    The effect of Coriandrum sativum seed extract on the learning of newborn mice by electric shock: interaction with caffeine and diazepam

    Coriander has been recommended for the relief of pain, anxiety, flatulence, and loss of appetite. In traditional medicine, it is believed that coriander can induce some degree of amnesia in a child when his/her mother uses coriander during the pregnancy. We evaluated the effect of Coriandrum sativum seed extract on learning in second-generation mice. Ethanolic extract (2%) of coriander (100 mg/kg intraperitoneal) was dissolved in sunflower oil (oil) as a vehicle and injected into the control group mother mice during breastfeeding for 25 days at 5-day intervals. After feeding the newborn mice, their learning was evaluated using a step-through passive avoidance task with 0.4 mA electric shock for 2 or 4 seconds. While coriander extract showed a negative effect in the short term (1 hour) after the training session, it potentiated the mice’s learning in later assessments (24 hours post-training [P = 0.022] and 1 week post-training [P = 0.002] by a 4-second shock). Low-dose caffeine (25 mg/kg ip after training) improved the learning after 1 hour (P = 0.024); while diazepam (1 mg/kg ip) suppressed learning at all time points after the 4-second shock training (1 hour, P = 0.022; 24 hours, P = 0.002; and 1 week, P = 0.008). No modification in the pain threshold was elicited by electric stimuli both in coriander and control groups. In conclusion, coriander does not improve learning within a short period of time after training; however, learning after coriander administration can be improved in the long term

    Lipid peroxidation and large-scale deletions of mitochondrial DNA in asthenoteratozoospermic patients

    492-499The correlation between malondialdehyde (MDA) an index of lipid peroxidation (LPO) with large-scale deletion mitochondrial DNA (mtDNA) was investigated in a case-control study with a total of 50 semen samples from infertile men, including 25 normozoospermic donor as the control group and 25 asthenoteratozoospermic (AT) patients as the case group. Routine semen analysis was performed according to World Health Organization (WHO, 1999) guidelines. MDA levels of the seminal plasma and spermatozoa were measured by TBARS method. A long-range polymerase chain reaction (PCR) method was used for the analysis of multiple large-scale mtDNA deletions based in two areas of mtDNA. The results showed that mean concentration of MDA in seminal plasma (nmol/ml) and spermatozoa (nmol/10 Ă— 106 sperm) of AT men was higher than in normozoospermic patients, but the differences were not statistically significant. The products of PCR analysis showed multiple deletions of ~4.7, 4.8, 7.2, 7.3 and 7.4-kb in mtDNA of the spermatozoa in both AT and control groups. Multiple deletions were also observed in 64% of AT patients and 44% of the control group. Moreover, MDA level of the spermatozoa in deleted mtDNA samples group was significantly higher than in non-deleted mtDNA group (p = 0.01). Our findings indicated a positive correlation between increased MDA levels and large-scale mtDNA deletions in human spermatozoa. It is suggested that LPO or other oxidative stress factors might be causative elements in mtDNA damage, effect on sperm motility and morphology, resulting in decline of fertility in men. </span

    Prediction of the Presence of Lipid Derivatives in Follicular Fluid and Reproductive Outcome among Infertile Women by MALDI Mass Spectrometry Method

    It has been documented that specific follicular fluid (FF) biochemical characteristics may be essential to determining oocyte quality. Lipid derivatives have most important role in the fertilization process and embryonic development. MALDI mass spectrometry is used for the diagnosis of biomolecules in the FF and serum of infertile women. FF and blood samples from 13 infertile women (20-38years old) undergoing intracytoplasmic sperm injection (ICSI) was obtained, stored frozen at -80Âş C, and later analyzed for Lipid Derivatives. Both FF and serum samples were compared and the Mfg was used as similarity index. For comparison between the FF and serum samples for one person, the Mass spectrometry (MS) intensity ratio versus molecular weight, between the FF and serum samples were calculated. Out of 13 patients, three ongoing pregnancies were observed, so the percentage of pregnancy in the studied population was 25%. The patients who became pregnant after micro-injection had higher unsaturated fatty acids and omega-3 fatty acid than other samples, but Stearic and Palmitic fatty acids were not high in these patients. A number of specific fatty esters and peptide derivatives in the FF and serum samples were found that may help to improve oocyte quality

    Effect of zinc-deficient nutrition on craniofacial bone growth in rats

    Background: Zinc (Zn) is an essential nutrient that is required in humans and animals for the growth, development, and maintenance of healthy bones. The aim of this study is to investigate the effects of zinc-deficient nutrition on the dental, mandibular, maxillary, and cranial dimensions of rats. Materials and Methods: This experimental study was carried out on 14 male Wistar rats. The rats were randomly divided into two groups. Group I rats were fed with a Zn-deficient (ZD) diet, and Group II rats with a Zn-containing (ZC) diet. All the rats on the experimental diet were killed at the end of the fourth week and their blood samples were taken. The serum Zn levels were measured by an atomic absorption spectrophotometer. Radiographic assessment of the jaw bone density was done at the end of the study. Subsequently, the final measurements were made on the dry skulls, the mandibles, and teeth in both the groups. Statistical evaluation was performed by the student′s t-test and repeated measures analysis. The difference between the groups was considered statistically significant if P < 0.05. Results: The ZD group showed a significantly lower value in body weight (P < 0.05), serum level of zinc (P < 0.0001), and radiographic bone density of the mandible (P = 0.02). With regard to the craniofacial parameters, a significant difference was observed only in the length of the clinical crowns of the teeth (L13), which were longer in group II as compared to group I (P = 0.03). Conclusion: This study confirmed that changes in zinc intake could not affect the growth of craniofacial structures. Also, it might change the radiographic bone density of the mandible
