    Activity and expression of urokinase-type plasminogen activator and matrix metalloproteinases in human colorectal cancer

    BACKGROUND: Matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9), and urokinase-type plasminogen activator (uPA) are involved in colorectal cancer invasion and metastasis. There is still debate whether the activity of MMP-2 and MMP-9 differs between tumors located in the colon and rectum. We designed this study to determine any differences in the expression of MMP-2, MMP-9 and uPA system between colon and rectal cancer tissues. METHODS: Cancer tissue samples were obtained from colon carcinoma (n = 12) and rectal carcinomas (n = 10). MMP-2 and MMP-9 levels were examined using gelatin zymography and Western blotting; their endogenous inhibitors, tissue inhibitor of metalloproteinase-2 (TIMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1), were assessed by Western blotting. uPA, uPAR and PAI-1 were examined using enzyme-linked immunosorbent assay (ELISA). The activity of uPA was assessed by casein-plasminogen zymography. RESULTS: In both colon and rectal tumors, MMP-2, MMP-9 and TIMP-1 protein levels were higher than in corresponding paired normal mucosa, while TIMP-2 level in tumors was significantly lower than in normal mucosa. The enzyme activities or protein levels of MMP-2, MMP-9 and their endogenous inhibitors did not reach a statistically significant difference between colon and rectal cancer compared with their normal mucosa. In rectal tumors, there was an increased activity of uPA compared with the activity in colon tumors (P = 0.0266), however urokinase-type plasminogen activator receptor (uPAR) and plasminogen activator inhibitor-1 (PAI-1) showed no significant difference between colon and rectal cancer tissues. CONCLUSION: These findings suggest that uPA may be expressed differentially in colon and rectal cancers, however, the activities or protein levels of MMP-2, MMP-9, TIMP-1, TIMP-2, PAI-1 and uPAR are not affected by tumor location in the colon or the rectum

    The Genetic Effect of Copy Number Variations on the Risk of Type 2 Diabetes in a Korean Population

    BACKGROUND: Unlike Caucasian populations, genetic factors contributing to the risk of type 2 diabetes mellitus (T2DM) are not well studied in Asian populations. In light of this, and the fact that copy number variation (CNV) is emerging as a new way to understand human genomic variation, the objective of this study was to identify type 2 diabetes-associated CNV in a Korean cohort. METHODOLOGY/PRINCIPAL FINDINGS: Using the Illumina HumanHap300 BeadChip (317,503 markers), genome-wide genotyping was performed to obtain signal and allelic intensities from 275 patients with type 2 diabetes mellitus (T2DM) and 496 nondiabetic subjects (Total n = 771). To increase the sensitivity of CNV identification, we incorporated multiple factors using PennCNV, a program that is based on the hidden Markov model (HMM). To assess the genetic effect of CNV on T2DM, a multivariate logistic regression model controlling for age and gender was used. We identified a total of 7,478 CNVs (average of 9.7 CNVs per individual) and 2,554 CNV regions (CNVRs; 164 common CNVRs for frequency>1%) in this study. Although we failed to demonstrate robust associations between CNVs and the risk of T2DM, our results revealed a putative association between several CNVRs including chr15:45994758-45999227 (P = 8.6E-04, P(corr) = 0.01) and the risk of T2DM. The identified CNVs in this study were validated using overlapping analysis with the Database of Genomic Variants (DGV; 71.7% overlap), and quantitative PCR (qPCR). The identified variations, which encompassed functional genes, were significantly enriched in the cellular part, in the membrane-bound organelle, in the development process, in cell communication, in signal transduction, and in biological regulation. CONCLUSION/SIGNIFICANCE: We expect that the methods and findings in this study will contribute in particular to genome studies of Asian populations

    Gender discriminations among young children in Asia

    Subsequent to the demographic transition, Asian countries have been experiencing deep-rooted changes in family structures. In this context, the question of gender relations within the family, and more generally within society, is crucial, in view of the increase in discriminatory practices toward women, beginning at foetal conception and continuing through all stages of life. Asia is the “black continent” for women. Estimates place the deficit in the number of women in the world at between 60 and 100 million, the vast majority of which is found on this continent. This book focuses on the intensity of female discrimination, from a demographic perspective, in the earliest stages of life, and more specifically around birth, in China, India, Pakistan, the Republic of Korea and Taiwan. These societies share cultural characteristics that are not favourable to women: patriarchal systems, patrilineal families, socialization processes encouraging the submission of wives to their husband's family, etc. In these societies, a son is needed to perpetuate the family line and ensure social and biological reproduction of the family. These are among the reasons why they share a strong son preference, which is in some cases accentuated by economic constraints. A son is generally the only person to support his parents in old age, and as a rule help with work in the fields. Moreover, girls and women still occupy a marginal position in society, whereas a male heir offers countless advantages.La transition démographique suscite, en Asie, de profondes transformations dans les structures familiales. Dans ce contexte, la question des rapports sociaux des sexes au sein de la famille, et plus généralement au sein de la société, est cruciale, dans la mesure où elle entraîne l’augmentation des discriminations à l’égard des femmes dès la conception et à tous les âges de la vie. L’Asie est le “continent noir” pour les femmes. Ce livre traite, par une approche démographique, des discriminations des femmes aux jeunes âges de la vie, et en particulier autour de la naissance, en Chine, en Inde, au Pakistan, en République de Corée et à Taiwan. Ces sociétés d’Asie partagent des caractéristiques culturelles qui ne sont pas favorables aux femmes ; un fils est désiré pour perpétuer la lignée et assurer la reproduction biologique et sociale de la famille.     Trans-ethnic meta-analysis of genome-wide association studies for Hirschsprung disease.

    Hirschsprung disease (HSCR) is the most common cause of neonatal intestinal obstruction. It is characterized by the absence of ganglia in the nerve plexuses of the lower gastrointestinal tract. So far, three common disease-susceptibility variants at the RET, SEMA3 and NRG1 loci have been detected through genome-wide association studies (GWAS) in Europeans and Asians to understand its genetic etiologies. Here we present a trans-ethnic meta-analysis of 507 HSCR cases and 1191 controls, combining all published GWAS results on HSCR to fine-map these loci and narrow down the putatively causal variants to 99% credible sets. We also demonstrate that the effects of RET and NRG1 are universal across European and Asian ancestries. In contrast, we detected a European-specific association of a low-frequency variant, rs80227144, in SEMA3 [odds ratio (OR) = 5.2, P = 4.7 × 10-10]. Conditional analyses on the lead SNPs revealed a secondary association signal, corresponding to an Asian-specific, low-frequency missense variant encoding RET p.Asp489Asn (rs9282834, conditional OR = 20.3, conditional P = 4.1 × 10-14). When in trans with the RET intron 1 enhancer risk allele, rs9282834 increases the risk of HSCR from 1.1 to 26.7. Overall, our study provides further insights into the genetic architecture of HSCR and has profound implications for future study designs
