in demographic factors in Korea. Methods: With the data from Korea Central Cancer Registry, International Agency for Research on Cancer, Korean Death Registry, and World Health Organization`s Statistical Information System, we calculated age-standardized incidence and mortality rates for ovarian cancer. Also we estimated future incidence of ovarian and cervical cancer using linear regression model. To assess the demographic trend, data from national surveys in Korea or results from published papers were searched. Results: Ovarian cancer incidence rate was similar to that in women worldwide but lower than those in Western countries, and the trend has been increased steadily. Ovarian cancer-related mortality rates have been increasing in Korea, even though those in western and some Asian countries, such as China, have been decreasing. Age-specific incidence rate and mortality rate showed steep increases with advancing age. The incidence rate of ovarian cancer was estimated to surpass that of uterine cervix cancer in 2015. Korea showed rapid changes in nutritional, reproductive, and anthropometric factors. Conclusion: These recent trends in ovarian cancer incidence and mortality may be partly attributed to gradual westernizing of life styles and to changes in socio-demographic behavior factors.     OGLE-2016-BLG-1227L: A Wide-separation Planet from a Very Short-timescale Microlensing Event

    We present the analysis of the microlensing event OGLE-2016-BLG-1227. The light curve of this short-duration event appears to be a single-lens event affected by severe finite-source effects. Analysis of the light curve based on single-lens single-source (1L1S) modeling yields very small values of the event timescale, t_E ∼ 3.5 days, and the angular Einstein radius, θ_E ∼ 0.009 mas, making the lens a candidate of a free-floating planet. Close inspection reveals that the 1L1S solution leaves small residuals with amplitude ΔI ≲ 0.03 mag. We find that the residuals are explained by the existence of an additional widely-separated heavier lens component, indicating that the lens is a wide-separation planetary system rather than a free-floating planet. From Bayesian analysis, it is estimated that the planet has a mass of _p = 0.79^(+1.30)_(−0.39) M_J and it is orbiting a low-mass host star with a mass of M_(host) = 0.10+0.17−0.05 M_⊙ located with a projected separation of a_ = 3.4^(+2.1)_(−1.0) au. The planetary system is located in the Galactic bulge with a line-of-sight separation from the source star of D_(LS) = 1.21^(+0.96)_(−0.63) kpc. The event shows that there are a range of deviations in the signatures of host stars for apparently isolated planetary lensing events and that it is possible to identify a host even when a deviation is subtle

    Changes of fat-mass and obesity-associated protein expression in the hippocampus in animal models of high-fat diet-induced obesity and D-galactose-induced aging

    Abstract Fat-mass and obesity-associated protein (Fto) is highly expressed in the brain including, the hippocampus, and its expression is significantly decreased in the brain of Alzheimers disease patients. In the present study, we measured Fto immunoreactivity and protein levels in the hippocampus of obese and aged mice, which were induced by high-fat diet for 12 weeks and D-galactose treatment for 10 weeks, respectively. The obesity and aging phenotypes were assessed by physiological parameters and Morris water maze test, respectively. High fat diet fed mice showed significant increases in body weight and blood glucose levels compared to that in the control or D-galactose-induced aged mice. In addition, treatment with D-galactose significantly decreased the spatial memory. Fto immunoreactivity in the control group was mainly detected in the pyramidal cells of the CA1 and CA3 regions and in the granule cells of the dentate gyrus. In the hippocampus of high-fat diet-fed mice, Fto immunoreactive structures were similarly found in the hippocampus compared to that in the control group, but Fto immunoreactivity in high-fat diet-fed mice was also found in the stratum oriens and radiatum of the CA1 and CA3 regions and the polymorphic layer of the dentate gyrus. In the hippocampus of D-galactose-induced aged mice, fewer Fto immunoreactive structures were detected in the granule cell layer of the dentate gyrus compared to the control group. Fto mRNA and protein levels based on quantitative real-time polymerase chain reaction and western blot assays were slightly increased in the hippocampus of high-fat diet-fed mice compared to that in control mice. In addition, Fto mRNA and protein levels were significantly decreased in the aged hippocampus compared to that in the control group. Fto protein levels are susceptible to the aging process, but not in the hippocampus of high-fat diet-induced obesity. The reduction of Fto in aged mice may be associated with reduced memory impairment in mice
