Quantification of the soluble Receptor of Advanced Glycation End-Products (sRAGE) by LC-MS after enrichment by strong cation exchange (SCX) solid-phase extraction (SPE) at the protein level
The study of low abundant proteins contributes to increasing our knowledge about (patho) physiological processes and may lead to the identification and clinical application of disease markers. However, studying these proteins is challenging as high-abundant proteins complicate their analysis. Antibodies are often used to enrich proteins from biological matrices prior to their analysis, though antibody-free approaches have been described for some proteins as well. Here we report an antibody-free workflow on the basis of strong cation exchange (SCX) enrichment and liquid chromatography-mass spectrometry (LC-MS) for quantification of the soluble Receptor of Advanced Glycation End-products (sRAGE), a promising biomarker in chronic obstructive pulmonary disease (COPD). sRAGE was quantified in serum at clinically relevant low to sub ng mL(-1) levels. The method was validated according to U.S. Food and Drug Administration (FDA) and European Medicines Agency (EMA) guidelines and was compared to an antibody-based LC-MS sRAGE method. The SCX-based method builds upon the bipolar charge distribution of sRAGE, which has a highly basic N-terminal part and an acidic C-terminal part resulting in an overall neutral isoelectric point (pI). The highly basic N-terminal part (pI(calculated) = 10.3) allowed for sRAGE to be enriched by SCX at pH 10, a pH at which most serum proteins do not bind. This study shows that ion exchange-based enrichment is a viable approach for the LC-MS analysis of several low abundant proteins following a thorough analysis of their physical-chemical properties. Collins and Sivers asymmetries in muonproduction of pions and kaons off transversely polarised protons
Measurements of the Collins and Sivers asymmetries for charged pions and charged and neutral kaons produced in semi-inclusive deep-inelastic scattering of high energy muons off transversely polarised protons are presented. The results were obtained using all the available COMPASS proton data, which were taken in the years 2007 and 2010. The Collins asymmetries exhibit in the valence region a non-zero signal for pions and there are hints of non-zero signal also for kaons. The Sivers asymmetries are found to be positive for positive pions and kaons and compatible with zero otherwise. © 2015
Transverse Spin Effects and TMDs in Semi-Inclusive DIS from the COMPASS Experiment at CERN
The measurement of azimuthal single spin asymmetries in semi-inclusive deep-inelastic scattering (SIDIS) on a transversely polarized target is an important part of the COMPASS program, giving access to the transversity distribution functions as well as transverse momentum dependent distribution functions. After COMPASS took data in the years 2002- 2004 by scattering a 160 GeV muon beam off a transversely polarized deuteron (LiD) target, in 2007 and 2010 additional data was collected on a transversely polarized proton (NH) target. In this contribution, the results of the 2007 measurements of the Collins and Sivers asymmetries in single hadron production as well as two-hadron asymmetries are presented
Transversity signals in two hadron correlation at COMPASS
Over the last couple of years, transverse spin physics has gained increasing attention as well from theoretical as from experimental side. To fully specify the quark structure of the nucleon at the twist-two level, the transverse spin distribution function has to be taken into account. The measurement of two hadron production introducing the chiral odd interference fragmentation function is considered a new probe of the transverse spin distribution function. COMPASS is a fixed target experiment on the SPS M2 beamline at CERN. Its target can be polarised both longitudinally and transversally with respect to the polarised 160 GeV/c beam. In 2002, 2003, and 2004, 20% of the beam-time was spent in the transverse configuration on a LiD target, allowing the measurement of transversity effects on a deuterium target. The results of the analysis of two hadron production based on the full statistics on the deuterium target are reported
