Global exponential stabilization of language constrained switched linear discrete-time system based on the s-procedure approach
This paper considers global exponential stabilization (GES) of switched linear discrete-time system under language constraint which is generated by non-deterministic finite state automata. A technique in linear matrix inequalities called S-procedure is employed to provide sufficient conditions of GES which are less conservative than the existing Lyapunov-Metzler condition. Moreover, by revising the construction of Lyapunov matrices and the corresponding switching control policy, a more flexible result is obtained such that stabilization path at each moment might be multiple. Finally, a numerical example is given to illustrate the effectiveness of the proposed results
The efficacy of nasal administration of esketamine in patients having moderate-to-severe pain after preoperative CT-guided needle localization: a randomized, double-blind, placebo-controlled trial
BackgroundWhether nasal administration of esketamine can provide effective analgesia is unclear in patients with acute pain after preoperative CT-guided needle localization.MethodsIn this double-blind, randomized, placebo-controlled trial, patients were assigned to receive either nasal administration of esketamine (0.3 mg/kg or 0.5 mg/kg) or saline (identical in appearance to esketamine) when they had visual analog scale (VAS) pain scores >3/10 during deep breathing after preoperative CT-guided needle localization. The primary outcome was the percentage of patients with satisfactory pain relief, which was defined as VAS pain scores ≤3/10 measured 15 min after intranasal of esketamine or saline. Secondary outcomes included VAS measured following esketamine or saline, the incidence and cumulative dose of rescue hydromorphone use, and related adverse events.ResultsA total of 90 patients were included in the final analysis. Following intranasal treatment, the percentage of patients with satisfactory pain relief was 16.7% (5/30) in the saline group, 56.7% (17/30) in the 0.3 mg/kg esketamine group, and 53.3% (16/30) in the 0.5 mg/kg esketamine group (p = 0.002). The median VAS during deep breathing was less after the intranasal administration of esketamine {median (IQR), 3 (3, 5) in 0.3 mg/kg or 0.5 mg/kg esketamine compared to the saline group [5 (4, 6)], p = 0.009}. The incidence of rescue hydromorphone use was detected less in the esketamine group compared to the saline group (43.3% in the 0.3 mg/kg esketamine group, 36.7% in the 0.5 mg/kg esketamine group, and 73.3% in the saline group, p = 0.010). The adverse events were similar among the three groups (p > 0.05).ConclusionIntranasal administration of esketamine is easier and more effective in alleviating acute pain in patients after preoperative CT-guided needle localization without significant adverse effects
Investigation and epidemiological analysis of chronic diseases and comorbidities in hospitalized patients
Objective·To investigate the current situation and distribution characteristics of chronic comorbidities, and to provide reference for further improving the self-management of comorbidities and implementing the whole course and all-round management of comorbidity.Methods·Two thousand and forty-five inpatients in the Department of Geriatrics,Renji Hospital, Shanghai Jiao Tong University School of Medicine were enrolled in this study from December 2020 to February 2023. The general vital signs, routine laboratory examination and disease status were collected. The epidemiological distribution characteristics of chronic diseases and comorbidities were analyzed.Results·The incidence of chronic diseases in the surveyed population was 99.6%, and the incidence of comorbidities was 94.2%. The top 5 chronic diseases were hypertension (43.68%), diabetes mellitus (24.81%), malignant tumor (21.48%), hyperlipidemia (18.38%) and coronary heart disease (11.99%). The detection rates of hypertension, diabetes mellitus, coronary heart disease, chronic obstructive pulmonary disease, stoke and chronic kidney disease in males were significantly higher than those in females (P<0.05). The proportion of patients with 5 chronic diseases was the highest (11.99%), followed by 7 chronic diseases (10.26%) and 6 chronic diseases (10.04%). Among the patients of different ages, the comorbidity rate was the highest in the patients aged 50‒59 years (27.78%). In different age groups, patients aged 50 to 59 with 2 chronic diseases had the highest incidence of comorbidity, which was as high as 40.82%. Although the overall proportion of comorbidities among male patients (95.37%) was higher than that among females (93.77%), there was no statistically significant difference (P=0.125). However, the proportions of male patients with 2 and 5 chronic diseases were 70.41% and 60.63%, respectively, which were significantly higher than those of female patients (29.59% and 39.37%). The correlations between coronary heart disease and diabetes mellitus, hypertension and coronary heart disease, hypertension and diabetes mellitus were higher (r=0.24, r=0.27, r=0.35, all P<0.05).Conclusion·The prevalence of chronic diseases and comorbidities is high in the middle-aged and elderly population, and the number of comorbidities increases significantly with the increase of age
Atmospheric deposition and river runoff stimulate the utilization of dissolved organic phosphorus in coastal seas
In coastal seas, the role of atmospheric deposition and river runoff in dissolved organic phosphorus (DOP) utilization is not well understood. Here, we address this knowledge gap by combining microcosm experiments with a global approach considering the relationship between the activity of alkaline phosphatases and changes in phytoplankton biomass in relation to the concentration of dissolved inorganic phosphorus (DIP). Our results suggest that the addition of aerosols and riverine water stimulate the biological utilization of DOP in coastal seas primarily by depleting DIP due to increasing nitrogen concentrations, which enhances phytoplankton growth. This “Anthropogenic Nitrogen Pump” was therefore identified to make DOP an important source of phosphorus for phytoplankton in coastal seas but only when the ratio of chlorophyll a to DIP [Log10 (Chl a / DIP)] is larger than 1.20. Our study therefore suggests that anthropogenic nitrogen input might contribute to the phosphorus cycle in coastal seas
Heterogeneity Impacts of Farmers’ Participation in Payment for Ecosystem Services Based on the Collective Action Framework
Payments for ecosystem services (PES) are designed to reduce the impact of human activities on eco-sensitive areas. PES programs often adopt economic-incentive and command-control strategies. Increasing the enthusiasm of farmers’ participation is crucial for the sustainability of PES programs and ecosystem restoration. The watershed ecological compensation in Xin’an River Basin is the first horizontal ecological compensation pilot in China. In this study, economic-incentive strategy and command-control strategies in living and farming are implemented simultaneously to improve water quality. Under the collective action framework, we analyze the drivers of farmers’ participation in three concurrent strategies using hierarchical linear models (HLM). The results show: (1) Overall, 81.79%, 76.26%, and 79.11% of farmers are willing to participate in economic-incentive strategy, command-control strategy in living, and command-control strategy in farming, respectively, while 18.21%, 23.74%, and 20.89% are from the village level. (2) Among statistically significant (p < 0.01) factors at the farmer level, social trust (β = 0.305), and social participation (β = 0.134) have positive effects on the economic-incentive strategy; the number of communication and entertainment equipment has a positive effect on the willingness to participate in command-control strategy in living (β = 0.287) and command-control strategy in farming (β = 0.336). (3) At the village level, village characteristics have a direct impact on the farmers’ willingness to participate in strategies. Village woodland area is positively correlated with strategies participation. In addition, village characteristics play a moderating role by influencing farmers’ sustainable livelihood capital. We conclude that different concurrent strategies and collective actions need to be considered in the design of PES programs, particularly in ecologically sensitive areas, which can enrich the theory of collective action and the connotation of PES
Comprehensive functional characterization of cancer–testis antigens defines obligate participation in multiple hallmarks of cancer
Tumours frequently activate genes whose expression is otherwise biased to the testis, collectively known as cancer–testis antigens (CTAs). The extent to which CTA expression represents epiphenomena or confers tumorigenic traits is unknown. In this study, to address this, we implemented a multidimensional functional genomics approach that incorporates 7 different phenotypic assays in 11 distinct disease settings. We identify 26 CTAs that are essential for tumor cell viability and/or are pathological drivers of HIF, WNT or TGFβ signalling. In particular, we discover that Foetal and Adult Testis Expressed 1 (FATE1) is a key survival factor in multiple oncogenic backgrounds. FATE1 prevents the accumulation of the stress-sensing BH3-only protein, BCL-2-Interacting Killer (BIK), thereby permitting viability in the presence of toxic stimuli. Furthermore, ZNF165 promotes TGFβ signalling by directly suppressing the expression of negative feedback regulatory pathways. This action is essential for the survival of triple negative breast cancer cells in vitro and in vivo. Thus, CTAs make significant direct contributions to tumour biology
Evolution of Cisplatin Resistance Through Coordinated Metabolic Reprogramming of the Cellular Reductive State
BACKGROUND: Cisplatin (CDDP) is a mainstay treatment for advanced head and neck squamous cell carcinomas (HNSCC) despite a high frequency of innate and acquired resistance. We hypothesised that tumours acquire CDDP resistance through an enhanced reductive state dependent on metabolic rewiring.
METHODS: To validate this model and understand how an adaptive metabolic programme might be imprinted, we performed an integrated analysis of CDDP-resistant HNSCC clones from multiple genomic backgrounds by whole-exome sequencing, RNA-seq, mass spectrometry, steady state and flux metabolomics.
RESULTS: Inactivating KEAP1 mutations or reductions in KEAP1 RNA correlated with Nrf2 activation in CDDP-resistant cells, which functionally contributed to resistance. Proteomics identified elevation of downstream Nrf2 targets and the enrichment of enzymes involved in generation of biomass and reducing equivalents, metabolism of glucose, glutathione, NAD(P), and oxoacids. This was accompanied by biochemical and metabolic evidence of an enhanced reductive state dependent on coordinated glucose and glutamine catabolism, associated with reduced energy production and proliferation, despite normal mitochondrial structure and function.
CONCLUSIONS: Our analysis identified coordinated metabolic changes associated with CDDP resistance that may provide new therapeutic avenues through targeting of these convergent pathways
