    Wnt2 and WISP-1/CCN4 induce intimal thickening via promotion of smooth muscle cell migration

    Objective—Increased vascular smooth muscle cell (VSMC) migration leads to intimal thickening which acts as a soil for atherosclersosis, as well as causing coronary artery restenosis after stenting and vein graft failure. Investigating factors involved in VSMC migration may enable us to reduce intimal thickening and improve patient outcomes. In this study, we determined whether Wnt proteins regulate VSMC migration and thereby intimal thickening. Approach and Results—Wnt2 mRNA and protein expression were specifically increased in migrating mouse aortic VSMCs. Moreover, VSMC migration was induced by recombinant Wnt2 in vitro. Addition of recombinant Wnt2 protein increased Wnt1-inducible signaling pathway protein-1 (WISP-1) mRNA by ≈1.7-fold, via β-catenin/T-cell factor signaling, whereas silencing RNA knockdown of Wnt-2 reduced WISP-1 mRNA by ≈65%. Treatment with rWISP-1 significantly increased VSMC migration by ≈1.5-fold, whereas WISP-1 silencing RNA knockdown reduced migration by ≈40%. Wnt2 and WISP-1 effects were integrin-dependent and not additive, indicating that Wnt2 promoted VSMC migration via WISP-1. Additionally, Wnt2 and WISP-1 were significantly increased and colocated in human coronary arteries with intimal thickening. Reduced Wnt2 and WISP-1 levels in mouse carotid arteries from Wnt2+/− and WISP-1−/− mice, respectively, significantly suppressed intimal thickening in response to carotid artery ligation. In contrast, elevation of plasma WISP-1 via an adenovirus encoding WISP-1 significantly increased intimal thickening by ≈1.5-fold compared with mice receiving control virus. Conclusions—Upregulation of Wnt2 expression enhanced WISP-1 and promoted VSMC migration and thereby intimal thickening. As novel regulators of VSMC migration and intimal thickening, Wnt2 or WISP-1 may provide a potential therapy for restenosis and vein graft failure

    Aneurysm Severity is Increased by Combined Mmp-7 Deletion and N-cadherin Mimetic (EC4-Fc) Over-Expression

    Abstract There is an unmet need for treatments to reduce abdominal aortic aneurysm (AAA) progression. Vascular smooth muscle cell (VSMC) apoptosis precipitates AAA formation, whereas VSMC proliferation repairs the vessel wall. We previously demonstrated that over-expression of EC4-Fc (truncated N-cadherin), or deletion of matrix-metalloproteinase-7 (Mmp-7) reduced VSMC apoptosis in mouse atherosclerotic plaques. Additionally, MMP-7 promotes VSMC apoptosis by cleavage of N-cadherin. We investigated their combined effect on AAA formation. Increased apoptosis and proliferation were observed in human AAA (HAAA) sections compared to normal aortae (HA). This coincided with increased MMP-7 activity and reduced N-cadherin protein levels in HAAA sections compared to HA. Using a mouse model of aneurysm formation, we showed that the combination of Mmp-7 deletion and EC4-Fc overexpression significantly increased AAA severity. Medial apoptosis and proliferation were both significantly reduced in these mice compared to control mice. In vitro, MMP-7 inhibition and EC4-Fc administration significantly supressed human aortic VSMC apoptosis (via activation of PI-3 kinase/Akt signalling) and proliferation. In conclusion, combined Mmp-7 deletion and systemic over-expression of EC4-Fc reduced both proliferation and apoptosis. Reduced proliferation-mediated repair over-rides any benefit of reduced apoptosis, increasing aneurysm severity. Future studies should therefore focus on retarding VSMC apoptosis whilst promoting VSMC proliferation

    Breaking the Cycle of Incarceration: Strategies for Successful Reentry Final Report for Labyrinth Outreach Services for Women

    Working with a local reentry organization, Labyrinth Outreach Services to Women, the purpose of this study was to gather information about opportunities and barriers related to two aspects of their program: employment services and establishment of a microbusiness. Information was obtained through a 22-item questionnaire given to a sample of local businesses, key informant interviews, and secondary data analysis. Thirty-nine businesses in the Bloomington-Normal area responded to the questionnaire via on-line and paper survey methods, nine face-to-face interviews were conducted, along with three case studies of similar reentry microbusiness programs and a review of current literature. Stigmas of formerly incarcerated women, such as being unmotivated, irresponsible, disobedient, and violent were found to be major barriers to hiring. Significant facilitators identified for increased consideration for employment were: having support of a job coach, professionalism, expressing passion for the job, and seeking jobs with low customer contact. Successful microbusinesses within similar reentry organizations involved realistic expectations, client control over business operations, local community involvement, practice of a holistic approach, insurance of high product quality, and a focus on multiple products. Major barriers identified were obtaining start-up capital and revenue not meeting expenses. The most appropriate structure was found to be a social enterprise, which focuses more on non-monetary benefits for the employees rather than a profit focus of a traditional microbusiness.     Understanding the landscape of policing when responding to vulnerability: interviews with frontline officers across Wales

    The National Adverse Childhood Experiences Approach to Policing Vulnerability: Early Action Together (E.A.T) programme is a unique collaboration between Public Health Wales and the four Welsh Police Forces and Police and Crime Commissioners, in partnership with Criminal Justice, Youth Justice and third sector organisations, funded by the Home Office to deliver a national programme of change across Wales. Whilst public safety, welfare and vulnerability is the highest demand for UK police forces, frontline officers and staff face many challenges responding to vulnerable individuals. The E.A.T. programme sets out to address these challenges and transform how police and partner agencies work together to respond to vulnerability beyond statutory safeguarding. Recognising the importance of early intervention and preventative action, this programme will develop a whole systems response to vulnerability to ensure pathways for support are available for the police when vulnerability falls below thresholds for statutory support. Building into current systems, this work will utilise existing community assets to develop a bank of resources for police and partners to draw upon when supporting people in their communities. This report is the first in a series of reports that has sought to understand the landscape in policing vulnerability across Wales, which in turn will support the E.A.T programme approach. It outlines the reality of responding to vulnerable individuals for frontline officers, the enablers and blockers in current service delivery and examines the introduction of the Adverse Childhood Experience Trauma-Informed Multi-agency Early Action Together training (ACE TIME training). This report provides the individual, situational and organisational context within which to view post ACE TIME training findings and provide key recommendations when preparing to deliver a National transformational and cultural change programme within policing. To capture the rich, complex picture of policing vulnerability 152 semi-structured interviews with a range of different policing roles across Wales were conducted. A review of the literature provided an understanding of the nature of vulnerability demand, examined key aspects that influence responses to incidents of vulnerability and explored potential factors that may affect engagement with the transformational change the E.A.T programme attempts to achieve. A number of key areas emerged from the literature that informed interviews with frontline staff: (1) Previous knowledge and understanding of vulnerability through training, systems, policy and guidance; (2) Understanding and attitudes towards ‘trauma-informed’ approaches and ACEs, in a police context; (3) Experiences and views on multi-agency working and collaborative working; (4) Workforce wellbeing, with a strong evidential link between the importance of wellbeing, organisational support and organisational belonging; (5) Attitudes and perceptions of transformational and organisational change programmes within policing

    Global data for ecology and epidemiology: a novel algorithm for temporal Fourier processing MODIS data

    Background. Remotely-sensed environmental data from earth-orbiting satellites are increasingly used to model the distribution and abundance of both plant and animal species, especially those of economic or conservation importance. Time series of data from the MODerate-resolution Imaging Spectroradiometer (MODIS) sensors on-board NASA's Terra and Aqua satellites offer the potential to capture environmental thermal and vegetation seasonality, through temporal Fourier analysis, more accurately than was previously possible using the NOAA Advanced Very High Resolution Radiometer (AVHRR) sensor data. MODIS data are composited over 8- or 16-day time intervals that pose unique problems for temporal Fourier analysis. Applying standard techniques to MODIS data can introduce errors of up to 30% in the estimation of the amplitudes and phases of the Fourier harmonics. Methodology/Principal Findings. We present a novel spline-based algorithm that overcomes the processing problems of composited MODIS data. The algorithm is tested on artificial data generated using randomly selected values of both amplitudes and phases, and provides an accurate estimate of the input variables under all conditions. The algorithm was then applied to produce layers that capture the seasonality in MODIS data for the period from 2001 to 2005. Conclusions/Significance. Global temporal Fourier processed images of 1 km MODIS data for Middle Infrared Reflectance, day- and night-time Land Surface Temperature (LST), Normalised Difference Vegetation Index (NDVI), and Enhanced Vegetation Index (EVI) are presented for ecological and epidemiological applications. The finer spatial and temporal resolution, combined with the greater geolocational and spectral accuracy of the MODIS instruments, compared with previous multi-temporal data sets, mean that these data may be used with greater confidence in species' distribution modelling
