    Protective effect of recombinant staphylococcal enterotoxin A entrapped in polylactic-co-glycolic acid microspheres against Staphylococcus aureus infection

    Staphylococcus aureus is an important cause of nosocomial and community-acquired infections in humans and animals, as well as the cause of mastitis in dairy cattle. Vaccines aimed at preventing S. aureus infection in bovine mastitis have been studied for many years, but have so far been unsuccessful due to the complexity of the bacteria, and the lack of suitable vaccine delivery vehicles. The current study developed an Escherichia coli protein expression system that produced a recombinant staphylococcal enterotoxin A (rSEA) encapsulated into biodegradable microparticles generated by polylactic-co-glycolic acid (PLGA) dissolved in methylene chloride and stabilized with polyvinyl acetate. Antigen loading and surface properties of the microparticles were investigated to optimize particle preparation protocols. The prepared PLGA-rSEA microspheres had a diameter of approximately 5 ÎĽm with a smooth and regular surface. The immunogenicity of the PLGA-rSEA vaccine was assessed using mice as an animal model and showed that the vaccine induced a strong humoral immune response and increased the percent survival of challenged mice and bacterial clearance. Histological analysis showed moderate impairment caused by the pathogen upon challenge afforded by immunization with PLGA-rSEA microspheres. Antibody titer in the sera of mice immunized with PLGA-rSEA microparticles was higher than in vaccinated mice with rSEA. In conclusion, the PLGA-rSEA microparticle vaccine developed here could potentially be used as a vaccine against enterotoxigenic S. aureus

    The complete genome sequences of seven closely related Vibrio cholerae phages isolated from environmental sites in southeastern China are reported here. Phages QH, CJY, H1, H2, H3, J2, and J3 are members of the Podoviridae family and are highly similar to the previously sequenced Vibrio phages VP2, VP5, and phiVC8

    The capBCA Locus is Required for Intracellular Growth of Francisella tularensis LVS

    Francisella tularensis is the causative agent of tularemia and a category A bioterrorism agent. The molecular basis for the extreme virulence of F. tularensis remains unclear. Our recent study found that capBCA, three neighboring genes, are necessary for the infection of F. tularensis live vaccine strain (LVS) in a respiratory infection mouse model. We here show that the capBCA genes are necessary for in vivo growth of F. tularensis LVS in the lungs, spleens, and livers of BALB/c mice. Unmarked deletion of capBCA in type A strain Schu S4 resulted in significant attenuation in virulence although the level of the attenuation in Schu S4 was much less profound than in LVS. We further demonstrated that CapB protein is produced at a low level under the in vitro culture conditions, and capB alone is necessary for in vivo growth of F. tularensis LVS in the lungs of BALB/c mice. Finally, deletional mutations in capB alone or capBCA significantly impaired intracellular growth of F. tularensis LVS in cultured macrophages, thus suggesting that the capBCA genes are necessary for intracellular adaptation of F. tularensis. The requirement of this gene locus in intracellular adaption at least in part explains the significant attenuation of F. tularensis capBCA mutants in virulence

    Reviving phage therapy for the treatment of cholera

    Cholera remains a major risk in developing countries, particularly after natural or man-made disasters. Vibrio cholerae El Tor is the most important cause of these outbreaks, and is becoming increasingly resistant to antibiotics, so alternative therapies are urgently needed. In this study, a single bacteriophage, Phi_1, was used prophylactically and therapeutically to control cholera in an infant rabbit model. In both cases, phage-treated animals showed no clinical signs of disease, compared with 69% of untreated control animals. Bacterial counts in the intestines of phage-treated animals were reduced by up to 4 Log10 CFU/g. There was evidence of phage multiplication only in animals which received a V. cholerae challenge. No phage-resistant bacterial mutants were isolated from the animals, despite extensive searching. This is the first evidence that a single phage could be effective in the treatment of cholera, without detectable levels of resistance. Clinical trials in human patients should be considered

    Metrological Software Test for Simulating the Method of Determining the Thermocouple Error in Situ During Operation

    The simplified metrological software test (MST) for modeling the method of determining the thermocouple (TC) error in situ during operation is considered in the paper. The interaction between the proposed MST and a temperature measuring system is also reflected in order to study the error of determining the TC error in situ during operation. The modelling studies of the random error influence of the temperature measuring system, as well as interference magnitude (both the common and normal mode noises) on the error of determining the TC error in situ during operation using the proposed MST, have been carried out. The noise and interference of the order of 5-6 μV cause the error of about 0.2-0.3°C. It is shown that high noise immunity is essential for accurate temperature measurements using TCs

    Three-dimensional (3D) meshes have been widely used in graphic applications for the representation of 3D objects. They often require a huge amount of data for storage and/or transmission in the raw data format. Since most applications demand compact storage, fast transmission, and efficient processing of 3D meshes, many algorithms have been proposed to compress 3D meshes efficiently since early 1990s. In this survey paper, we examine 3D mesh compression technologies developed over the last decade, with the main focus on triangular mesh compression technologies. In this effort, we classify various algorithms into classes, describe main ideas behind each class, and compare the advantages and shortcomings of the algorithms in each class. Finally, we address some trends in the 3D mesh compression technology development
