    Understanding the phase behavior of a proto-biomembrane

    The rich thermotropic behavior of lipid bilayers is addressed using phenomenological theory informed by many experiments. The most recent experiment not yet addressed by theory has shown that the tilt modulus in DMPC lipid bilayers decreases dramatically as the temperature is lowered toward the main transition temperature TM . It is shown that this behavior can be understood by introducing a simple free energy functional for tilt that couples to the area per molecule. This is combined with a chain melting free energy functional in which the area is the primary order parameter that is the driver of the main transition. Satisfactory agreement with experiment is achieved with values of the model parameters determined by experiments, but the transition is directly into the gel phase. The theory is then extended to include the enigmatic ripple phase by making contact with the most recent experimentally determined ripple structure.Comment: 11 pages, 9 figure

    Structural Determinants of Water Permeability through the Lipid Membrane

    Despite intense study over many years, the mechanisms by which water and small nonelectrolytes cross lipid bilayers remain unclear. While prior studies of permeability through membranes have focused on solute characteristics, such as size, polarity, and partition coefficient in hydrophobic solvent, we focus here on water permeability in seven single component bilayers composed of different lipids, five with phosphatidylcholine headgroups and different chain lengths and unsaturation, one with a phosphatidylserine headgroup, and one with a phosphatidylethanolamine headgroup. We find that water permeability correlates most strongly with the area/lipid and is poorly correlated with bilayer thickness and other previously determined structural and mechanical properties of these single component bilayers. These results suggest a new model for permeability that is developed in the accompanying theoretical paper in which the area occupied by the lipid is the major determinant and the hydrocarbon thickness is a secondary determinant. Cholesterol was also incorporated into DOPC bilayers and X-ray diffuse scattering was used to determine quantitative structure with the result that the area occupied by DOPC in the membrane decreases while bilayer thickness increases in a correlated way because lipid volume does not change. The water permeability decreases with added cholesterol and it correlates in a different way from pure lipids with area per lipid, bilayer thickness, and also with area compressibility

    Comparing Energy Expenditure During Land and Shallow Water Walking in Overweight and Obese Females

    Introduction: The prevalence of overweight and obesity in the United States has reached epidemic levels. Reduction in body weight is of great importance for overweight and obese individuals through the increase in physical activity. One particular mode of physical activity that is currently growing in popularity is shallow water walking, although little research has been done examining the energy cost of this activity in an overweight and obese population. Purpose: To compare the energy expenditure (kcal/min) and rating of perceived exertion (RPE) of a bout shallow water walking at a self-selected pace to a bout of land walking at a matched heart rate response and to a bout of land walking at a self-selected pace in overweight and obese women. Methods: Nineteen participants completed three 10-minute experimental trials including a self-selected pace shallow water walking trial, a matched heart rate response land walking trial, and a self-selected pace land walking trial. Results: Significantly lower energy expenditure (p= 0.046) was observed for shallow water walking (6.46 ± 1.38 kcal/min) compared to the matched heart rate response land walking bout (7.26 ± 1.29 kcal/min), although no significant differences were detected for energy expenditure for shallow water walking and self-selected pace land walking (6.92 ± 1.61 kcal/min). No significant differences were detected for RPE across conditions (p=0.439). Exploratory analyses revealed correlations between measures of body composition (BMI and percent body fat) and the difference in energy expenditure between shallow water walking and matched heart rate response land walking. Conclusions: Findings from the current study suggest that although producing energy expenditure compared to heart rate matched land walking, shallow water walking is a viable alternative that can elicit and increase in energy expenditure performed at a moderate intensity, meeting ACSM criteria. Results of the exploratory analyses revealed an association between measures of body composition and differences in energy expenditure. Although only a limited number of participants of the present study had BMI’s above 35.0 kg/m2 (n=2), findings suggest that water exercise may be an alternative form of exercise and produce higher caloric expenditure at higher ranges of BMI and percent body fat
