    Genome-wide identification and comprehensive analysis of tubby-like protein gene family in multiple crops

    IntroductionThe highly conserved tubby-like proteins (TLPs) play key roles in animal neuronal development and plant growth. The abiotic stress tolerance function of TLPs has been widely explored in plants, however, little is known about comparative studies of TLPs within crops.MethodsBioinformatic identification, phylogenetic analysis, Cis-element analysis, expression analysis, Cis-element analysis, expression analysis and so on were explored to analysis the TLP gene family of multiple crops.ResultsIn this study, a comprehensive analysis of TLP genes were carried out in seven crops to explore whether similar function of TLPs in rice could be achieved in other crops. We identified 20, 9, 14, 11, 12, 35, 14 and 13 TLP genes in Glycine max, Hordeum vulgare, Sorghum bicolor, Arabidopsis thaliana, Oryza sativa Japonica, Triticum aestivum, Setaria italic and Zea mays, respectively. All of them were divided into two groups and ten orthogroups (Ors) based on amino acids. A majority of TLP genes had two domains, tubby-like domain and F-box domain, while members of Or5 only had tubby-like domain. In addition, Or5 had more exons and shorter DNA sequences, showing that characteristics of different Ors reflected the differentiated function and feature of TLP genes in evolutionary process, and Or5 was the most different from the other Ors. Besides, we recognized 25 cis-elements in the promoter of TLP genes and explored multiple new regulation pathway of TLPs including light and hormone response. The bioinformatic and transcriptomic analysis implied the stresses induced expression and possible functional redundancy of TLP genes. We detected the expression level of 6 OsTLP genes at 1 to 6 days after seed germination in rice, and the most obvious changes in these days were appeared in OsTLP10 and OsTLP12.DiscussionCombined yeast two-hybrid system and pull down assay, we suggested that the TLP genes of Or1 may have similar function during seed germination in different species. In general, the results of comprehensive analysis of TLP gene family in multiple species provide valuable evolutionary and functional information of TLP gene family which are useful for further application and study of TLP genes

    A Two-Year Surveillance of 2009 Pandemic Influenza A (H1N1) in Guangzhou, China: From Pandemic to Seasonal Influenza?

    In this two-years surveillance of 2009 pandemic influenza A (H1N1) (pH1N1) in Guangzhou, China, we reported here that the scale and duration of pH1N1 outbreaks, severe disease and fatality rates of pH1N1 patients were significantly lower or shorter in the second epidemic year (May 2010-April 2011) than those in the first epidemic year (May 2009-April 2010) (P<0.05), but similar to those of seasonal influenza (P>0.05). Similar to seasonal influenza, pre-existing chronic pulmonary diseases was a risk factor associated with fatal cases of pH1N1 influenza. Different from seasonal influenza, which occurred in spring/summer seasons annually, pH1N1 influenza mainly occurred in autumn/winter seasons in the first epidemic year, but prolonged to winter/spring season in the second epidemic year. The information suggests a tendency that the epidemics of pH1N1 influenza may probably further shift to spring/summer seasons and become a predominant subtype of seasonal influenza in coming years in Guangzhou, China

    Visual Detection of Water Content Range of Seabuckthorn Fruit Based on Transfer Deep Learning

    To realize the classification of sea buckthorn fruits with different water content ranges, a convolution neural network (CNN) detection model of sea buckthorn fruit water content ranges was constructed. In total, 900 images of seabuckthorn fruits with different water contents were collected from 720 seabuckthorn fruits. Eight classic network models based on deep learning were used as feature extraction for transfer learning. A total of 180 images were randomly selected from the images of various water content ranges for testing. Finally, the identification accuracy of the network model for the water content range of seabuckthorn fruit was 98.69%, and the accuracy on the test set was 99.4%. The program in this study can quickly identify the moisture content range of seabuckthorn fruit by collecting images of the appearance and morphology changes during the drying process of seabuckthorn fruit. The model has a good detection effect for seabuckthorn fruits with different moisture content ranges with slight changes in characteristics. The migration deep learning can also be used to detect the moisture content range of other agricultural products, providing technical support for the rapid nondestructive testing of moisture contents of agricultural products

    Simultaneous detection of enterovirus-D68 and vaccine-related poliovirus 3 in the stool samples of a 5-month hospitalized child with acute respiratory disease: A case report

    Human enterovirus (EV) infections can lead to various manifestations, with variable correlations between genotypes and symptoms. Human enterovirus D68 (EV-D68) was considered to be associated with acute respiratory disease and acute flaccid myelitis. In this short report, both EV-D68 and poliovirus 3 were detected in the stool of a hospitalized 5-month child who presented with acute respiratory symptoms and who was recently vaccinated with oral polio vaccine (OPV), using a metatranscriptomic high-throughput sequencing method. The nearly full-length genome sequences with complete open reading frames of EV-D68 and poliovirus 3 were assembled. One previously-reported neurovirulence-related amino acid substitution (T860N) in the EV-D68 VP1 region was observed, but the patient showed no neurological symptoms. More attention should be paid to EV-D68, and continuous multiple syndrome-based surveillance on non-polio enterovirus is called for

    Identification of a New Wide-Compatibility Locus in Inter-Subspecific Hybrids of Rice (Oryza sativa L.)

    As a special class of rice germplasm, wide-compatibility varieties (WCVs) guarantee the fertility of hybrids when there is cross-fertilization between two subspecies. In this study, Chenghui9348 was identified as a new member of the WCV family that improves pollen fertility in an inter-subspecific hybrid. Cytological analysis showed that the abnormal mitosis of microspores resulted in the sterility of pollens at the early bicellular stage in the inter-subspecific hybrid. Furthermore, the new F12 locus, corresponding to improvements in fertility of the indica-japonica hybrid, was found to co-segregate with the RM1047 marker and associated with a region of approximately 630 kb flanked by the D1101 and D1164 markers on chromosome 12. In this region, two putative genes were predicted as the candidates for wide-compatibility genes (WCGs). Sequence analysis revealed that, compared with indica/japonica alleles, deletion/insertion occurred within exons of both putative genes. Together, the present study identified another new WC locus, F12, and offers more opportunities for further exploitation of inter-subspecific hybrids in rice

    Current Progress in Natural Degradation and Enhanced Removal Techniques of Antibiotics in the Environment: A Review

    Antibiotics are used extensively throughout the world and their presence in the environment has caused serious pollution. This review summarizes natural methods and enhanced technologies that have been developed for antibiotic degradation. In the natural environment, antibiotics can be degraded by photolysis, hydrolysis, and biodegradation, but the rate and extent of degradation are limited. Recently, developed enhanced techniques utilize biological, chemical, or physicochemical principles for antibiotic removal. These techniques include traditional biological methods, adsorption methods, membrane treatment, advanced oxidation processes (AOPs), constructed wetlands (CWs), microalgae treatment, and microbial electrochemical systems (such as microbial fuel cells, MFCs). These techniques have both advantages and disadvantages and, to overcome disadvantages associated with individual techniques, hybrid techniques have been developed and have shown significant potential for antibiotic removal. Hybrids include combinations of the electrochemical method with AOPs, CWs with MFCs, microalgal treatment with activated sludge, and AOPs with MFCs. Considering the complexity of antibiotic pollution and the characteristics of currently used removal technologies, it is apparent that hybrid methods are better choices for dealing with antibiotic contaminants

    OsGRP3 Enhances Drought Resistance by Altering Phenylpropanoid Biosynthesis Pathway in Rice (Oryza sativa L.)

    As a sessile organism, rice often faces various kinds of abiotic stresses, such as drought stress. Drought stress seriously harms plant growth and damages crop yield every year. Therefore, it is urgent to elucidate the mechanisms of drought resistance in rice. In this study, we identified a glycine-rich RNA-binding protein, OsGRP3, in rice. Evolutionary analysis showed that it was closely related to OsGR-RBP4, which was involved in various abiotic stresses. The expression of OsGRP3 was shown to be induced by several abiotic stress treatments and phytohormone treatments. Then, the drought tolerance tests of transgenic plants confirmed that OsGRP3 enhanced drought resistance in rice. Meanwhile, the yeast two-hybrid assay, bimolecular luminescence complementation assay and bimolecular fluorescence complementation assay demonstrated that OsGRP3 bound with itself may affect the RNA chaperone function. Subsequently, the RNA-seq analysis, physiological experiments and histochemical staining showed that OsGRP3 influenced the phenylpropanoid biosynthesis pathway and further modulated lignin accumulation. Herein, our findings suggested that OsGRP3 enhanced drought resistance in rice by altering the phenylpropanoid biosynthesis pathway and further increasing lignin accumulation
