    An improved mixture of probabilistic PCA for nonlinear data-driven process monitoring

    An improved mixture of probabilistic principal component analysis (PPCA) has been introduced for nonlinear data-driven process monitoring in this paper. To realize this purpose, the technique of a mixture of probabilistic principal component analyzers is utilized to establish the model of the underlying nonlinear process with local PPCA models, where a novel composite monitoring statistic is proposed based on the integration of two monitoring statistics in modified PPCA-based fault detection approach. Besides, the weighted mean of the monitoring statistics aforementioned is utilized as a metrics to detect potential abnormalities. The virtues of the proposed algorithm are discussed in comparison with several unsupervised algorithms. Finally, Tennessee Eastman process and an autosuspension model are employed to demonstrate the effectiveness of the proposed scheme further

    An Allosteric Inhibitor of KRas Identified Using a Barcoded Rapid Assay Microchip Platform

    Protein catalyzed capture agents (PCCs) are synthetic antibody surrogates that can target a wide variety of biologically relevant proteins. As a step toward developing a high-throughput PCC pipeline, we report on the preparation of a barcoded rapid assay platform for the analysis of hits from PCC library screens. The platform is constructed by first surface patterning a micrometer scale barcode composed of orthogonal ssDNA strands onto a glass slide. The slide is then partitioned into microwells, each of which contains multiple copies of the full barcode. Biotinylated candidate PCCs from a click screen are assembled onto the barcode stripes using a complementary ssDNA-encoded cysteine-modified streptavidin library. This platform was employed to evaluate candidate PCC ligands identified from an epitope targeted in situ click screen against the two conserved allosteric switch regions of the Kirsten rat sarcoma (KRas) protein. A single microchip was utilized for the simultaneous evaluation of 15 PCC candidate fractions under more than a dozen different assay conditions. The platform also permitted more than a 10-fold savings in time and a more than 100-fold reduction in biological and chemical reagents relative to traditional multiwell plate assays. The best ligand was shown to exhibit an in vitro inhibition constant (IC_(50)) of ∼24 μM

    Process monitoring based on orthogonal locality preserving projection with maximum likelihood estimation

    By integrating two powerful methods of density reduction and intrinsic dimensionality estimation, a new data-driven method, referred to as OLPP-MLE (orthogonal locality preserving projection-maximum likelihood estimation), is introduced for process monitoring. OLPP is utilized for dimensionality reduction, which provides better locality preserving power than locality preserving projection. Then, the MLE is adopted to estimate intrinsic dimensionality of OLPP. Within the proposed OLPP-MLE, two new static measures for fault detection TOLPP2 and SPEOLPP are defined. In order to reduce algorithm complexity and ignore data distribution, kernel density estimation is employed to compute thresholds for fault diagnosis.     Do Emotional Faces Affect Inhibition of Return? An ERP Study

    Inhibition of Return (IOR) refers to an individual’s slowed localization or discrimination performance for targets that appear in previously cued versus uncued location after a relatively long delay after cue (∼300–500 ms). The current study adopted a cue-target paradigm and used behavioral and event-related potential (ERP) measures to investigate whether IOR would be modulated by emotional faces during an emotion recognition task. For reaction time measure, we found IOR effect and the magnitude of IOR effect were comparable for fearful face target and neutral face target. For ERP measures, valid cues were associated with smaller P1 and larger N1 waveform than that for invalid cues. Fearful faces were associated with a larger N170 than neutral faces. The onset latency of the stimulus-locked lateralised readiness potential (LRP) in the valid cue condition was longer than that in the invalid cue condition, while there was no significant difference on the onset latency of the response-locked LRP between the valid cue and invalid cue condition. These results support the notion that, regardless the emotion component of the stimulus, the inhibitory bias of attention to previous visited location before response contributes to the IOR

    Clinical Observation of Employment of Umbilical Cord Derived Mesenchymal Stem Cell for Juvenile Idiopathic Arthritis Therapy

    Juvenile idiopathic arthritis (JIA), known as Juvenile rheumatoid arthritis, is the most common type of arthritis in children aged under 17. It may cause sequelae due to lack of effective treatment. The goal of this study is to explore the therapeutic effect of umbilical cord mesenchymal stem cells (UC-MSCs) for JIA. Ten JIA patients were treated with UC-MSCs and received second infusion three months later. Some key values such as 28-joint disease activity score (DAS28), TNF-α, IL-6, and regulatory T cells (Tregs) were evaluated. Data were collected at 3 months and 6 months after first treatment. DAS28 score of 10 patients was between 2.6 and 3.2 at three months after infusion. WBC, ESR, and CRP were significantly decreased while Tregs were remarkably increased and IL-6 and TNF-α were declined. Similar changes of above values were found after 6 months. At the same time, the amount of NSAIDS and steroid usage in patients was reduced. However, no significant changes were found comparing the data from 3 and 6 months. These results suggest that UC-MSCs can reduce inflammatory cytokines, improve immune network effects, adjust immune tolerance, and effectively alleviate the symptoms and they might provide a safe and novel approach for JIA treatment

    The habitual characteristic of smart phone use under relevant cues among Chinese college students

    Excessive smartphone use may be habitual behavior induced by cues associated with the phone. Habitual behavior occurs outside of awareness and is characterized by lack of control. It is not like problematic smartphone use (PSU) (Brand et al., 2016), which is used to either limit behavior or produce pleasure and relieve feelings of pain, stress, and failure despite significant harmful consequences. 62 college students participated in experiments to test the effects of visual cues and self-control, which are the important characteristic of habitual behavior in smartphone-related behavior. The results showed that a significantly larger amount of cue-related phone use behavior occurred in the setting where participants (a) had their smartphones in sight and (b) were given no instructions to exert self-control, compared to when neither of the two conditions was imposed. The habitual model is a useful framework for understanding PSU and can help people avoid it with less stress. The results provide substantial implications for reducing the frequency and duration of smartphone use among college populations
