    Characterization of the fertilization independent endosperm (FIE) gene from soybean

    Reproduction of angiosperm plants initiates from two fertilization events: an egg fusing with a sperm to form an embryo and a second sperm fusing with the central cell to generate an endosperm. The tryptophan-aspartate (WD) domain polycomb protein encoded by fertilization independent endosperm (FIE) gene, has been known as a repressor of hemeotic genes by interacting with other polycomb proteins, and suppresses endosperm development until fertilization. In this study, one Glycine max FIE (GmFIE) gene was cloned and its expression in different tissues, under cold and drought treatments, was analyzed using both bioinformatics and experimental methods. GmFIE showed high expression in reproductive tissues and was responsive to stress treatments, especially induced by cold. GmFIE overexpression lines of transgenic Arabidopsis were generated and analyzed. Delayed flowering was observed from most transgenic lines compared to that of wild type. Overexpression of GmFIE in Arabidopsis also leads to semi-fertile of the plants.Keywords: Polycomb proteins, fertilization independent endosperm (FIE), Glycine max, Arabidopsis thalian

    Attenuating effect of Polygala tenuifolia Willd. seed oil on progression of MAFLD

    Introduction: Metabolic-associated fatty liver disease (MAFLD) is a common chronic metabolic disease that seriously threatens human health. The pharmacological activity of unsaturated fatty acid-rich vegetable oil interventions in the treatment of MAFLD has been demonstrated. This study evaluated the pharmacological activity of Polygala tenuifolia Willd, which contains high levels of 2-acetyl-1,3-diacyl-sn-glycerols (sn-2-acTAGs).Methods: In this study, a mouse model was established by feeding a high-fat diet (HFD, 31% lard oil diet), and the treatment group was fed a P. tenuifolia seed oil (PWSO) treatment diet (17% lard oil and 14% PWSO diet). The pharmacological activity and mechanism of PWSO were investigated by total cho-lesterol (TC) measurement, triglyceride (TG) measurement and histopathological observation, and the sterol regulatory element-binding protein-1 (SREBP1), SREBP2 and NF-κB signaling pathways were evaluated by immunofluorescence and Western blot analyses.Results: PWSO attenuated the increases in plasma TC and TG levels. Furthermore, PWSO reduced the hepatic levels of TC and TG, ameliorating hepatic lipid accumulation. PWSO treatment effectively improves the level of hepatitic inflammation, such as reducing IL-6 levels and TNF-α level.Discussion: PWSO treatment inactivated SREBP1 and SREBP2, which are involved in lipogenesis, to attenuate hepatic lipid accumulation and mitigate the inflammatory response induced via the NF-κB signaling pathway. This study demonstrated that PWSO can be used as a relatively potent dietary supplement to inhibit the occurrence and development of MAFLD

    PGAweb: A Web Server for Bacterial Pan-Genome Analysis

    An astronomical increase in microbial genome data in recent years has led to strong demand for bioinformatic tools for pan-genome analysis within and across species. Here, we present PGAweb, a user-friendly, web-based tool for bacterial pan-genome analysis, which is composed of two main pan-genome analysis modules, PGAP and PGAP-X. PGAweb provides key interactive and customizable functions that include orthologous clustering, pan-genome profiling, sequence variation and evolution analysis, and functional classification. PGAweb presents features of genomic structural dynamics and sequence diversity with different visualization methods that are helpful for intuitively understanding the dynamics and evolution of bacterial genomes. PGAweb has an intuitive interface with one-click setting of parameters and is freely available at http://PGAweb.vlcc.cn/

    Screening and identification of seed-specific genes using digital differential display tools combined with microarray data from common wheat

    <p>Abstract</p> <p>Background</p> <p>Wheat is one of the most important cereal crops for human beings, with seeds being the tissue of highly economic value. Various morphogenetic and metabolic processes are exclusively associated with seed maturation. The goal of this study was to screen and identify genes specifically expressed in the developing seed of wheat with an integrative utilization of digital differential display (DDD) and available online microarray databases.</p> <p>Results</p> <p>A total of 201 unigenes were identified as the results of DDD screening and microarray database searching. The expressions of 6 of these were shown to be seed-specific by qRT-PCR analysis. Further GO enrichment analysis indicated that seed-specific genes were mainly associated with defense response, response to stress, multi-organism process, pathogenesis, extracellular region, nutrient reservoir activity, enzyme inhibitor activity, antioxidant activity and oxidoreductase activity. A comparison of this set of genes with the rice (<it>Oryza sativa</it>) genome was also performed and approximately three-fifths of them have rice counterparts. Between the counterparts, around 63% showed similar expression patterns according to the microarray data.</p> <p>Conclusions</p> <p>In conclusion, the DDD screening combined with microarray data analysis is an effective strategy for the identification of seed-specific expressed genes in wheat. These seed-specific genes screened during this study will provide valuable information for further studies about the functions of these genes in wheat.</p

    Impact of Resource-Saving and Environment-Friendly Society Construction on Sustainability

    Promoting coordinated and environmentally sustainable development of the Chinese economy is one of the primary tasks at the moment, as well as one of the sustainable development goals of the United Nations. The Resource-saving and Environment-friendly Society (RES) has created a pilot promotion policy aimed at China, with the goal of supporting the sustainable development of economic production activities while preserving natural ecosystems. In this investigation, we used the global Malmquist&ndash;Luenberger index with a slack-based measure of the directional distance function to calculate the industrial green total factor productivity (IGTFP) of 105 prefecture-level cities along the Yangtze River from 2004 to 2019; IGTFP was used as a proxy for sustainable development. Then, by considering the RES construction as a quasi-natural experiment, we used propensity score matching difference-in-differences (PSM-DID) to determine the effect of RES construction on sustainable development of the Yangtze River economic belt. We also employed the mediating effect model and triple difference (DDD) model to further analyze the mechanisms underlying the heterogeneous impacts of different urban characteristics. The results revealed three key findings. (1) According to the IGTFP calculation results, RES construction can effectively promote green technological progress while inhibiting green technological efficiency. (2) After carrying out a series of robustness tests, we found that RES construction increased the IGTFP of pilot cities by 4%, indicating that RES construction can significantly promote the sustainable development of pilot cities along the Yangtze River. (3) The results of the mechanism analysis showed that RES construction had a significantly positive impact on sustainable development through technological innovation, human capital, energy conservation, and emission control. In terms of different urban characteristics, the RES construction promoted sustainable development in first-tier cities, second-tier cities, and resource-based cities. We summarized the practical experience of the RES construction as a typical pilot promotion policy. It provides an empirical basis for planning the construction of eco-friendly urban areas in the 21st century and responding to the international community&rsquo;s sustainable development goals from a Chinese perspective

    A Secure Multimedia Data Sharing Scheme for Wireless Network

    A large number of wireless devices like WiFi cameras and 4G robots have been deployed in the rapidly growing wireless network such as Internet of Things. All of the devices (sensors) are collecting and analyzing multimedia data all the time while they are actively working, and it is also required to share data among these the sensors. Typically, the wireless data is transmitted through the network gateway or the cloud platforms. In such a wireless environment, if there is no appropriate protection to the data, it is easy to cause potential data leakage. In reality, the owner of the sensor might only want to share the multimedia data stored in the sensor with a trusted third party (e.g., a family member or a coworker) through an internet gateway or the cloud platform. Ideally, the gateway or the cloud platform in the wireless network should transform one user’s encrypted data (wireless multimedia data) directly into another ciphertext under a set of new users (e.g., a trusted third party) without accessing the user’s plaintext data. In this work, a new secure notion called fuzzy-conditional proxy broadcast re-encryption (FC-PBRE) is presented to address the concern. In a FC-PBRE scheme, the proxy (the gateway or cloud server) uses a broadcast re-encryption key to re-encrypt the encrypted wireless multimedia data which can be decrypted by a set of delegatees if and only if the broadcast key’s conditional set W is close to the conditional set W′ of the ciphertext. With the FC-PBRE scheme, the wireless multimedia data is not disclosed and cannot be learnt by the proxy (the gateway or cloud server). In this paper, we first present the definition of security against chosen-ciphertext attacks for FC-PBRE. Second, we propose an efficient fuzzy-conditional proxy broadcast re-encryption scheme. Third, we prove that our FC-PBRE scheme is CCA-secure in the random oracle model based on the Decisional nBDHE assumption

    Synergistic Antibiofilm Effects of Ultrasound and Phenyllactic Acid against Staphylococcus aureus and Salmonella enteritidis

    This study evaluated the effect of the combination of ultrasound and phenyllactic acid (PLA) on inactivating Staphylococcus aureus and Salmonella enteritidis biofilm cells and determined the possible antibiofilm mechanism. S. aureus and S. enteritidis biofilm cells were separately treated with ultrasound (US, 270 W), phenyllactic acid (PLA, 0.5% and 1%), and their combination (US + 0.5% PLA, and US + 1% PLA) for 5, 10, 20, 30, and 60 min. Biofilm inactivation, polysaccharide, and respiratory chain dehydrogenase assays were conducted. US and PLA had a synergistic effect on inactivating bacterial cells in S. aureus and S. enteritidis biofilms. The combination of US and PLA significantly decreased the contents of soluble and insoluble polysaccharides and the activity of respiratory chain dehydrogenase in the biofilm cells compared to the single treatment. Confocal laser scanning microscopy, scanning electron microscopy, and intracellular adenosine-triphosphate (ATP) analyses indicated that the combination of US and PLA seriously destroyed the cell membrane integrity of the S. aureus and S. enteritidis biofilms and caused the leakage of intracellular ATP. These findings demonstrated the synergistic antibiofilm effect of US combined with PLA and offered a research basis for its application in the food industry
