    The increasing diversity of youth in the United States necessitates a shift in the ways that youth services and programming are designed and implemented. This article examines existing scholarship on developing the cultural competency of volunteers in youth development programs in an effort to improve 4-H YDP protocol. Drawing from a diverse, interdisciplinary range of peer-reviewed, academic articles, this literature review plots out recent pedagogical trends, theoretical concepts, and empirical studies dealing with the cultural competence of service workers and mentors interacting with youth. Based on a synthesis of the findings, this paper presents guiding principles for increasing cultural competence of youth program design through both training and organizational changes

    Hydrogeology and drilling phase 1 for Scott Creek catchment

    Scott Creek Catchment is the first of a number of catchments that will be used as case studies to investigate the sustainability of groundwater resources in the Mount Lofty Ranges over the next 45 years. This report provides a collation of background information for the Scott Creek Catchment including geological, hydrological, meteorological and surface water quality data. Site selection criteria, drilling methods, construction details and lithological logs are presented for the first phase of drilling in this catchment. A total of nine wells (one completed in the Quaternary alluvium and eight in the fractured Woolshed Flat Shale) were drilled at strategic locations on either side of Scott Creek upstream of the weir at Scott Bottom. These wells will be used for a variety of hydraulic and hydrochemical tests to define the local hydrogeology in terms of streamaquifer interactions and groundwater recharge and flow rates.repor

    Impact of DNA ligase IV on the fidelity of end joining in human cells

    A DNA ligase IV (LIG4)‐null human pre‐B cell line and human cell lines with hypomorphic mutations in LIG4 are significantly impaired in the frequency and fidelity of end joining using an in vivo plasmid assay. Analysis of the null line demonstrates the existence of an error‐prone DNA ligase IV‐independent rejoining mechanism in mammalian cells. Analysis of lines with hypomorphic mutations demonstrates that residual DNA ligase IV activity, which is sufficient to promote efficient end joining, nevertheless can result in decreased fidelity of rejoining. Thus, DNA ligase IV is an important factor influencing the fidelity of end joining in vivo. The LIG4‐defective cell lines also showed impaired end joining in an in vitro assay using cell‐free extracts. Elevated degradation of the terminal nucleotide was observed in a LIG4‐defective line, and addition of the DNA ligase IV–XRCC4 complex restored end protection. End protection by DNA ligase IV was not dependent upon ligation. Finally, using purified proteins, we demonstrate that DNA ligase IV–XRCC4 is able to protect DNA ends from degradation by T7 exonuclease. Thus, the ability of DNA ligase IV–XRCC4 to protect DNA ends may contribute to the ability of DNA ligase IV to promote accurate rejoining in vivo

    Testing galaxy formation models with the GHOSTS survey: The color profile of M81's stellar halo

    We study the properties of the stellar populations in M81's outermost part, which hereafter we will term the stellar halo, using HST ACS/WFC observations of 19 fields from the GHOSTS survey. The observed fields probe the stellar halo out to a projected distance of ~ 50 kpc from the galactic center. Each field was observed in both F606W and F814W filters. The 50% completeness levels of the color magnitude diagrams (CMDs) are typically at 2 mag below the tip of the red giant branch (TRGB). Fields at distances closer than 15 kpc show evidence of disk-dominated populations whereas fields at larger distances are mostly populated by halo stars. The RGB of the M81's halo CMDs is well matched with isochrones of ~ 10 Gyr and metallicities [Fe/H] ~ -1.2 dex, suggesting that the dominant stellar population of M81's halo has a similar age and metallicity. The halo of M81 is characterized by a color distribution of width ~ 0.4 mag and an approximately constant median value of (F606W - F814W) ~ 1 mag measured using stars within the magnitude range 23.7 < F814W < 25.5. When considering only fields located at galactocentric radius R > 15 kpc, we detect no color gradient in the stellar halo of M81. We place a limit of 0.03+/-0.11 mag difference between the median color of RGB M81 halo stars at ~ 15 and at 50 kpc, corresponding to a metallicity difference of 0.08+/-0.35 dex over that radial range for an assumed constant age of 10 Gyr. We compare these results with model predictions for the colors of stellar halos formed purely via accretion of satellite galaxies. When we analyze the cosmologically motivated models in the same way as the HST data, we find that they predict no color gradient for the stellar halos, in good agreement with the observations.Comment: 16 pages, 7 figures.     Vaccinia virus protein A46R targets multiple Toll-like-interleukin-1 receptor adaptors and contributes to virulence

    Viral immune evasion strategies target key aspects of the host antiviral response. Recently, it has been recognized that Toll-like receptors (TLRs) have a role in innate defense against viruses. Here, we define the function of the vaccinia virus (VV) protein A46R and show it inhibits intracellular signalling by a range of TLRs. TLR signalling is triggered by homotypic interactions between the Toll-like-interleukin-1 resistance (TIR) domains of the receptors and adaptor molecules. A46R contains a TIR domain and is the only viral TIR domain-containing protein identified to date. We demonstrate that A46R targets the host TIR adaptors myeloid differentiation factor 88 (MyD88), MyD88 adaptor-like, TIR domain-containing adaptor inducing IFN-beta (TRIF), and the TRIF-related adaptor molecule and thereby interferes with downstream activation of mitogen-activated protein kinases and nuclear factor kappaB. TRIF mediates activation of interferon (IFN) regulatory factor 3 (IRF3) and induction of IFN-beta by TLR3 and TLR4 and suppresses VV replication in macrophages. Here, A46R disrupted TRIF-induced IRF3 activation and induction of the TRIF-dependent gene regulated on activation, normal T cell expressed and secreted. Furthermore, we show that A46R is functionally distinct from another described VV TLR inhibitor, A52R. Importantly, VV lacking the A46R gene was attenuated in a murine intranasal model, demonstrating the importance of A46R for VV virulence

    Advancing loneliness and social isolation as global health challenges: taking three priority actions

    Loneliness and social isolation have been identified as critical global health issues in the aftermath of the coronavirus disease 2019 (COVID-19) crisis. While there is robust scientific evidence demonstrating the impact of loneliness and social isolation on health outcomes and mortality, there are fundamental issues to resolve so that health authorities, decision makers, and practitioners worldwide are informed and aligned with the latest evidence. Three priority actions are posited to achieve a wider and more substantial impact on loneliness and social isolation. They are 1) strengthening the evidence base; 2) adopting a whole-of-systems approach; 3) developing policy support for governments worldwide. These priority actions are essential to reduce the pervasive impact of loneliness and social isolation as social determinants of health

    Constraining the age of the NGC 4565 HI Disk Warp: Determining the Origin of Gas Warps

    We have mapped the distribution of young and old stars in the gaseous HI warp of NGC 4565. We find a clear correlation of young stars (<600 Myr) with the warp, but no coincident old stars (>1 Gyr), which places an upper limit on the age of the structure. The formation rate of the young stars, which increased ~300 Myr ago relative to the surrounding regions, is (6.3 +2.5/-1.5) x 10^-5 M_sol/yr/kpc^2. This implies a ~60+/-20 Gyr depletion time of the HI warp, similar to the timescales calculated for the outer HI disks of nearby spiral galaxies. While some stars associated with the warp fall into the asymptotic giant branch (AGB) region of the color magnitude diagram, where stars could be as old as 1 Gyr, further investigation suggests that they may be interlopers rather than real AGB stars.
