    The Role of Gut Microbiota in Atherosclerosis and Hypertension

    In recent years, accumulating evidence has indicated the importance of gut microbiota in maintaining human health. Gut dysbiosis is associated with the pathogenesis of a number of metabolic diseases including obesity, type 2 diabetes mellitus (T2DM), non-alcoholic fatty liver disease (NAFLD), and cardiovascular diseases (CVDs). Indeed, CVD has become the leading cause of death worldwide, especially in developed countries. In this review, we mainly discuss the gut microbiota-involved mechanisms of CVD focusing on atherosclerosis and hypertension, two major risk factors for serious CVD. Then, we briefly discuss the prospects of gut microbiota-targeted therapeutic strategies for the treatment of CVD in the future

    Study on cosmogenic activation in copper for rare event search experiments

    The rare event search experiments using germanium detectors are performed in the underground laboratories to prevent cosmic rays. However, the cosmogenic activation of the cupreous detector components on the ground will generate long half-life radioisotopes and contribute continually to the expected background level. We present a study on the cosmogenic activation of copper after 504 days of exposure at an altitude of 2469.4 m outside the China Jinping Underground Laboratory (CJPL). The specific activities of the cosmogenic nuclides produced in the copper bricks were measured using a low background germanium gamma-ray spectrometer at CJPL. The production rates at sea level, in units of nuclei/kg/day, are 18.6 \pm 2.0 for Mn-54, 9.9 \pm 1.3 for Co-56, 48.3 \pm 5.5 for Co-57, 51.8 \pm 2.5 for Co-58 and 39.7 \pm 5.7 for Co-60, respectively. Given the expected exposure history of the germanium detectors, a Monte Carlo simulation is conducted to assess the cosmogenic background contributions of the detectors' cupreous components.Comment: 6 pages, 4 figure

    The Theory and Practice of Ageing Housing Transformation in China and Abroad: A Review

    This paper investigates the theoretical research and practical exploration in housing the ageing in western developed countries and China after France first entered the ageing society. The subject of housing the ageing was reviewed using the methods of literature search and induction analysis. In terms of practical exploration, three basic models of ageing adaptation in developed countries were extracted: public welfare policy, nursing insurance system, and market economy. The objectives, objects, contents, advantages, and disadvantages of the transformation were analysed; the practical exploration and problems encountered in China's pilot cities for ageing adaptation were summarized. Through comparative analysis, it is proposed that we should learn from the advanced experience of western countries to avoid their shortcomings. It is also suggested that the transformation of the people, the content, and the processes to be adapted to suit the local conditions in China

    An Intragenic SRF-Dependent Regulatory Motif Directs Cardiac-Specific microRNA-1-1/133a-2 Expression

    Transcriptional regulation is essential for any gene expression including microRNA expression. MiR-1-1 and miR-133a-2 are essential microRNAs (miRs) involved in cardiac and skeletal muscle development and diseases. Early studies reveal two regulatory enhancers, an upstream and an intragenic, that direct the miR-1-1 and miR-133a-2 transcripts. In this study, we identify a unique serum response factor (SRF) binding motif within the enhancer through bioinformatic approaches. This motif is evolutionarily conserved and is present in a range of organisms from yeast, flies, to humans. We provide evidence to demonstrate that this regulatory motif is SRF-dependent in vitro by electrophoretic mobility shift assay, luciferase activity assay, and endogenous chromatin immunoprecipitation assay followed by DNA sequence confirmation, and in vivo by transgenic lacZ reporter mouse studies. Importantly, our transgenic mice indicate that this motif is indispensable for the expression of miR1-1/133a-2 in the heart, but not necessary in skeletal muscle, while the enhancer is sufficient for miR1-1/133a-2 gene expression in both tissues. The mutation of the motif alone completely abolishes miR-1-1/133a-2 gene expression in the animal heart, but not in the skeletal muscle. Our findings reveal an additional architecture of regulatory complex directing miR-1-1/133a-1 gene expression, and demonstrate how this intragenic enhancer differentially manages the expression of the two miRs in the heart and skeletal muscle, respectively

    Integrated Metagenomic and Transcriptomic Analyses Reveal the Dietary Dependent Recovery of Host Metabolism From Antibiotic Exposure

    The balance of gut microbiome is essential for maintaining host metabolism homeostasis. Despite widespread antibiotic use, the potential long-term detrimental consequences of antibiotics for host health are getting more and more attention. However, it remains unclear whether diet affects the post-antibiotic recovery of gut microbiome and host metabolism. In this study, through metagenomic sequencing and hepatic transcriptome analysis, we investigated the divergent impacts of short-term vancomycin (Vac), or combination of ciprofloxacin and metronidazole (CM) treatment on gut microbiome and host metabolism, as well as their recovery extent from antibiotic exposure on chow diet (CD) and high-fat diet (HFD). Our results showed that short-term Vac intervention affected insulin signaling, while CM induced more functional changes in the microbiome. However, Vac-induced long-term (45 days) changes of species were more apparent when recovered on CD than HFD. The effects of antibiotic intervention on host metabolism were long-lasting, antibiotic-specific, and diet-dependent. The number of differentially expressed gene was doubled by Vac than CM, but was comparable after recovery on CD as revealed by the hepatic transcriptomic analysis. In contrast, HFD intake during recovery could worsen the extent of post-antibiotic recovery by altering infection, immunity, and cancer-related pathways in short-term Vac-exposed rats and by shifting endocrine system-associated pathways in CM-exposed rats. Together, the presented data demonstrated the long-term recovery extent after different antibiotic exposure was diet-related, highlighting the importance of dietary management during post-antibiotic recovery

    Global genome expression analysis of rice in response to drought and high-salinity stresses in shoot, flag leaf, and panicle

    To elucidate genome-level responses to drought and high-salinity stress in rice, a 70mer oligomer microarray covering 36,926 unique genes or gene models was used to profile genome expression changes in rice shoot, flag leaf and panicle under drought or high-salinity conditions. While patterns of gene expression in response to drought or high-salinity stress within a particular organ type showed significant overlap, comparison of expression profiles among different organs showed largely organ-specific patterns of regulation. Moreover, both stresses appear to alter the expression patterns of a significant number of genes involved in transcription and cell signaling in a largely organ-specific manner. The promoter regions of genes induced by both stresses or induced by one stress in more than one organ types possess relative enrichment of two cis-elements (ABRE core and DRE core) known to be associated with water stress. An initial computational analysis indicated that novel promoter motifs are present in the promoters of genes involved in rehydration after drought. This analysis suggested that rice might possess a mechanism that actively detects rehydration and facilitates rapid recovery. Overall, our data supports a notion that organ-specific gene regulation in response to the two abiotic stresses may primarily be mediated by organ-specific transcription responses.
