Bidirectional regulation of angiogenesis and miR-18a expression by PNS in the mouse model of tumor complicated by myocardial ischemia
BACKGROUND: Panax Notoginseng Saponins (PNS) is the major class of active constituents of notoginseng, a natural product extensively used as a therapeutic agent in China. Tumor when accompanied by cardiovascular disorders poses a greater challenge for clinical management given the paradoxical involvement of angiogenesis, therefore gaining increased research attention. This study aim to investigate effects of PNS and its activity components in the mouse model of tumor complicated with myocardial ischemia. METHODS: Tumor complexed with myocardial ischemia mouse model was first established, which was followed by histological and immunohistochemistry examination to assess the effect of indicated treatments on tumor, myocardial ischemia and tissue specific angiogenesis. MicroRNA (miRNA) profiling was further carried out to identify potential miRNA regulators that might mechanistically underline the therapeutic effects of PNS in this complex model. RESULTS: PNS and its major activity components Rg1, Rb1 and R1 suppressed tumor growth and simultaneously attenuated myocardial ischemia. PNS treatment led to decreased expression of CD34 and vWF in tumor and increased expression of these vascular markers in heart. PNS treatment resulted in reduced expression of miR-18a in tumor and upregulated expression of miR-18a in heart. CONCLUSIONS: Our data demonstrated for the first time that PNS exerts tissue specific regulatory effects on angiogenesis in part through modulating the expression of miR-18a, which could be responsible for its bidirectional effect on complex disease conditions where paradoxical angiogenesis is implicated. Therefore, our study provides experimental evidence warranting evaluation of PNS and related bioactive component as a rational therapy for complex disease conditions including co-manifestation of cancer and ischemic cardiovascular disease
Improving the accuracy of cotton seedling emergence rate estimation by fusing UAV-based multispectral vegetation indices
Timely and accurate estimation of cotton seedling emergence rate is of great significance to cotton production. This study explored the feasibility of drone-based remote sensing in monitoring cotton seedling emergence. The visible and multispectral images of cotton seedlings with 2 - 4 leaves in 30 plots were synchronously obtained by drones. The acquired images included cotton seedlings, bare soil, mulching films, and PE drip tapes. After constructing 17 visible VIs and 14 multispectral VIs, three strategies were used to separate cotton seedlings from the images: (1) Otsu’s thresholding was performed on each vegetation index (VI); (2) Key VIs were extracted based on results of (1), and the Otsu-intersection method and three machine learning methods were used to classify cotton seedlings, bare soil, mulching films, and PE drip tapes in the images; (3) Machine learning models were constructed using all VIs and validated. Finally, the models constructed based on two modeling strategies [Otsu-intersection (OI) and machine learning (Support Vector Machine (SVM), Random Forest (RF), and K-nearest neighbor (KNN)] showed a higher accuracy. Therefore, these models were selected to estimate cotton seedling emergence rate, and the estimates were compared with the manually measured emergence rate. The results showed that multispectral VIs, especially NDVI, RVI, SAVI, EVI2, OSAVI, and MCARI, had higher crop seedling extraction accuracy than visible VIs. After fusing all VIs or key VIs extracted based on Otsu’s thresholding, the binary image purity was greatly improved. Among the fusion methods, the Key VIs-OI and All VIs-KNN methods yielded less noises and small errors, with a RMSE (root mean squared error) as low as 2.69% and a MAE (mean absolute error) as low as 2.15%. Therefore, fusing multiple VIs can increase crop image segmentation accuracy. This study provides a new method for rapidly monitoring crop seedling emergence rate in the field, which is of great significance for the development of modern agriculture

Theoretical Investigation on the ESIPT Process and Detection Mechanism for Dual-Proton Type Fluorescent Probe
Recently, a new fluorescent probe AE-Phoswas reported to detect the activity of alkaline phosphatases (ALP) in different living cell lines. Here, we present an in-depth computational analysis of the mechanism and source of the fluorescence of the AE-Phos probe. There is an intermediate product (AE-OH-Phos) in the experiment as well as a different configuration of products that may emit fluorescence. It is essential to investigate the origin of fluorescence and the detection mechanism of the probe, which could help us eliminate the interference of other substances (including an intermediate product and possible isomers) on fluorescence during the experiment. According to the change of geometric parameters and Infrared spectra, we deduce that the dual intramolecular hydrogen bonds of salicylaldehyde azine (SA) were enhanced at the excited state, while AE-OH-Phos was attenuated. Considering the complex ESIPT behavior of the dual proton-type probe, the potential energy surfaces were further discussed. It can be concluded that the single proton transfer structure of SA (SA-SPT) is the most stable form. Both the concerted double proton transfer process and stepwise single proton transfer process of SA were forbidden. The fluorescence for SA was 438 nm, while that of SA-SPT was 521 nm, which agrees with the experimentally measured fluorescence wavelength (536 nm). The conclusion that single proton transfer occurs in SA is once again verified. In addition, the distribution of electron-hole and relative index was analyzed to investigate the intrinsic mechanism for the fluorescence quenching of the probe and the intermediate product. The identification of the origin of fluorescence sheds light on the design and use of dual-proton type fluorescent probes in the future
A DFT/TD-DFT Study on the ESIPT-Type Flavonoid Derivatives with High Emission Intensity
To reveal the influence of different substituents on the excited-state intramolecular proton transfer (ESIPT) process and photophysical properties of 4′-N, N-dimethylamino-3-hydroxyflavone (DMA3HF), two novel molecules (DMA3HF-CN and DMA3HF-NH2) were designed by introducing the classical electron-withdrawing group cyano (-CN) and electron-donating group amino (-NH2). The three molecules in the acetonitrile phase were systematically researched by applying the density functional theory (DFT) and time-dependent DFT (TD-DFT) methods. The excited-state hydrogen bond enhancement mechanism was confirmed, and the hydrogen bond intensity followed the decreasing order of DMA3HF-NH2 > DMA3HF > DMA3HF-CN, which can be explained at the electronic level by natural bond orbital, fuzzy bond order, and frontier molecular orbital analyses. Moreover, we found from the electronic spectra that the fluorescence intensity of the three molecules in keto form is relatively strong. Moreover, the calculated absorption properties indicated that introducing the electron-withdrawing group -CN could significantly improve the absorption of DMA3HF in the ultraviolet band. In summary, the introduction of an electron-donating group -NH2 can promote the ESIPT reaction of DMA3HF, without changing the photophysical properties, while introducing the electron-withdrawing group -CN can greatly improve the absorption of DMA3HF in the ultraviolet band, but hinders the occurrence of the ESIPT reaction
Profiles of Proinflammatory Cytokines and T Cells in Patients With Tourette Syndrome: A Meta-Analysis
BackgroundTic disorder is a neurodevelopmental disorder characterized by motor and phonic tic symptoms. Tourette syndrome (TS) is a subtype of tic disorder that shows more persistent tic symptoms. The etiological mechanism of TS concerning immune dysfunction remains unclear due to limited evidence, especially for pediatric TS patients. MethodIn the present study, a meta-analysis was performed to confirm the identified changes in proinflammatory cytokines and T cells of pediatric TS patients. A total of five databases, including PubMed, Web of Science, PsycINFO, Google Scholar and the China National Knowledge Infrastructure (CNKI), were used for the literature search. The standardized mean difference (SMD) and mean difference (MD) with a 95% confidence interval (CI) were used to present the effect size of each type of proinflammatory cytokine and T cell. Sensitivity analysis, subgroup analysis and meta-regression analysis were used to explore the heterogeneity of the meta-analysis. This meta-analysis was registered in the International Platform of Registered Systematic Review and Meta-analysis Protocols (number: INPLASY2021110079). ResultsIn the 25 studies included in this meta-analysis, thirteen studies focused on the levels of T cells, and twelve studies focused on the levels of proinflammatory cytokines. Based on the random-effects model, the pooled MDs are -1.45 (95% CI: -3.44, 0.54) for CD3 cells, -4.44 (95% CI: -6.80, -2.08) for CD4 cells, and 1.94 (95% CI: -0.08, 3.97) for CD8 cells. The pooled SMDs are1.36 for IL-6 (95% CI: 0.00, 2.72) and 2.39 for tumor necrosis factor alpha (TNF-alpha) (95% CI: 0.93, 3.84). ConclusionWe provided evidence of immune dysfunction in pediatric TS patients, with elevated levels of particular proinflammatory cytokines and disproportionate changes in T-cell subpopulations. Small to large effect sizes were identified for increased IL-6 levels as well as a reduced number of T helper cells, while a large effect size was identified for increased TNF-alpha levels. These results indicate a close association between peripheral immune activation and TS. However, the most direct and meaningful interaction between peripheral immune status and microglial activation in the central nervous system in TS patients requires further exploration
Effect of Electronic Acceptor Segments on Photophysical Properties of Low-Band-Gap Ambipolar Polymers
Stimulated by a recent experimental report, charge transfer and photophysical properties of donor-acceptor ambipolar polymer were studied with the quantum chemistry calculation and the developed 3D charge difference density method. The effects of electronic acceptor strength on the structure, energy levels, electron density distribution, ionization potentials, and electron affinities were also obtained to estimate the transporting ability of hole and electron. With the developed 3D charge difference density, one visualizes the charge transfer process, distinguishes the role of molecular units, and finds the relationship between the role of DPP and excitation energy for the three polymers during photo-excitation
Computer Simulation of Polyethylene Terephthalate Carbonated Beverage Bottle Bottom Structure Based on Manual–Automatic Double-Adjustment Optimization
PET bottlesare often used as airtight containers for filling carbonated drinks. Because carbonated drinks contain large volumes of CO2 gas, the container needs to bear a tremendous pressure from the inside of the bottle.If the stress exceeds the bearing limit, the material will show the phenomenon of local cracking and liquid overflow.For the structural design, the method of manual adjustment before automatic adjustment was adopted. First, through manual optimization, the initial optimal parameter combination was as follows:the inner diameter of the bottle bottom was 17 mm, the dip angle of the valley bottom was 81°, the deepest part of the valley bottom was 25 mm, and the outer diameter was 27 mm. Comsol software was used for automatic optimization. Compared with the original bottle bottom, the total maximum principal stress and total elastic strain energy in the bottle bottom after manual–automatic double optimization decreased by 69.4% and 40.0%, respectively, and the displacement caused by deformation decreased by 0.60 mm (74.1%). Maize PPR278 Functions in Mitochondrial RNA Splicing and Editing
Pentatricopeptide repeat (PPR) proteins are a large protein family in higher plants and play important roles during seed development. Most reported PPR proteins function in mitochondria. However, some PPR proteins localize to more than one organelle; functional characterization of these proteins remains limited in maize (Zea mays L.). Here, we cloned and analyzed the function of a P-subfamily PPR protein, PPR278. Loss-function of PPR278 led to a lower germination rate and other defects at the seedling stage, as well as smaller kernels compared to the wild type. PPR278 was expressed in all investigated tissues. Furthermore, we determined that PPR278 is involved in the splicing of two mitochondrial transcripts (nad2 intron 4 and nad5 introns 1 and 4), as well as RNA editing of C-to-U sites in 10 mitochondrial transcripts. PPR278 localized to the nucleus, implying that it may function as a transcriptional regulator during seed development. Our data indicate that PPR278 is involved in maize seed development via intron splicing and RNA editing in mitochondria and has potential regulatory roles in the nucleus
